[Dependence of gravitotropic reaction in segments of flax stems on frequency and amplitude of variable components of a weak combined magnetic field].

We determined the dependence of the effect of weak combined magnetic field (CMF) tuned to Ca2+ resonance (Ca(2+)-CMF) on the rate of gravitropic response in apical segments (25 mm long) excised from the 4-day-old seedlings of flax (Linum bienne) on the amplitude and frequency of the alternating component of Ca(2+)-CMF. The results indicate that test-systems derived from plant and animal sources are affected by Ca(2+)-CMF via an identical mechanism.

[Activation and inhibition of the gravitropic response in the flax stem segments exposed to the permanent magnetic field with magnetic density ranging from 0 to 350 microT].

It was found that the rate of gravitropic bending in apical segments excised from 4-day-old etiolated seedlings of flax Linum bienne is strongly and nonlinearly dependent on the value of the magnetic flux density of a static magnetic field, BDC, in the range from 0 to 350 microT. The gravitropic bending is stimulated at 0 < or = BDC < or = 2 microT and 200 < or = BDC < or = 350 microT but is inhibited at 100 < or = BDC < or = 170 microT relative to control samples being in the magnetic field of the Earth equal to 46.5 microT.

[Effects of extremely weak alternating magnetic fields on the plant gravitropism].

It was shown that the rate of gravitropic response in apical segments excised from the 4-day-old seedlings of flax (Linum bienne) may be substantially influenced by combined magnetic fields (CMF) of two different types: 1) CMF tuned to the parametric resonance for Ca2+; 2) CMF containing extremely weak alternating component with the values of magnetic density ranging from 10(-6) to 10(-10) T. Our experimental data indicate that CMF affect the gravitropic response via at least two different mechanisms. The first one corresponds to the ion parametric resonance well established earlier in studies with test-systems prepared from animals. The origin of the bioeffects induced by CMF containing extremely weak alternating component remains to be established.

[Chemotaxis as a method for testing of the biological effects of silver nanoparticles].

A method for assessing the abiotic efficiency of water-dispersed nano-sized silver particles is suggested. Nanoparticles were obtained by the method of biochemical synthesis in reversed micelles made from anionic surfactant bis-(2-ethylhexyl) sodium sulfosuccinate (AOT). A water dispersion of nanoparticles was prepared from the micellar solution in organic solvent by means of a special procedure. The abiotic efficiency assay is based on the capacity of cells to respond by the negative chemotaxis to chemical agents with harmful metabolic action. In plasmodium of myxomicete Physarum polycephalum, the biocide and repellent effects of silver nanoparticles, Ag+ ions, and AOT were tested in order to compare the abiotic efficiency of these substances in water solution and after introduction into agar substrate. The increase in the oscillation period, the reduction in the area of spreading, and the avoidance reaction in the spatial test, taken together, revealed a much higher repellent efficiency of silver nanoparticles as compared with that of Ag+ ions and AOT. The NSP concentrations lethal for Physarum were similar to those found earlier for bacteria and viruses. The chemotaxis-based tests applied in this study allow one to quantitatively assess cell reactions and monitor their time course. Besides, they have a much higher resolving capacity than the tests based on lethal effects of abiotic substances.

[The structure of thick filaments on longitudinal sections of rabbit psoas muscle].

By means of electron microscopy the longitudinal sections of chemically skinned fibres of rigorised rabbit psoas muscle have been examined at pH of rigorising solutions equal to 6, 7, 8 (I = 0.125) and ionic strengths equal to 0.04, 0.125, 0.34 (pH 7.0). It has been revealed that at pH 6.0 the bands of minor proteins localization in A-disks were seen very distinctly, while at pH 7.0 and I = 0.125 these bands can be revealed only by means of antibody labelling technique. At the ionic strength of 0.34 (pH 7.0) the periodicity of 14.3 nm in thick filaments was clearly observed, which was determined by packing of the myosin rods into the filament shaft and of the myosin heads (cross-bridges) on the filament surface. The number of cross-bridge rows in the filament equals 102. A new scheme of myosin cross-bridge distribution in thick filaments of rabbit psoas muscle has been suggested according to which two rows of cross-bridges at each end of a thick filament are absent. The filament length equals 1.64 +/- 0.01 micron. It has been shown that the length of thick filament as well as the structural organization of their end regions in rabbit psoas muscle and frog sartorius one are different.

