[Effect of estradiol dipropionate on the development of uterine sarcoma induced by 1,2-dimethylhydrazine in mice].

The study was concerned with the influence of single dose of estradiol dipropionate on its promoting effect on 1,2-dimethylhydrazine-induced tumors in female CBA/CaLac mice. Estradiol dipropionate given within 19 weeks did not affect carcinogenesis whereas 39-week treatment stimulated uterine sarcoma development irrespective of single dose and schedule of administration.

Carcinogenicity of deuterium-labeled 1,2-dimethylhydrazine in mice.

To study the effect of deuterium substitution on the carcinogenicity of 1,2-dimethylhydrazine (DMH) in mice, two comparisons were made between DMH and its fully methyl-deuterated analogue, [2H6]DMH. In a lifetime study with the CBA strain, groups of 19-30 animals of each sex were dosed s.c. weekly with 8 mg/kg of either DMH or [2H6]DMH for 8, 16, or 32 weeks. In the second study, female CF-1 mice were given DMH or [2H6]DMH in 10 weekly s.c. doses of 12 mg/kg each (13.2 mg/kg for [2H6]DMH) and examined for colon tumors 36 weeks after the first dose. Deuteration significantly decreased tumor incidence in the colon of males (P less than 0.01) and the anal tissue of both sexes (P less than 0.05) but increased that of hepatomas and lung tumors in males (P less than 0.01). Substrate deuteration did not significantly affect overall incidence of any other tumor type, however, including hemangioendotheliomas and kidney tumors in both sexes, as well as colon, uterine, ovarian, liver, and lung tumors in females. The results indicate that C--H bond breakage is kinetically important in the activation of DMH to its ultimately carcinogenic form in organs such as the male colon (relative risk in DMH-versus [2H6]DMH-treated animals approximately equal to 6), and that inhibition of this process by substrate deuteration allows a diversionary mechanism having a smaller isotope effect to become relatively more extensive. The qualitatively different effect in other organs (e.g., kidney, relative risk approximately equal to 1) supports recent suggestions that the net mechanism of activation can differ from one target tissue to another, possibly by striking a different balance between parallel metabolic pathways. The lack of a significant isotope effect on overall colon tumor incidence in females of either strain suggests that differences in relative importance among competing enzymes may also be responsible for sexual dimorphism in tumor induction by DMH.

1,2-Dimethylhydrazine carcinogenesis in neonatally androgenized CBA mice.

The aim of this study was to investigate the possible influence of exposure to steroid hormones early in life on the susceptibility of animals as adults to chemical carcinogens. CBA male and female mice received a single subcutaneous injection of 0.5 mg testosterone propionate (TP) in olive oil within 24 h after birth. At the age of 2 months, neonatally androgenized and control mice started receiving weekly subcutaneous injections of 1,2-dimethylhydrazine (DMH). By the end of the experiment, 90% of neonatally androgenized females treated with DMH developed uterine sarcoma against 9% in control females treated with DMH, this difference being attributed to the hyperoestrogenization of androgenized females. In neonatally androgenized males treated with DMH 79% developed pararenal sarcoma and 71% colon tumours versus 25 and 32% respectively of control males treated with DMH.

[Carcinogenic effect of benz(a)pyrene depending on its mode of application to the skin of mice].

The role of dose and time factors in intensification of the carcinogenic effect is studied on the model of skin carcinogenesis in (CBA x C57Bl)F1 female mice under application of BP. The total dose is not a single parameter determining the carcinogenic effect. The single dose interval between the actions and their number are of essential importance under definite conditions. The total time (T) of the carcinogen action is a significant factor. The total and single doses and the number of carcinogen applications to the mouse skin being similar, the effect is the stronger, the longer is T, i.e. the longer is the interval between certain applications of the carcinogen.

[Effects of castration and estradiol dipropionate on carcinogenesis of methylcholanthrene-induced gallbladder cancer in male hamsters].

The study used 8 groups of 8-10 week-old male hamsters. Beeswax pellets containing 5 mg of 20-methylcholanthrene (group 4) or 2 mg (group 5) were injected into the gallbladder. In group 6, the latter treatment was supplemented with castration and subcutaneous injection of 40 micrograms of estradiol dipropionate (EP) once a week. Other experimental exposures included: sham operation (group 2), beeswax pellet injection into the gallbladder (group 3), beeswax pellet injection + castration + EP (group 7) and castration + EP (group 8) with the animals is group 1 being in control. The gallbladder tumor development frequency in groups 4, 5 and 6 was 58.3; 39.3 and 77.8%, respectively. The difference between the data on group 5 and 6 proved significant. The study on cumulative likelihood of tumor development showed a significant increase in tumor frequency in group 6 as compared with group 4 and 5. A strong stimulating effect of castration and EP treatment on tumorigenesis in the gallbladder was demonstrated.

[Histogenesis of endocardial tumors in rats].

10 endocardial tumours (two tumours induced by N-nitrosoethylurea and eight spontaneous) of BD VI rats were examined for protein S-100 using specific antiserum in an immunoperoxidase reaction. The presence of protein S-100 in tumour cells is demonstrated in all cases and is considered as a proof of Schwann cell origin of these tumours.

[Histogenesis of neurinomas induced in rats by nitrosoethylurea].

Twenty-five tumors of the peripheral nerves induced in BD rats by N-nitrosoethylurea were studied by peroxidase-antiperoxidase method using specific anti-serum against protein S-100. Eighteen tumors gave positive reactions for S-100 protein, including those with invasive growth. Protein S-100 was always present in cystic neurinomas. No protein was found in seven tumors. It is concluded that most tumors studied originated from Schwann cells, while at least some of them had a connection with the peri(endo)neurium.

