Adhesive properties of blood cells.

The adhesive interactions of the cells with each other and with the extracellular matrix (ECM) are important for their normal growth and differentiation. Currently, very little is known concerning the adhesive and cell recognition factors that maintain cell-to-cell contacts in haemopoietic tissues. The majority of peripheral blood cells are known to possess an ability to interact with each other (aggregation phenomenon) or with various artificial materials, which has important implications for medicine, particularly in the fields of thrombosis, vascular prosthetics and dialysis. On the other hand, recent studies indicate that blood cells can interact with the main important components of ECM, such as collagen, fibronectin, etc. These data, as well as the newly discovered receptor structures for various adhesive proteins on the surface of blood cells suggest that the haemopoietic tissue has a diverse function involving interaction with the ECM.

Quantitative problems in bone marrow transplantation by peripheral blood stem cells.

Investigation into radiation bone marrow aplasia in mice, guinea pigs, dogs and clinical trials in man presented clear evidence of successful engraftment of autologous or allogeneic peripheral blood stem cells. The quantitative donation problems are discussed arising with the use of continuous cytopheresis to obtain a sufficient quantity of peripheral blood mononuclears (stem cells) for repopulation of aplastic bone marrow. Although bone marrow repopulation is possible by using peripheral blood mononuclears (stem cells) in individual cases, the method can only be used in practice after discovering an appropriate stimulator able to augment several times the number of bone marrow stem cells in the peripheral blood, or a new method for stem cell multiplication in vitro.

Function and role of peritoneal macrophages in experimental leukaemia.

The specific and nonspecific function of peritoneal macrophages of leukaemic mice has been studied as well as the role of these cells in the induction of lymphocyte cytotoxicity against leukaemic cells in vitro. It was found that the phagocytic and digestive function of the peritoneal macrophages remains normal, but the macrophage-mediated cytotoxicity against leukaemic cells is rare and weak in both allogeneic and syngeneic forms of grafted leukaemia. Utilizing the system of generation of cytotoxic lymphocytes in mixed lymphocyte tumour cell cultures in vitro, it was observed that the addition of specifically activated syngeneic macrophages lowers the rate of generated cytotoxicity. The mechanism of macrophage-induced suppression is discussed.

[Radiometric method of determining the phagocytic activity of macrophages in vitro].

The authors describe a radiometric method for determination of phagocytic activity of macrophages, using as an object for phaging hen erythrocytes labeled with 51Cr. The basic principle consists of quantitative determination of phaged cells by means of measuring the activity, found in the phagocytes. The method is described in details and the obtained results in normal mice and such with experimental leucosis NKLI are compared with those, checked by the classical methods for determining phagocytic activity in vivo and in vitro. On the basis of these data the authors indicate the advantage of the method, prepared by them, because it alows objective checking of phagocytosis on a large number of cells and thus increases the possibilities of obtaining significant results.

Kinetics of mononuclear phagocytes after thymectomy.

Some kinetic parameters of mononuclear phagocytes have been studied. Wistar rats were injected with 3H-thymidine. Samples of peripheral blood and peritoneal macrophages were obtained at 12, 14, 24, 36, 48, 60, and 84 hours respectively after injection. The labelling index and the mean grain count were determined. The turnover of monocytes was slowed down in the thymectomized animals. It is assumed that thymectomy had changed some biological qualities of mononuclear phagocytes and/or the humoral relations connected with the function and the regulation of mononuclear phagocytes.

[Action of the sera from acute leukemia patients on lymphocytic transformation].

The authors examined in vitro the action of sera, obtained from patients with acute leucosis during the first attack and relapse on PHA (phytoheagglutinins)-M-reactivity of lymphocytes, obtained from healthy donors and olymphocytes obtained from leucemic patients. They found that sera inhibited in vitro the effects of PHA-M both on lymphocytes of patients and of healthy donors. There was stimulating effect of sera in vitro in single cases under autologic conditions.

[Search for embryonic antigens in the serum of leukemia patients].

The authors examined patients with blastic leucosis, myelosis and lympholeucosis for the presence of specific embrional antigens by immunodiffusion, using antisera, obtained after immunization of rabbits with extracts from human embryos and absorbed with tissue extracts from a normal adult. There were no specific embryonal components in the patients with leucemic sera or in the women from the control group.

Kinetics of immunoglobulin G in plasmapheresis of non-immunized and immunized donors.

The balance of immunoglobulins G has been studied during plasmapheresis in donors immunized and non-immunized with D antigen. For 42 days, the life-span of IgG, rhythmical withdrawal of plasma was carried out. During the procedure, 50% or 100% of the extracellular IgG was lost. A rhythmic loss of IgG amounting to double its production is the upper limit which can still sufficiently be compensated by the organism.