["Freezing" of the conformation of thin filaments in a single skinned fiber of the rabbit muscle by glutaraldehyde].

It is shown that short treatment of a single skinned rigor fibre from rabbit m X psoas with 0.05% glutaraldehyde in the absence of Ca ions leads to a modified state of the contractile apparatus. After the addition of 5 mM MgATP in the absence of Ca ions to the fibre a sharp rise and subsequent slow decay of tension were observed in contrast to the tension drop in case of the control (unmodified) specimen. The tension transients following quick stretch (L 0.5%) were similar to those for Ca-activated tension. In case of the modified relaxed fibre such a phenomenon was not observed. These results can be explained by "freezing" with glutaraldehyde the thin filament structure either in the "on" or "off" states. The relation of these results to the cooperativity in the regulation mechanism of contraction is discussed.

[Functional role of the structural nonequivalence of the F-actin subunits of thin filaments].

According to the model of the actin-containing filaments suggested earlier by Frank and author thin filaments of sceletal muscle can be considered as a linear polymer chain composed of structural units each of which can adopt either of two different structural states -- I (inhibited) and A (activated). The structural unit can be represented by a block of two complementary functional units of the thin filaments. The data available indicate that the length of the A-state is slightly longer than the length of the I-state. In this case the relative amount of the functional units in A- and I-states in thin filaments must depend not only on the Ca2+ concentration in sarcomere, but also on the value of the external force pulling the thin filaments in contracting muscle. It is shown that this property of the thin filaments may provide the molecular basis of Hill-equation and the selfadaption of contractile machinery to the value of the external load on muscle.

[X-ray study of the effect of pyrophosphate on thick filament structure in skinned fiber bundles of the rabbit psoas muscle].

Structure of thick filaments in the chemically skinned fibre bundles of rabbit psoas muscle in a state of pseudorelaxation induced by adding 2 mM pyrophosphate (PP) and of PP-mixture with 40% ethyleneglycol to the bathing rigor solution was studied with the help of X-ray diffraction technique. Reduction in the isometric rigor tension by about 50-70% in a state of pseudorelaxation is accompanied by significant changes in the relative intensities of a number of meridional reflections, indicating that in situ the structure and location of S-2 segment may be regulated by the structural changes in the acto S-1-complex during its cyclic interaction with ATP.

[X-ray diffraction study of the molecular nature of the dependence of rigor tension developed by skinned rabbit psoas muscle on the ionic strength of the solution].

Molecular origin of the dependence of rigor tension developed by skinned rabbit psoas muscle on the value of ionic strength in rigorizing solution was studied by means of X-ray diffraction technique. It is shown that a deviation of the ionic strength from "normal" value (= 0.125) either to high (= 0.34) or to low (= 0.04) values is accompanied by the changes in the relative intensities of some meridional reflections while their axial positions remained unchanged. This may be explained as the result of the detachment of myosin subfragments-2 from the surfaces of thick filaments. The data obtained indicate that there is a correlation between the structure and localization of myosin subfragments-2 and the value of force generated by the muscle.

[X-ray study of pH-dependence of thick filament structure in stripped fiber bundles of vertebrate skeletal muscles].

Molecular origin of pH-dependence of rigor tension in chemically skinned fibre bundles of rabbit psoas muscle was studied with the help of X-ray diffraction technique. It was found that a shift of pH from the neutral value of about 7.0 either to basic or to acidic regions by one unit is accompanied by changes of relative intensities in a number of meridional reflections. These effects are explained as the result of pH-induced detachment of subfragments-2 of myosin molecules from the thick filament surface. The data obtained indicate that force generation in muscle may be caused by the structural changes of subfragments-2 of myosin molecules.
