Morphology and histogenesis of anal region and clitoral gland tumors induced in mice by 1,2-dimethylhydrazine.

Ninety-seven tumors induced in the anal region in female CBA and BALB/c mice by 1,2-dimethylhydrazine were examined histologically. Tumors originated mainly from the following sources: epidermis (papillomas and basal cell neoplasms), pilosebaceous complexes of the anal region (keratoacanthomas, cystic squamous cell tumors, and adenomas of the perianal sebaceous glands), and clitoral (preputial) glands (adenomas and adenoacanthomas). A subcutaneous location and late ulceration were characteristics of many tumors originating from the pilosebaceous complexes and clitoral glads. From 31 to 43% of all anal tumors were of sebaceous gland (perianal or clitoral) origin. Cystic squamous cell tumors were morphologically similar to type II keratoacanthomas of rodents. Squamous cell tumors of any origin finally became squamous carcinomas.

[Stimulating action of estradiol dipropionate on uterine sarcomagenesis induced by 1,2-dimethylhydrazine in mice].

CBA female mice received weekly subcutaneous injections of 1,2-dimethylhydrazine (1,2-DMH) in a dose of 8 mg/kg for 10 weeks (group 1). In groups 2,3 and 4 1,2-DMH plus estradiol dipropionate were administered for 10, 20 or 25 weeks, respectively. The incidence of uterine sarcoma in groups 1, 2, 3 and 4 was 3.3, 2.5, 65.4 and 73%, respectively. The mice given estradiol dipropionate alone developed no uterine sarcomas. Possible mechanisms of the uterine sarcomogenesis stimulation by estrogen are discussed.

[Effect of age, castration and pregnancy on carcinogenesis, induced by 1,2-dimethylhydrazine, in CBA mice].

Subcutaneous injection of 1,2-dimethylhydrazine into female CBA mice once a week resulted in the development of tumours of the colon, anal region, uterus and liver. In 12-13-month-old mice treated with DMH an earlier appearance (week 8) of uterine sarcomas and more rapid increase in the incidence of tumours of the anal region were noted as compared to 3-month-old mice. In pregnant females treated with DMH a statistically significant decrease in the uterine sarcoma incidence was observed (10.3% versus 48.3% in nonpregnant). Pregnancy exerted no effect on the incidence of tumours at other sites. Castration did not affect the time of appearance and the incidence of tumours of any site.

[Morphology of gastrointestinal tract tumors induced in rats by N'-nitro-N-nitrosoguanidine].

For 14 months 200 white noninbred male rats received N-methyl-N'-nitro-N-nitrosoguanidine in various concentrations in drinking water. The animals were sacrificed in 18--22 months. Out of 178 rats surviving the time of appearance of the first tumor 64 animals developed tumours of the gastrointestinal tract. Histologically, gastric tumours in 34 cases were presented by adenomatous outgrowths, in 11 the changes were extremely suspect of cancer, in 2 cases morphologically true cancer was observed. The induced gastric tumours had certain peculiarities as compared with the corresponding tumours of man which consisted in tumour cells growing through the thickness of the wall without marked atypism and lack of metastases. Intestinal tumours were sarcomas in which simultaneously some mesenchymal components were present.

[Pararenal angiosarcoma as a manifestation of sexual dimorphism in carcinogenesis].

Pararenal angiosarcoma developed in 42% of CBA male mice injected subcutaneously 1.2-dimethyl hydrazine (DMH) in a dose of 8 mg/kg body weight within 30 weeks. None of 176 CBA female mice given the same treatment developed such tumors. Histologically, the tumors represented various angiosarcomas characterized by a marked invasive growth into the renal parenchyma.

Non-epithelial uterine tumours induced in CBA mice by 1,2-dimethylhydrazine.

In CBA mice treated with weekly subcutaneous injections of 1,2-dimethylhydrazine an unusually high incidence of uterine sarcomas was observed in two successive experiments. The tumours are easily transplantable. The description of their histological structure is presented and their histogenesis is discussed.

[Modifying effect of DDT and of its various metabolites on the toxic effect of 7,12-dimethylbenz(alpha)anthracene].

In rats a preliminary three times injection of DDT in doses of 10 and 100 mg/Kg eliminated completely DMBA (30 mg/per 100 g of weight) lethal effect and prevented or decreased the development of adrenal necrosis. Among the metabolites under study (DDT, DDON, DDMU, DDE, DDA, DVR) DDE was found to render the strongest protective action in its single administration in a dose of 10 mg/Kg a day before the exposure to DMBA.

[Tumors in CBA line mice caused by 1,2-dimethylhydrazine].

Female CBA mice were treated weekly with subcutaneous injections of 1,2-dimethyl-hydrazine (8 mg/Kg b. w.) for 38--40 weeks. All animals but one developed tumors, mainly of the uterus, and region and intestine. Uterine tumors were most likely endometrial sarcomas, although myometrial and vascular components were also present. Large tumors of the anal region were squamous cell or baso-squamous carcinomata, while small growths were sebaceous adenomas or basal-cell tumors. 1.2-dimethylhydrazine appears to be a more multipotent carcinogen for mice than for the rat. Uterine sarcomas are of special interest in view of their rarity in mice.
