[An evaluation of the quality of tetanus anatoxins by gel electrophoresis].

The study of tetanus toxoids obtained from different manufacturers in the USSR has shown that these preparations exhibit molecular heterogeneity. The method of gel filtration has made it possible to find out that tetanus toxoids from different manufacturers differ in the degree of their purification. The preparations produced by the manufacturing enterprises in Perm and Ufa have been found to contain considerably less ballast substances than the preparations produced in Moscow.

[A gel filtration study of the fractional composition of tetanus antitoxic sera].

The fraction composition of tetanus antitoxic sera has been studied by the method of gel filtration in columns packed with Sephadex G-200. Differences in the fraction composition of preparations manufactured by different enterprises have been revealed. The specific activity of antitoxic sera has been found to depend on the degree of their fragmentation.

[Determination of the antigenic activity of diphtheria anatoxin in liquid and sorbed preparations using an erythrocyte diphtheria antigen (anatoxin) diagnostic agent].

On the basis of the experimentally established dependence of the degree of binding of diphtheria toxoid with standard diphtheria antitoxin on the duration of their joint incubation with the maximum binding occurring in 3-4 hours of incubation at 37 degrees C, the method of the in vitro determination of the antigenic activity of diphtheria toxoid in liquid and adsorbed preparations is proposed. The method is based on the principle of double binding with the use of diphtheria antigenic (toxoid) erythrocyte diagnosticum. The antigenic activity of diphtheria toxoid, evaluated by the degree of its maximum binding with diphtheria antitoxin, correlated with its antitoxin-binding activity in animal experiments and did not correlate with its flocculating activity. The antigenic activity of diphtheria toxoid in adsorbed preparations, evaluated by their maximum binding with standard diphtheria antitoxin, was shown to be closely related to the immunogenic potency of these vaccines in animals.

[Dependence of the specific activity of antitetanus immunoglobulin on the degree of its fragmentation].

Antitetanus immunoglobulin preparations with the increasing content of Fab-fragments (15, 30, 53%) have been obtained under specific experimental conditions. Tests for specific activity have revealed an insignificant decrease (13%) in this activity in the preparation containing 15% of Fab-fragments and its sharp drop in the preparations containing 30-50% of Fab-fragments. The specific activity of antitetanus immunoglobulin has been found to be related to the degree of its fragmentation.

[Limited controlled trial of the reactogenic properties of ADTP vaccines containing stab cultures as the pertussis component].

The reactogenic properties of adsorbed DPT vaccine containing Bordetella pertussis cultures, grown by submerged cultivation, as the pertussis component were studied. The study revealed that the vaccine was low-reactogenic: no severe postvaccinal reactions were recorded in immunized children. The frequency and intensity of febrile and local reactions in children immunized with adsorbed DPT vaccine under trial and the commercial preparation commonly used in the USSR were identical, but considerably less pronounced than in children immunized with foreign vaccines having similar composition.

[Relationship of the sensitizing and protective properties of pertussis antigens].

When comparing delayed hypersensitivity (DN) to B. pertussis corpuscular antigens with the agglutinogenic composition of B. pertussis, as well as to its histamine-sensitizing, leukocyte-sensitizing, adjuvant and hemagglutinating activity, no correlation was detected between the specific sensitizing properties of these antigens and the serotype and studied nonspecific properties of B. pertussis. The DH level correlated with the protective activity of B. pertussis corpuscular antigens and the ultrasonic fractions of B. pertussis. The close correlation of these two phenomena suggest that DH-inducing and protective B. pertussis antigens are identical, though their action has different manifestations, depending on the method of administration of the antigen preparation. On these grounds a new method for evaluating the immunogenicity of B. pertussis antigens is proposed. This method consists in comparing the sensitizing activity of the antigen under test and that of the national standard.

[Action of cyclic adenosine monophosphate on the spleen antibody--forming cells of rabbits immunized with Escherichia coli].

Cyclic adenosine monophosphate (cAMP) parenterally injected to rabbits (immunized intraperitoneally with thymus-independent antigen of killed E. coli 0127/545) during January--April inhibited production of antibody-forming cells (AFC) in the spleen of these animals, and during May--June it increased the AFC count. Both the stimulating and inhibitory effect of cAMP was associated with the administration of the same doses of 25--250 microgram/kg. The nature of the cAMP effect on the production of the AFC depends on the initial immune response level. At the maximum immune response and in the absence of the dose-effect dependence cAMP inhibited the antibody formation, but when the immunological reaction was below the maximal level and in the presence of the dose-effect relationship cAMP increased the AFC production.

[Role of the adenyl cyclase system in achieving the immunogenesis-stimulating action of bacterial lipopolysaccharides--pyrogenal and endogenous serum pyrogen].

Experiments were conducted on rabbits immunized intraperitoneally with corpuscular typhoid vaccine; the number of antibody-forming cells in the spleen proved to increase after tha administration of bacterial lipopolysaccharide (LPS)--pyropeneal, and endogenous serum pyrogen (EPS) together with theopylline. The data obtained indicated that the adenylcyclase system played a certain role in the mechanism of the stimulating action of LPS and EPS.

[The standardization of the methods for assessing the immunogenic activity of all of the components of an adsorbed DTP vaccine].

The reproducibility of laboratory methods, recommended by WHO and used by manufacturing and control institutions of most countries for the evaluation of the immunogenic potency of the pertussis, diphtheria and tetanus components of adsorbed DPT vaccine in the process of its production, was studied. This immunogenic potency was experimentally studied in comparison with the immunogenic potency of national standard samples from Bulgaria, Czechoslovakia, Hungary, Poland and Russia. The results of this study indicated that the coded commercial lots of adsorbed DPT vaccine corresponded to WHO requirements with respect to the immunogenic potency of all their components. At the same time, according to data provided by different laboratories, the indices of their immunogenic potency varied within rather wide limits. Taking into account the necessity of dosing all components of adsorbed DPT vaccine by immunizing (protective) units, the reliability of the recommended control methods should be enhanced by the maximum standardization of all testing conditions and the organization of periodic workshops on the basis of the leading WHO laboratory with a view to improving the qualification of specialists carrying out the quality control of commercial lots of adsorbed DPT vaccine.

[The efficacy of recombinant vaccines against hepatitis B and the results of their use in a schedule for pediatric prophylactic inoculations].

The results of field clinical trials of Russian and American yeast vaccines against hepatitis B are presented. The study revealed that both vaccines were faintly reactogenic, safe and exhibited high immunological activity. After the full course of immunization following the schedule 0-1-2 months 92.5% and 97.5% of patients receiving, respectively. Russian vaccine "Combiotech" and American vaccine "H-B-Vax II" were found to have specific antibodies. The maximum effect was registered when the vaccines were introduced according to the schedule 0-1-6 months. Seroconversions were observed in 97.5% and 100% of the vaccinees receiving the Russian and American vaccines respectively, in the latter case the highest antibody level being observed. The use of the vaccines within the prophylactic immunization schedule showed that antibodies to hepatitis B appeared in immunized children in 93-100% of cases. Seroconversion indices and the levels of antibodies to diphtheria, tetanus, poliomyelitis and measles were statistically significant and were the same in children receiving only the vaccines according to the immunization schedule and in children immunized, in addition to these vaccines, against hepatitis B.
