[The effect of indomethacin and leukinferon on the thromboxane B2 level in the blood plasma of patients with endometrial cancer in the perioperative period].

The paper discusses the effect of indomethacin, leukinferon as well as their combined effect on blood thromboxane B2 (TxB2) level in 40 endometrial cancer patients in the perioperative period. Perioperative treatment with indomethacin was followed by a significant decrease in blood TxB2 level before surgery and in the postoperative period. Treatment with leukinferon exerted similar effect which, however, was less pronounced than that of indomethacin. The effect was most apparent when the two drugs were combined. The influence of indomethacin and leukinferon on metabolism of arachidonic acid in tumor cells and those of the immune system of endometrial cancer patients are discussed as well possible role of eicosanoids in the pathogenetic mechanism of growth and dissemination of reproductive tumors.

Aged

[Leukinferon in immunological correction of acute inflammatory diseases of the abdominal organs].

Addition of (intramuscular+intravenous) leukinferon (LF) to the schemes for the treatment of acute peritonitis promoted a more rapid positive development of the time course of clinical signs and decreasing of leukocytosis in the presence of a pronounced tendency to normalization of the main immunological indices i. e. the counts of differential T-lymphocytes and T-helper cells. There was also activation of neutrophil phagocytic function. A rapid decrease in objective signs of endotoxicosis was recorded: the intoxication leukocytic index and the level of medium-mass molecules. In parallel with the decrease in the intoxication leukocytic index, there was a decrease in cytosis of the peritoneal exudate. The use of LF in the treatment of elderly patients with acute cholecystitis eliminated the clinical signs and normalized the main laboratory indices without surgical interventions which allowed one to make a planned operation with the minimum risk.

Acute Disease

[Effects of interferon preparations on the course of Salmonella infection (experimental and clinical studies)].

The effect of type I interferons on the process of experimental salmonellosis in mice and rabbits, as well as their effect on salmonellosis in patients was studied. It was shown that homologous interferon increased the animal survival rate, activated bactericidal activity of the phagocytic cells and increased elimination of the infectious agents from the host. The complex of the etiotropic and pathogenetic treatment of 32 patients with salmonellosis included human leukocytic interferon, leukinferon. It was administered intramuscularly in a dose of 1 x 10(4)-2 x 10(4) IU: three injections at intervals of 48 to 72 hours (the treatment course). The course was repeated 10 days after the last injection of the first course. Addition of leukinferon to the routine scheme of the medicinal treatment of salmonellosis provided recovery of all the patients without complications or appearance of the bacteria carriers (the observation period of more than 3 years).

[Effects of leukinferon on the phagocytic function of puerperal blood].

Leukinferon effects on the phagocytic activities of the monocytes and polymorphonuclear leukocytes of 12 pregnant women and puerperants with various chronic infectious diseases were studied in vitro. Blood samples of 5 female donors were examined for control. It was found possible to stimulate and correct phagocytic function by leukinferon, proceeding from the initial function of the blood cells and the presence of serum factors. The immunocorrecting properties of leukinferon were found particularly valuable in cases with immunodeficiencies, for infectious diseases and labor complications could thus be prevented in this high-risk group of puerperants.

Cytokines

[The effect of leukinferon on the activity of phagocytosing cells in human salmonellosis].

The functional activity of phagocytic cells in 52 salmonellosis patients was studied with regard to the following characteristics: percent share of phagocytosis, phagocytic index, nitro blue tetrazolium test results, digestive activity. In patients with the unfavorable course of salmonellosis (the formation of carrier state) disturbances in the bactericidal activity of neutrophils and monocytes were established. For 32 patients leukinferon was included in the complex of etiotropic and pathogenetic treatment. The preparation was introduced in 3 intramuscular injections of 10,000 I. U. at intervals of 48 hours (the course of treatment); 10 days after the last injection this course was repeated. The use of leukinferon restored the normal functioning of phagocytes and the number of T-lymphocytes.

[The effect of interferon preparations on the ultrastructural organization of Legionella pneumophila].

