Functional activity of hepatocytes in tissue fragments in a new bioreactor biological artificial liver.

Functional activity of hepatocytes in a new bioreactor designed for culturing of liver tissue fragments under perfusion conditions was tested. Specific hepatic functions such as ammonium detoxification, urea and protein synthesis, and P-450-dependent metabolism of p-nitroanisole were maintained for 1.5 days. The bioreactor can be used as a bioartificial liver support apparatus.

[The use of radioisotope study methods in cardiology practice].

Methods of radionuclide diagnosis modified with regard to the clinical problems and combined with new mathematic approaches to the processing of the data obtained make it possible to substantially enlarge the diagnostic information on myocardial contractility, central hemodynamics, the status and perfusion of the myocardium and to use it in combination with the latest research methods applied in cardiological patients.

[Complementary addressed alkylation of Escherichia coli 16S rRNA with 2',3'-O-[4-N-methyl-N-(2-chloroethyl)aminobenzylidene]m derivatives of oligodeoxyribonucleotides. IV. Determination of binding sites of 16S rRNA with benzylidene derivatives of d(pACCTTGTT)rA, d(pTTACGACT)rU, d(TTTGCTCCCC)rA].

By site-directed alkylation of 16S rRNA with benzylidene derivatives of d(pACCTTGTT)rA (II), d(pTTACGACT)rU (III), d(pTTTGCTCCCC)rA (IV) (reagents (II)--(IV] followed by the RNase H treatment a number of 16S rRNA fragments have been obtained. Hybridisation of these fragments with restriction fragments of plasmid pKK 3535, containing operon rrnB of E. coli rRNAs, led to the identification of all reagents' binding sites in 16S rRNA. Good correlation is found between estimated stability of non-perfect 16S rRNA.oligodeoxyribonucleotide duplexes and the level of modification of this site with alkylating derivative of the same oligodeoxyribonucleotide. With high concentration of the reagents (II)--(IV) ((2-5) x 10(-5) M) the site-directed alkylation proceeds not only at the desired site but also at other sites corresponding to non-perfect duplexes between 16S rRNA and the reagents. It should be noted that the modification mainly occurs in the non-perfect duplexes, carrying mismatched bases at the termini. Influence of the secondary structure of 16S rRNA on the site-directed modification is discussed.

[Platelet aggregation and plasma lipid spectrum in patients with myocardial infarct developing on the background of first-time appearing angina pectoris].

Patients with myocardial infarction in the presence of first angina pectoris have been found to display less platelet aggregability in the acute period of the disease than those with recurrent myocardial infarction, which is associated with a lower incidence of postinfarction complications and smaller plasma triglyceride levels. The changes observed in plasma lipid concentrations and platelet aggregation in different periods of the disease have been shown to be similar to those seen in patients with myocardial infarction concurrent with sustained angina.

[Theoretical analysis of mechanisms of spontaneous and induced mutations in DNA based on repeated sequences].

Mechanisms of spontaneous and chemically induced point mutations' emergence in DNA have theoretically been investigated using the statistical weight method. We have analysed 12 nucleotide sequences containing 95 point spontaneous mutations and 3 sequences comprising 30 mutations induced by such mutagens as 4'-hydroxymethyl-4,5',8-threemethylpsoralene, natrium bisulfite, hydroxylamine. The possibility of occurrence of point mutations by repair correction of heteroduplexes formed via mispairing of imperfect direct and inverted repeats in DNA has been studied. Statistically reliable connection of position of spontaneous mutations in DNA with repeats has been revealed for a number of nucleotide sequences. Statistically reliable connection of mutations induced by 4'-hydroxymethyl-4,5',8-threemethylpsoralene with imperfect repeats is also shown.

[The role of abnormally located chordae in the origin of accidental heart murmurs in children].

A study of 55 children with accidental systolic and diastolic heart murmurs demonstrated that abnormal chorda position (ACP), detected at echocardiography in various heart cavities, was the principal contributing factor. The relationship between the intensity and other parameters of systolic murmurs and abnormal chorda topography is discussed. Abnormal position of the chordae was not associated with any deviations from basic morphometric characteristics of the heart and should therefore be regarded as a normal variant.

[Presence in the mobile genetic elements of regions homologous to the heat shock regulatory site].

The results of contextual analysis of 13 different mobile genetical elements (MGE) MDG1, MDG2, MDG3, MDG4, HOBO, P, F, MDG 17.6, H. M. S. Beagle, CIN1, BS1, TDD1, EV1 are presented. A search for regions revealing marked and statistically non-random homology with the consensus sequence of the heat-shock regulatory site (HSRS) has been carried out in these elements. Seven MGE (MDG1, MDG4, HOBO, P, CIN1, BS1, EV1) were shown to contain the regions of non-random homology with the HSRS consensus and the real HSRS. Evolutionary significance of connection between the environment and genetical cell system, based on the transcriptional activation of mobile genetical elements by heat-shock and some other factors, has been discussed.