[Transzygotic carcinogenic effect of nitrosoethylurea in rats].

The results of two experiments on the transzygotic transfer of the nitrosoethylurea (NEU) carcinogenic effect are presented. In the first experiment female rats received 60 mg/kg b. w. of NEU at 16, 18 and 20th day of the pregnancy. Their F1 descendants developed multiple tumours of the nervous system after a short latency (up to 40 weeks). The rats of F2, F3, F4 and F5 generations which were not in a direct contact with NEU developed a statistically higher incidence of tumours occurring spontaneously (those of mammary gland, pituitary, and haematopoietic tissue). However, the incidence of nervous system tumours was at the level of controls. In the second experiment male rats received 80 mg/kg b. w. of NEU and then were mated with untreated females. The incidence of the nervous system tumours in the descendants of treated males was somewhat higher than in the controls. The manifestations and mechanisms of the transzygotic effect are discussed.

Experimental carcinoma of the stomach in rats with chronic ulcers and following gastric surgery.

The data obtained did not prove the stimulating effect of experimental chronic ulcer on the induced carcinogenesis of the stomach in the rat. It is shown that some surgical procedures followed by a gastroduodenal reflux enhance the gastric carcinogenesis while the procedures without such a reflux do not reveal any modifying effect on stomach cancer. The results of this study together with data from literature may have some relevance to the treatment procedures of patients with gastric ulcers.

[Benzo(a)pyrene and the carcinogenic effect of coal tar].

The carcinogenic effects of coal tar and benzo(a)pyrene were compared. The agents were applied to murine skin, with coal tar dose being adjusted for benzo(a)pyrene. The carcinogenic effect of coal tar appeared higher than that of benzo(a)pyrene, the relative carcinogenic risk of the former being 3.2 times that of the latter. The coefficient of relative carcinogenic risk of coal tar calculated on the basis of TD56 regression coefficients for dose-effect relationship and mean latent period ratios was averagely 1.5.

[Effect of the duration of administration of 1,2-dimethylhydrazine on its carcinogenic effect].

The (CBA x C57Bl)F1 female mice were treated with weekly injections of 1,2-dimethylhydrazine (DMH) at a dose rate of 4.15 mg/kg of body weight during different time periods. Relations between the incidence of organ specified particular tumours depend on the total DMH dose. Incidence of haemoblastoses decreases with an increase in the DMH dose. Dose relationships of the tumour incidence are analyzed statistically by the method with intercurrent mortality corrections and carcinogen effect expressed by relations of the observed and expected numbers.

Pararenal angiosarcoma induced in male mice by 1,2-dimethylhydrazine--a model for studying the role of androgens in chemical carcinogenesis.

CBA male mice treated with 1,2-dimethylhydrazine (DMH) developed high incidence (up to 97%) of pararenal angiosarcomas. Castration that preceded DMH-treatment almost completely inhibited the induction of these tumours while castration that followed DMH-treatment had on influence on their development. Testosterone propionate (TP) was efficient in restoring the incidence of DMH-induced pararenal tumours in castrated males only when given simultaneously with DMH and was totally inefficient when given after the cessation of DMH-administration. Castrated CBA female mice developed 92% of pararenal angiosarcomas when they received combined treatment with DMH and TP; no such tumours appeared in the intact females treated with DMH alone. The incidence of pararenal tumours in males of different strains was as follows: CBA, 97%; (CBA X C57Bl)F1, 36%; C57Bl, 4%; C3H, 35%; BALB/c, 13%; C3HA, 7%.

[Experimental models of 2-stage carcinogenesis].

The regularities observed in the experiments carried out by the two-stage scheme (initiation-promotion) are considered. Apart from the mouse skin in which these regularities are established, the phenomenon of initiation-promotion is reproduced on the epithelial tumours of the liver, bladder, colon, thyroid, mammary gland, epithelial and mesenchymal tumours of the uterus. The following properties of the promoters are listed: lack of mutagenicity; weak carcinogenicity towards the target organ; threshold of the promoting effect; cessation of carcinogenesis after the promoting treatment has been stopped; organotropism. Approaches and principles of chemical compound testing for cocarcinogenic and promoting activity are considered.

[Comparative quantitative assessment of a carcinogenic effect taking intercurrent mortality into account].

The paper discusses the advantages offered by the statistical evaluation of experimental carcinogenic effect with due regard to mortality from concomitant pathology. The method used complex tables of contingents and analysis was carried out separately in 3 groups of tumors--independent of mortality, fatal and incidental ones. The method was compared with standard procedures which do not take care of the said mortality rate (table of contingents 2 X 2 and precise procedure of Fisher). The data on development of 1,2-dimethylhydrazine-induced tumors of the skin, uterus and ovaries in 5 strains of mice were used in the study.

[Modifying action of neonatal androgenization on 1,2-dimethylhydrazine-induced carcinogenesis in male CBA-strain mice].

Newborn male CBA mice received a single treatment with 0.5 mg testosterone propionate. Weekly subcutaneous injections of 1,2-dimethylhydrazine (DMH) were given to 2-month-old mice. The incidence of pararenal angiosarcomas and colonic tumors in neonatally androgenized mice reached 78.5 and 71.0%, respectively by the 35th week after the DMH treatment was commenced. In DMH-treated control mice, the incidence of the above tumors amounted to 25 and 32%, respectively.
