[Modification of the ligand load and structure of human serum albumin with different methods of isolation].

The influence of isolation procedures on physico-chemical properties of human serum albumin (HSA) has been studied using differential scanning calorimetry and infrared spectroscopy. Fractionation of blood plasma with polyethylene glycol (PEG) followed by ion-exchange chromatography produced healthy donor albumin whose melting thermograms were identical to those of the HSA in non-fractionated plasma. The endotherms of HSA samples isolated by affinity chromatography (AC) and native electrophoresis (EP) are bimodal as distinct from monophasic endotherms of PEG-HSA preparations. The changes in the melting curves result from increased levels of nonesterified fatty acids in AC-HSA samples and modification of the protein secondary structure revealed in EP-HSA samples by IR-spectroscopy. Analysis of HSA melting thermograms in patients with uremia, chronic hepatitis and peritonitis revealed that the PEG method permits to preserve, in the greatest degree, the thermodynamic features of various "pathological" HSA. The experimental results demonstrate the advantage of the PEG technique in isolation of native HSA in the norm and under pathology.

[A method of determining middle molecules].

The authors suggest a method for the detection of 'medium molecules' to be used for rapid diagnosis of poisoning. They present their arguments in favor of a higher specificity of this method as against other rapid methods for 'medium molecule' detection in biologic fluids of the body. The authors' data evidence a high correlation between the results of the suggested method and gel filtration technique. The procedure may be used for the rapid diagnosis of peptidemia severity in the patients with grave endogenic intoxication and in cases when endotoxicosis is not the underlying condition.

[Middle molecules--formation and methods of determination].

Medium molecules have been detected by two methods, gel filtration and screening technique, in patients with diffuse purulent peritonitis and with chronic renal insufficiency. No close correlation between the findings of the two methods has been observed. The results lead to a conclusion that, despite being rapid and simple, the screening method is hardly adequate for cases with endogenous intoxication. Approaches to improvement of this method are discussed.

[Disorders of protein metabolism in neonatal hyperbilirubinemia and their correction by extracorporeal detoxification].

A close relationship was established between the gravity of bilirubin intoxication, dysproteinemia and hyperpeptidemia in neonates. It is shown that accumulation of peptides belonging to the medium molecule group and associated pathophysiological manifestations may account for a graver course and less favourable prognosis of hemolytic disease of the newborn as compared to conjugation hyperbilirubinemia. It is suggested that the measures aimed at bilirubin removal by the control of the medium molecule effects should be supplemented. The results of hemosorption (51 sessions in 42 children) and substitution transfusion (27 operations in 24 children) were subjected to clinical and laboratory comparisons. In addition to bilirubin removal from the body, hemoperfusion permits the correction of protein metabolism abnormalities occurring in hemolytic disease.

[Peptides of the "middle molecule" group].

The "middle molecules", endotoxins of peptide nature appearing in biological fluids in several diseases, cause the disorder of many regulatory processes, suppress the functions of blood cells, affect the transport characteristics of cell membranes. The review covers different aspects of formation, structure and numerous biological effects of "middle molecules", including the molecular mechanism of their biological action.

[Complications of extracorporeal hemosorption and their prevention].

The authors have analyzed complications in 395 hemosorptions in 189 patients with different diseases, burns being most frequent one associated with hypocalcemia resulting from great doses of heparin. Trental and small doses of heparin are proposed for prevention of the complications.

[Spatial structure of a vasoactive peptide--a fibrin fragment].

Empirical energy calculations were used to determine all low-energy conformations of vasoactive pentapeptide Ala-Arg-Pro-Ala-Lys, thereby three most stable conformations were distinguished. Biological testing of conformationally restricted analogs allowed to delineate the most probable "biologically active" conformation of the molecule.

[Effect of RNA and histone preparations on the properties of lymphoid cells].

Preparations of RNA and histones change the electrophoretic mobility of lymphoid cells, the intensity of their respiration and anaerobic glycolysis, and thermo-merchanical properties of DNA-complexes isolated from lymphoid cells. RNA and histones exert their influence on the properties of DNP-systems and the rate of anaerobic glycolysis in the opposite manner. But both these preparations depress the electrophoretic mobility and oxygen consumption.

[Physico-chemical properties of the DNP-systems of leukemia cells with different chromosome numbers].

The binding between the components of the isolated DNP-complex proved to be more stable in leukemic cells in comparison with the normal lymphoid spleen cells. This conclusion was based on the investigation of the thermomechanical and disaggregation properties of the supramolecular DNP systems isolated from leukemic cells. The change in the isolated DNP-complex were independent of the subpopulation variants of the leukemic lymphatic cells and their chromosome set (diploid or hyperdiploid). The observed changes in the chromatin properties were more specific for leukemic cells than the changes in thechromosome number.