The influence of the preparations of interferon on morphological changes in L. pneumophila on the ultrastructural level has been studied. Disturbances in the ultrastructure of L. pneumophila result from the direct bactericidal action of interferons without any interference of immune mechanisms. These disturbances are manifested by damages in the cell wall, plasma membrane, nuclear and ribosomal apparatuses of microbial cells. Leukinferon exhibits pronounced anti-Legionella activity, both in vitro in a liquid culture medium and in ovo, than reaferon.

Animals

[The treatment of different forms of B and delta hepatitis with interferon preparations].

A study was made of the therapeutic efficacy of human leukocytic interferon (HLI) and leikinferon in the treatment of HBV- and HDV-infection. 21 patients were placed under observation. Of these, 6 presented with lingering hepatitis B (HB), 8 with chronic HB, 1 was a HBsAg carrier, 4 had Grades I-IV, acute hepatic encephalopathy, and 2 acute hepatitis delta. 15 patients received leikinferon, 6 were given HLI for injections. Indications and schemes for the treatment with interferon preparations are provided as are the clinico-biochemical and serological criteria for estimating the efficacy of interferon therapy. In lingering and chronic forms of HB, leikinferon exerts a beneficial effect. It is not costly, thus enabling one to carry out continuous treatment in patients suffering from chronic forms. HLI may be recommended as an effective agent.

[Functional activity of platelets in preterm labor].

The platelet component of the hemostasis system was studied in women during preterm labor; platelet functional activity was correlated to changes in prostaglandins E and F-alpha, prostacyclin and thromboxane A2 concentrations. Preterm labor is associated with a significant elevation of platelet functional activity, compatible with that in timely labor. A marked elevation of blood prostaglandin levels was detected. The authors claim that elevated platelet functional activity results not from a gradual increase of the hemostatic potential, but from systemic effect of the labor activity trigger mechanisms on blood coagulation cellular mechanism.

Blood Platelets

[Highly active interferon in the combined treatment of patients with meningoencephalitis].

High-active interferon was used in the complex treatment of 49 patients with nervous system diseases of viral-bacterial etiology. A control group of 28 similar patients were also analyzed. A positive clinical effect was observed in early use of interferon. The clinical course of the disease was milder, the duration of clinical symptoms reduced, healing of the CSF was enhanced, the development of protracted forms of the disease was preventable, lethality reduced 2.9 times with interferon in the complex treatment of these patients.

Acute Disease

[Gallstone-induced intestinal obstruction].

Six patients with gallstone ileus were under observation. The diagnosis was not established before the operation. Surgery was undertaken for acute ileus without identification of the cause. Stones in the intestinal lumen were an unexpected operative finding. The operation of choice was a minimal intervention for the restoration of intestinal continuity (enterolithotomy, resection of the intestine whenever necessary). Five patients recovered, one died.

Acute Disease

[Immunomodulating properties of leukinferon].

Tolerance and immunomodulating properties of leukinferon for injections were studied in 13 healthy volunteers. The drug was administered by inhalation and intramuscularly in a single dose of 10,000 MU twice daily for 3 days. Intramuscular injections of the drug were accompanied by a number of side effects: pyrogenicity, weakness, headache, that ceased 8-12 hrs after the drug was discontinued. Despite these side effects, the drug was satisfactorily tolerated. Neither viscera nor systemic disorders nor allergic reactions were detected. T-lymphocytes proved to be the most sensitive to the drug. Leukinferon was found to activate the interferon system. The effect was the most marked in respect of IRL-gamma.

[The immunomodulating action of interferon preparations. The effect of medicinal alpha-interferon preparations on the rosette-forming activity of human blood lymphocytes].

Effects of alpha-interferon (alpha-IF) medical preparations on human blood lymphocyte rosette-forming activity was under study. Blood samples from 40 donors and 81 patients with chronic pyoderma, lupus erythematosus, and pemphigus were examined. Interferon preparations alpha-IF for nasal administration and IN-1 and IN-2 (or leukinterferon) for injections were used. The results evidence that IF preparations activities depend on the methods of the preparation synthesis and purification. alpha-IF and leukinterferon most effectively enhanced the lymphocyte ability to spontaneous, early, and active rosette formation. IN-1 was found less active and enhanced only the ability to spontaneous E-rosette formation, but had no effect on early active E-RFC subpopulations. IF preparations elevated the counts of E-RFC anf active RFC only if these levels were lowered; if these values were normal the agents had no effect.
