The purpose of this study was to determine whether association of vasoactive intestinal peptide with sterically stabilized liposomes (VIP on SSL) amplifies DNA synthesis evoked by the peptide in cultured chemically transformed hamster oral keratinocytes (HCPC-1) and, if so, whether this response in mediated, in part, by SSL-induced inactivation of neutral endopeptidase 24.11 (NEP; EC 3.4.24.11) and angiotensin I-converting enzyme (ACE; EC 3.4.15.1), two ectoenzymes that modulate HCPC-1 cell growth, in these cells. We found that VIP (10(-9)-10(-6) M) alone elicited a modest, albeit significant, concentration-dependent increase in DNA synthesis in HCPC-1 cells that was maximal after 48-72-h incubation (p < 0.05). VIP on SSL potentiated DNA synthesis in these cells relative to VIP alone. The magnitude of VIP on SSL-induced responses was 1.2-1.6-fold higher than that of VIP alone with maximal effects observed at 10(-9) M and 10(-6) M after 72- and 48-h incubation, respectively. Empty SSL had no significant effects on DNA synthesis. Empty SSL and VIP on SSL had no significant effects on NEP 24.11 and ACE activity in HCPC-1 cells. Collectively, these data indicate that association of VIP with SSL potentiates DNA synthesis in cultured oral keratinocytes relative to VIP alone and that this response is not related to non-specific effects of SSL.
The purpose of this study was to determine whether human vasoactive intestinal peptide (VIP) aggregates in aqueous solution and, if so, whether the peptide interacts with a biomimetic phospholipid monolayer and increases surface pressure. Using a custom-made Teflon trough containing HEPES buffer (pH 7.4) at room temperature and a surface tensiometer, we found that the critical micellar concentration (CMC) of VIP is 0.4 microM. Surface pressure of a dipalmitoylphosphatidylcholine (DPPC) monolayer spread over the HEPES buffer declined significantly over 120 min because of phospholipid decomposition. However, injection of VIP at concentrations above CMC into the subphase of the monolayer elicited a significant concentration-dependent increase in surface pressure that persisted for 120 min (P < 0.05). Unlike VIP, injection of [(8)Arg]-vasopressin at an equimolar concentration only prevented the time-dependent decline in DPPC monolayer surface pressure. Taken together, these data indicate that human VIP aggregates in aqueous solution and expresses surface-active properties at physiological concentrations in vitro. We suggest that these attributes could have a role in modulating the bioactive effects of the peptide in vivo.
Small cell neuroendocrine carcinoma of the cervix is very rare and is usually associated with dismal prognosis if treated by conventional surgery and radiotherapy even in early stage disease. This tumor is characterized by early lymphatic and hematogenous spread. Only one successfully treated case of small cell neuroendocrine carcinoma of the cervix with dissemination to the pelvic nodes had been reported before in the literature. We are reporting a case of small cell neuroendocrine carcinoma of the cervix with multiple pelvic nodal metastases including the common iliac nodes, which had been successfully treated with a multimodal approach including radical hysterectomy, pelvic/para-aortic lymphadenectomy, and postoperative chemotherapy using cisplatin-etoposide combinations and pelvic irradiation.
A 58-yr-old woman with a right atrial myxoma associated with a vascular malformation supplied by the right coronary artery is described. She presented with atypical chest pain that was reproduced during angiography of the malformation. Findings were confirmed during surgery.
Strategic information systems planning (SISP) is the process of deciding the objectives of computing for an organization and then identifying the applications that the organization should computerize. SISP has become increasingly important as information systems have begun to play a more critical role in implementing business strategies. However, SISP is beset with problems that hinder organizations from determining their computing objectives and applications. This article identifies the impediments to SISP and offers some constructive actions for business planners to take to increase their chances of success. It also suggests that planners may face greater difficulties implementing their information systems plans than in initially creating them.
This paper addresses both the increased use of marketing by hospitals and frustrations which have more recently been felt with such programs. A series of seven factors are presented as suggested explanations of current dissatisfaction with the effectiveness of marketing efforts in the hospital of the late 1980s.
Radioactive sources of finite volume containing 133Xe, 67Ga, 99Tcm and 111In were used to measure the attenuation coefficient, mu, in water at six different energies in the range 80-296 keV using an Anger camera. The experimental accuracy was about 7% for the volume range from 40-225 ml when corrections were made for background. The same radioactive sources were used to measure zero attenuation count rates per unit of activity. The theoretical basis was also derived, which confirms our experimental findings, i.e. the measurement of the thickness of the attenuator using a dual energy method. The determination of the linear attenuation coefficient in the broad-beam geometry situation is possible by accounting for cross-talk, scatter and out-of-target activity. By correcting for the broad-beam geometry, agreement with the narrow-beam geometry linear attenuation coefficient was obtained. We also demonstrate the use of the technique to accurately determine the depth of the organ using two separate energies. This methodology is independent of the organ volume for determination of the depth. It is hoped that our findings will provide a better understanding of the photon interactions when extended sources are used. Such a knowledge can also be applied to organ volume measurements.
Twenty-four patients with classical angina of effort completed this double-blind cross-over clinical trial of acebutolol and propranolol. Both acebutolol (100-400 mg) and propranolol (40-160 mg) three times a day produced a significant reduction in the incidence of anginal attacks, consumption of nitroglycerine tablets and increased exercise tolerance. Both drugs produced mild and tolerable side effects which did not require any reduction of doses of the drugs being used. No change in laboratory parameters assessing the function of the liver, the kidneys and the hemopoietic system was found. The commonly accepted dose ratio of 1:2.5 propranolol and acebutolol, respectively, was found to be true in less than 50% of the patients. Therefore, for optimal effect, the dose of acebutolol should be individualized depending upon the response.
Twenty patients with mild to moderate essential hypertension completed this double-blind cross-over randomized clinical trial of propranolol and acebutolol. Acebutolol was found to be as effective and safe as propranolol in lowering the systolic and diastolic blood pressure both in supine as well as in standing positions. However, the difference between the two drugs was not statistically significant. No significant change was produced in serum electrolytes, uric acid, fasting blood sugar with either drug. The accepted dose ratio of 1:2.5 propranolol and acebutolol, respectively, was found to be true only in 50% of the patients. Therefore, for optimal effect, the dose of acebutolol should be individualized depending upon the response.
Noncardiogenic pulmonary edema and acute respiratory failure were found to develop after blood transfusion. Pulmonary antiographic studies and hemodynamic measurements were performed in the presence of pulmonary edema. Normal wedge pressure was observed at the time of angiographic studies, and contrast material appeared to extravasate across the alveolocapillary membrane into the alveoli. The mechanisms of acute injury to the alveolocapillary membranes and of pulmonary edema secondary to reactions to transfusion are discussed.
