[Lectins, adhesins, and lectin-like substances of lactobacilli and bifidobacteria].

Cell-surface adhesion factors of lactobacilli and bifidobacteria, such as lectin/adhesin proteins of S-layers, secreted lectin-like bacteriocins, and lectin-like complexes, are considered and classified in the article. Certain general and specific properties of these factors are noted, such as in vitro and in vivo adhesion, cell co(aggregation), participation in the forming of microbial biofilms and colonization of mammalian alimentary tract, as well as complexation with biopolymers and bioeffectors, specificity to glycanes and natural glycoconjugates, domain and spatial organization of adhesion factors, co-functioning with other cytokines (pro- and anti-inflammatory ones), regulation of target cell properties, and other biological and physiological activities. The authors also note possibilities of application of lectins and lectin-like proteins of probiotic strains of lactobacilli and bifidobacteria in medicine and biotechnology.

[The protective activity of a new variant of the probiotic Acilact in exposure to ionizing radiation and anticancer chemotherapy under experimental conditions].

The aim of the study was to investigate the protective activity of a new variant of the probiotic Acilact in exposure to ionizing radiation and anticancer chemotherapy under experimental conditions. The study revealed protective effects of the preparation after combined exposure of mice to radiation and temperature. The preparation increased 30-day survival of the mice, inhibited intestinal colonization with gram-negative microorganisms, and increased resistance to LPS. A multidirectional protective effect of the probiotic was observed after the use of sublethal doses of radiation; this effect was manifested by quicker normalization of hematological parameters and bone marrow cellularity, as well as elevation of blood concentration of the hemoragulatory cytokines IL-3 and IL-6.

[Monkeys as subjects for study of immunobiological preparations].

The parameters of colon microbiocenosis were evaluated in healthy animals belonging to 6 species of marmosets; its disorders typical of patients with different forms of acute intestinal infections were revealed. Bifidoflora of young monkeys was characterized. The study demonstrated multiple similarities of the microflora of monkeys and humans, including small children. A preclinical study of dry bifidin, a probiotic, revealed its advantages over the commercial bifidumbacterin. 90 monkeys were used for preclinical evaluation of complex immunoglobulin preparation (CIP) and a combination of CIP with interferon (kipferon in suppositories). The study demonstrated sanifying and microflora-normalizing effects, increase of anti-microbial antibodies and serum interferon after both parenteral and rectal administration. [Biological characterization and prospects of application of a new bifidobacterial strain as an ingradient of probiotics].

The study was dedicated to investigation of a bifidobacterial strain, isolated from a healthy chick, its biological properties and lectins produced by the strain; a restrictive analysis of the chromosomal DNA was conducted as well. Presumably B. gallinarum was identified. The strain displayed adhesive, lectinic and antagonistic activities; it was resistant to the leading chemotherapeutic pharmaceuticals and antibiotics, and, when given to chicks, it had probiotic effect. The study identified a set of lectins binding phosphorylated mannan, with isoelectric points in the pH interval of 4 to 8; the set differed from the sets of lectins of other bifidobacteria.

[Formation of probioticotherapy in Russia].

Probioticotherapy and probioticoprophylaxis have been applied widely in Russia and the world of late. The article presents the main medical, strain-related, and biotechnological stages of their development in this country, in comparison with the results of the world practice. The authors substantiate appropriateness of creation of probiotics and which would contain either only growth medium components, able to activate normoflora (to increase colonization ability and stimulate production of biologically active substances) upon entering the organism, or only probiotic strain components and/or biologically active substances produced by biologically active strains in mono- or associative cultivation, or combination of the listed components.

[The effects of the microbial components of the probiotic Acilact on the cell-mediated immunity factors under experimental conditions].

The authors analyzed the effects of the microbial components of the probiotic Acilact on the cell-mediated factors of the immune homeostasis. According to the results, L. acidophilis stimulates the functional activity of immunocompetent cells, especially T-cell mediated immunity with strain- and dose-depending differences in intensity. The study demonstrated the ability of L. acidophilis to activate expression of the genes of pro-inflammatory cytokines (IFNgamma, IL-6, 12, and TNFalpha), to induce production of IFNalpha and IFNgamma, and increase interferon production, with additional stimulation of lymphoid organ cells with specific inductors in vitro. The study established that L. acidophilis stimulates production of macrophage migration inhibition factor by immunocompetent cells of Peyer's plaques and the splen, and induces its quantitative increase in the serum of CBA mice. The study detected immunocorrecting effect of L. acidophilis by the example of experimental shigellous infection, demonstrated intensification of the prolipherative activity of immunocompetent T-cells, induction of the expression of the genes of pro-inflammatory cytokines (IFNgamma, IL-1beta, 6, 12, and TNFalpha), but not IL4 and 10, increase of synthesis of IFNalpha and IFNgamma by the cells of Peyer's plaques, the splen and thymus. The obtained results prove the immunological effect of the probiotic preparation Acilact, based on the strains L. acidophilis NK1 and L. acidophilis K3(111)24.

