[Effects of DL-difluoromethylornithine on the cell surface charge of Ehrlich ascitic cancer during different phases of the mitotic cycle].

Difluoromethylornithine (DFMO)-induced polyamide deficiency has been studied for its effect on the electrophoretic mobility of the Ehrlich ascitic cells in G1, S and G2 phases of the mitotic cycle. The DFMO treatment alters the pattern of the electrophoretic mobility distribution of Ehrlich ascite cells. The most profound changes of the electrophoretic mobility pattern are observed in G1-phase and at the S phase beginning. Exogenic polyamines decrease the electrophoretic mobility of cancer cells, which is accompanied by restoration of the initial pattern of the electrophoretic mobility distribution.

[Structural dynamic properties of the cell membranes of Ehrlich ascites tumor associated with a polyamine deficiency caused by DL-difluoromethylornithine].

The blocking of polyamine synthesis in cancer cell by a specific inhibitor (DFMO) results in an increase in membrane viscosity and in significant changes in cell deformability as determined by cell penetration through a nuclear filter with given porosity. Almost two-fold reduction of the penetration rate through the filter pores was registered for the DFMO-treated cells in spite of the fact that the cell size decreases after such a treatment. The number of cells penetrating through the filter after DFMO treatment is diminished due to an increasing resistance to penetration. The addition of putrescine to the incubation medium at physiological concentration restores all these parameters to the initial values. The possible association of these changes with differential sensitivity of cancer cells towards the inhibitor action depending on the stage of cell cycles is discussed.

[Adsorption on charcoal sorbents of the nucleic acids from the blood plasma of leukemia patients and from the ascitic fluid of mice with lymphoma NK/Ly].

Using 3H-thymidine, 14C-orotic acid and ethidium bromide identification of the nucleic acid double-stranded structures from biological fluids and the method of hemocarboperfusion the data are obtained on the active and irreversible adsorption of nucleic acids and nucleoproteins from plasma of leukaemic patients and ascitic fluid of mice with lymphoma NK/Ly.

[Characteristics of the nucleic component of a nucleoprotein complex secreted by tumor cells].

The characteristic of the nucleic component from the nucleoprotein complex excreted by tumour cells into ascitic fluid is presented. Using enzymes which hydrolyze nucleic acids and applying the methods of cellulose chromatography and ethidium bromide identification of the double-stranded structure of nucleic acids it is shown that the nucleic component of the excreted complex is a heterogeneous system presented by double-stranded structures which involve DNA-RNA hybrid molecules.

[A case of Meigs' syndrome].

The described case of Meigs' [correction of Maigce] syndrome supports data on diversity of exudative pleurisies as some syndrome of manifestation of different pathological processes. Diagnosing and treatment by surgery technique of the female patient with Meigs' [correction of Magice] syndrome suggest expediency of examination by an obstetrician-gynecologist of all those women presenting with exudative pleurisy.

[Polysome ribonucleases of leukemic cells].

The composition of ribosomal proteins isolated from normal homopoietic and leucemic cells was studied by analytic electrophoresis in polyacrylamide gel. It was found that acid-soluble proteins from polysomal complexes can be separated into 21-22 components in the acid system. There was no significant differences in protein components from normal and leucemic polysomes by their quantity and mobility in polyacrylamide gel. Five protein components possess a ribonuclease activity as it was established by using technique of direct RNase assay in electrophoregrams. All ribonuclease-active components have pH optimum at 7.6-7.8. No differences were detected in the number and activity of particular enzyme components of polysomes isolated from normal hemopoietic and leucemic tissues. It is suggested that the number and presence of RNases in polysome complexes is likely to play not a significant role in regulation of polysome activity, but influence of inhibitors makes this supposition possible.

[Ribosomal apparatus of liver cells in the process of carinogenesis induced by 4-dimethylaminoazobenzene].

A content of membrane-bound and free ribosomes, sedimentation distribution of total polyribosomes in a linear sucrose density gradient and their functional activity in a cell-free system of protein synthesis were estimated in liver of animals feeded with protein-poor rice diet containing a cancerogen, 4-dimethylaminoazobenzene (DAB) at a dose of 15 mg per day. Periods of the experiments were 1-10 days, 1-2, 3-4 and 5 months from the beginning of DAB treatment. The decrease of protein-synthesizing activity, the decrease in the content of membrane-bound ribosomes and the oncrease in the content of free ribosomes in animal liver were observed at first days of DAB action. Changes in the quantitative ratio between membrane-bound and free ribosomes (in the direction of decreasing the content of membrane-bound ribosomes) was observed at all the stages of DAB-cancerogenesis. It is supposed that the uncoupling of ribosome-membrane bonds in endoplasmic reticulum takes an important part in cancerogenesis, disturbing intracellular regulation of protein synthesis.
