[Pathogenetic implications of lipid spectrum in some infectious diseases].

AIM: To estimate concentrations of blood serum lipids and erythrocytic membranes in the conditions of intoxication syndrome. MATERIAL AND METHODS: Lipids were studied in the serum and erythrocytic membranes of 48 patients with oropharyngeal diphtheria and 67 patients suffering from hemorrhagic fever with renal syndrome (HFRS) in different forms and severity of the disease. RESULTS: Various correlations were found between structural and serum lipids. This is pathogenetically important for characterizing biomembranes and variants of activation of lipid metabolism in conditions of intoxication syndrome in diphtheria and HFRS. CONCLUSION: At the height of diphtheria and HFRS organization of biomembrane phospholipids conformation creates conditions for cell disorganization.

[The phospholipid spectrum of erythrocyte membranes in hemorrhagic fever with renal syndrome].

The subjects of the study--patients with severe hemorrhagic fever with renal syndrome--were divided into two groups: those who were on hemodialysis, and those who were not. The study included evaluation of the phospholipid spectrum of erythrocyte membranes in the acute period and during recovery. The results revealed conformational shifts in the structure of the bilipid membrane layer, which were maximal during the acute phase of the disease, as well as less prominent and varied changes in the phospholipid spectrum during recovery. This allows determination of the terms of rehabilitation of the patients and substantiates administration of membrane stabilizers as a part of complex therapy of residual syndrome.

[Complex pathogenetic therapy of experimental botulism].

The study of survival and life span of mice and white rats upon the administration of LD50 of the toxin has shown that an antihypoxic agent--gutimine (50-200 mg/kg)--had a protecting effect in type C botulinum intoxication. A combined use of gutimine and 4-aminopyridine (1-5 mg/kg), facilitating a transmitter release in synapses, had a more marked protecting effect in botulinum intoxication. Due to the potentiation of the drugs effect during their combined application, the doses of each drug in the combination could be reduced.

4-Aminopyridine↗

[Effect of 4-aminopyridine on the development of experimental botulism].

The studies of survival and the life span of mice and the toxin in LD50 have shown that 4-aminopyridine (4-AP) at doses of 1, 2 and 5 mg/kg has a therapeutic effect on type C botulinic intoxication. Maximum protective effect is observed when 4-AP at a dose of 2 mg/kg is injected twice daily. Therefore, 4-AP, enhancing the transmitter release into cholinergic synapses, weakens the toxic effect of botulinic toxin.

4-Aminopyridine↗

[Changes in the electrogenic properties of striated muscle fibers during experimental botulism].

Changes in the electrical parameters of fibers of fast and slow muscles of rats during development of local botulism were studied. At the early stage of intoxication a decrease of the membrane potential of the fast and slow muscle fibers was found. At the stage of intoxication, alongside with an expressed membrane depolarization, there was a change of the input resistance, of the membrane electrical time constant, the threshold currents and the amplitude of action potentials, caused by direct intra-cell stimulation. Changes in the electrical parameters are more expressed in the fibers of the fast muscle.

[The lipid peroxidation processes and the antioxidant system in the dynamics of an experimental Pseudomonas aeruginosa intoxication].

Lipid peroxidation and antioxidant defense processes have been studied over the course of experimental Pseudomonas aeruginosa intoxication in albino rats injected with a lethal dose of exotoxin A. The development of intoxication is associated with intensification of lipid peroxidation, manifested by accumulation of malonic dialdehyde and diene conjugates and decreased peroxide resistance of erythrocytes. Superoxide dismutase and catalase activities were inhibited and the concentration of vitamin B dropped. Injection of antioxidant alpha-tocopherol or antihypoxant gutimine at the late stage of intoxication did not notably modify the studied parameters.

[Mechanism of impairment of energy metabolism in respiratory muscles in experimental botulism].

Phase alterations in content of adenylic nucleotides were observed in muscular and nervous tissues in dynamics of botulinic intoxication in rats. Experiments with administration of 4-aminopyridine and alpha-tocopherol showed that a decreased rate of mediator liberation in cholinergic synapses and activation of lipid peroxidation were apparently responsible for impairments in energy metabolism.

[Mechanism of the change in energy and electrolyte metabolism in the respiratory musculature in experimental botulism].

Distinct decrease in concentrations of creatine phosphate and K+ was observed in various respiratory muscles of cat in generalized botulism, accompanied by the syndrom of asphyxia. Content of glycogen and Na+ was unaltered in these cases. In local botulinic paralysis concentrations of creatine phosphate, glycogen and K+ were decreased in the respiratory muscles. Accumulation of Na+ was observed in diaphragm and intercostal muscles. Content of K+ and creatine phosphate was partially restored in respiratory muscles after intraperitoneal administration of guanidine HCl/50 mg/kg of body weight/.

[Change in the histamine and serotonin levels in the external respiratory apparatus in botulinic poisoning in cats].

General botulinic intoxication in cats, accompanied by severe respiratory insufficiency, was characterized by distinct impairments in histamine and serotonin metabolism in external respiratory system. Pronounced increase in content of histamine was observed in impaired muscles and their corresponding regional centers of medulla spinalis, when botulinic intoxication had the local type with unilateral paralysis of diaphragm and intercostal muscles. Content of serotonin was increased in the tissues studied only at the later steps of local botulinic intoxication.

[Role of hypoxia factor in changes of ionic composition of blood and muscle tissue in experimental botulinum poisoning].

Levels of sodium and potassium ions in muscle tissue, blood plasma and red cells were measured, and the index characterizing the capacity of tissue to accumulate cations from the environment and the discrimination coefficient were calculated in experiments on white rats subjected to experimental generalized botulinum poisoning. Late stages of botulism were found to involve a deficit of potassium and sodium ions in red cells. With the progress of intoxication, a deficit of potassium ions develops in muscle tissue and the capacity of this tissue to accumulate potassium ions from the environment declines. All these disorders are more expressed in rapidly contracting skeletal muscles. An antihypoxant gutimin reduces sodium deficit in the blood plasma and red cells and leads to normalization of potassium ion levels in muscle tissue.
