The significance of positive controls in testing the oncogenic potential of cell material for virus vaccine production.

For all the parts involved in the production and control of virus vaccine it is important that controls of the cell material should be carried out under optimal conditions for the detection of any abnormal characteristics of the cells. The experiments reported here attempt to establish some of the optimal parameters for testing out the tumor-inducing properties of a human diploid cell culture. All experiments were carried out on 6-week old Syrian hamsters and the main objectives were: (i) to devise an optimal immunosupression scheme and (ii) to find a more suitable positive control for testing the oncogenic potential of the human diploid cells. The anti-hamster lymphocyte serum proved to be a more potent immunosuppresser than hydrocortisone succinate as seen from the development of tumors following injections with KB cells; this scheme was subsequently used in assessing the usefulness of KB and Swiss-12 cells as positive controls. The experiments lend support to the suggestion that a heteroploid fibroblastic culture (such as Swiss-12) should become a standard positive control since not only are the tumors so obtained larger and less prone to early necrosis but, more important, they bear a closer resemblance to the kind of tumors likely to be induced by the diploid cell cultures being tested.

[Diagnostic and treatment difficulties in posttraumatic duodenal wounds].

Authors report a number of 5 patients operated for posttraumatic duodenal wounds between 1990 and 1996. Urgency diagnosis, and therapeutical conduct are commented upon, under conditions of surgical teams sometimes very little trained in solving such lesions. Two out of the 5 cases have presented extensive injuries to the duodenum and pancreas, and have brought about surgery tactical and technical problems. These have had an immediate postoperative morbidity full of serious complications, which have necessitated other interventions of a high vital risk. There have been no deaths registered. It has been reported about the advantage, of using "a la Roux" loop anastomosis for solving duodenal wounds which compromise the viability of the duodenal wall on a larger surface than that initially appreciated, when are combined with ampulla detachments and pancreatic lesions.

[Production and characterization of some mouse embryo cellular substrates in vitro].

Cell cultures with a different multiplication potential in vitro, depending upon the strain source used, were obtained from mouse embryos belonging to the CVA, C57 Black A2G and Swiss strains. Only the Swiss 12 culture underwent spontaneous transformation and was carried through more than 50 passages in vitro. The Swiss-12 substrate proved not to be contaminated either by viruses or micoplasma. It is less sensitive than other elective cell substrates to infection with attenuated polioviruses, cytopathogenic Coxsackie A9 virus and vaccinia virus, but its sensitivity to infection with Herpes simplex type 1 virus is similar to that of human embryo fibroblasts. After a high number of passages the Swiss-12 substrate permits, in comparison to other cell substrates (human heteroploid Hep-2 line, human embryo fibroblasts), a highly efficient qualitative differentiation between the growth media and calf serum.

The contribution of cytochemistry and electron microscopy to the detection of contaminants in the cell material used for the production of virus vaccines.

For the production of virus vaccine it is essential to use cellular material free of contaminants that could reach the final product. It is also important to check initial tissue for possible inherent infections. Studies on primary culture of chick embryo fibroblasts have shown in several cases that cultures which appeared normal by current cytological methods had a strongly positive reaction when investigated by the Feulgen reaction for nuclear DNA and acridine orange method proving intense RNA synthesis. Comparison of electron microscopic (EM) pictures of cell sections with results obtained from negatively stained preparations of identical cell material after pronase digestion have shown the presence of viruses, thus elucidating the nature of the inclusions. The combined approach of the above-mentioned problem by cytochemical and EM methods can usefully enlarge the rage of tests employed for the definition of cell populations acceptable for virus vaccine production.

[Term pregnancy, a factor favoring the bleeding of a gastroduodenal ulcer. A case report].

We present the case of a 31-years-old woman, in the 37th week of pregnancy who was urgently hospitalized in our section, with superior digestive bleeding, in a serious state, externalized in hematemesis and melena. The pregnancy being in due time, with sings of secondary fetal pain, probably due to mother's serious anemia, we proceeded to the extraction of the foetus using caesarean section, followed by hemostasis "in situ" of a bleeding duodenal ulcer and the excision of another healed ulcerous lesion (ulcer in mirror). The postoperative evolution was favorable both for mother and new-born child. Reexamined after two years, the patient didn't present subjective pains of an ulcerous type any more, neither lesions visible at an endoscopic examination.