[The adhesive properties of bacteria of intestinal origin].

Differences between strains of nonpathogenic Escherichia and lactobacilli, as well as some pathogenic bacteria of enteric origin (Escherichia, Shigella, Campylobacter), in their capacity to adhesion to rat enteric and colonic cells have been shown in vitro. The strains under study have been found to possess more pronounced adhesiveness with respect to colonic cells, which is indicative of their higher receptive capacity in comparison with enteric cells. In the absence of normal microflora lactobacilli and Escherichia exhibit increased adhesiveness with respect to enteric cells. Escherichia enterotoxigenic strains, Yersinia enterocolitica and Salmonella typhimurium virulent strains, Campylobacter jejuni clinical isolates possess more pronounced capacity for adhesion to enteric cells of Peyer's plaques than to other types of epithelial cells, which may be of importance in the pathogenesis of these infections.

[Mechanism of antagonistic activity of lactobacilli].

Different species of lactobacilli, when grown together with salmonellae and shigellae, have been found to inhibit their growth and proliferation. The supernatants of the culture fluids of 8 species of lactobacilli have been shown to contain natural metabolites, other than lactic acid, with a wide spectrum of antibacterial activity with regard to different opportunistic aerobic and anaerobic microorganisms. The relationship between the antagonistic activity and the presence of plasmid DNA in a number of the most active antagonistic lactobacilli is discussed.

[Intestinal microflora of monkeys, normal and in pathology, and its correction by preparations of live bacteria from human biocenosis (coli bacteria, bifikol and bifidum bacterin)].

The study of the qualitative and quantitative composition of intestinal microflora in 15 anthropoid apes, 60 lower primates and 72 monkeys with clinically pronounced dysbacteriosis was made, which revealed the prevalence of microorganisms belonging to the genus Proteus, a decrease in the biological activity of normal Escherichia coli and in the content of bifido- and lactobacteria. The treatment of 39 rhesus and pig-tailed macaques with the preparations of live bacteria occurring in normal human microflora led to clinical convalescence and the normalization of the bacteriocenosis, while in 33 control animals no positive shifts in clinical and bacteriological data were observed. The essential similarity of the composition of intestinal microflora in higher and lower primates and in man makes it possible to use these animals as models for testing the effectiveness of new biological preparations and determines the expediency of using bacterial therapy for the treatment of monkeys at the period of acclimatization.

[Interaction between Bifidobacterium bifidum, Proteus vulgaris, and Klebsiella pneumoniae 204 in the gastrointestinal tract of gnotobiotic chicks].

Experiments on gnotobiont chickens indicated that the strains B. bifidum 1/85 phi, P. vulgaris F-30 and K. pneumoniae 204, when introduced simultaneously into the gastrointestinal tract in a single administration, proliferate there with the pronounced predominance of bifidobacteria. 6 additional administrations of B. bifidum 1/85 phi culture resulted in the complete suppression of microorganisms belonging to the genera Rroteus and Klebsiella as early as 10 days after the introduction of bifidobacteria. These data suggest that it is necessary to use B. bifidum 1/85 phi in cases of intestinal dysbacteriosis characterized by the predominance of microorganisms belonging to the genera Proteus and Klebsiella.

[Antagonistic interrelationships of Bifidobacterium bifidum i Proteus vulgaris in vitro in the digestive tract of gnotobiotic chicks].

The antagonistic relations between Bacterium bifidum, strain I/850 phi, and Proteus vulgaris, strain F-30, were studied. These organisms, when introduced together in equal doses into the digestive tract of gnotobiotic chickens in a single administration, were shown to create certain ecological correlations in various organs with the prevalence of bifidobacteria which exerted no negative influence on Proteus vulgaris. The additional daily administration of bifidobacteria for 3 days running in doses 1000 times as great as the initial dose, the content of both dibifobacteria and Proteus vulgaris in the intestine being at that time at its maximum, resulted in the suppression of the growth of Proteus vulgaris. Our findings indicate that the influence of the pH of the medium should be considered in order to obtain the evidence of significantly pronounced antagonistic relations between the two organisms in vitro.

[Effect of bificol on the intestinal microflora of chronic colitis patients working in antiboitic prodiction].

Data on the use of bificol, a new Soviet preparation, and its effect on the intestine microflora of patients with chronic colitis occupied in production of penicillin are presented. It was shown that by the 28th day of the preparation use the level of the intestine bacteria in the patients' intestine reliably increased. The number of immobile strains decreased from 67.6 to 36.6 per cent. Bifidoflora normalized by the 14th day of the treatment. Some clinical inprovement, i.e. stool normalization, lessening of the stomach pain, increased appetite were observed by the 4th--5th day of the treatment with bificol. On the basis of the microbiological and clinical data it was shown that treatment of the patients with chronic colitis in antibiotic production should continue for at least 28 days and in individual cases for longer periods of time. It is recommended to use the preparation in 10 doses a day divided into 2 parts.
