[A new mathematical approach for determining antibiotic concentrations using the agar diffusion method].

A mathematical approach using the one-point intercept form of the equation of a straight line for the assay of antibiotic biological activity by the agar diffusion method is described. Insignificant alteration in the experimental conditions made it possible to markedly lower the volume of work and materials for the statistic processing. As distinct from the routine methods, the assay substance in three different concentrations is applied to three cavities on a dish and the standard substance in one control concentration is applied to the other three cavities on the same dish. The antibiotic biological activity is estimated from the equation lg cn = lg cst + (dn - dst)/bi when the relationship between the dose logarithm and the size of the diameters of the test microbe inhibition growth zones is direct. When the relationship is inverse e.g. when the dose is expressed as the dilution, the equation lg cn = lg cst + (dst - dn)/bi is used. Factor bi is determined experimentally for every sample by the mean value of the sizes of the inhibition growth zone diameter by the programs of the simple linear-regression.

[Effect of nitrogen-containing compounds on the biosynthesis of avermectins].

The influence of complex nitrogen-containing substrates (Difko yeast extract, EKD nutrient yeast extract, soy bean flour and cotton seed meal), ammonium salts and some amino acids (alanine, methionine, valine, isoleucine and threonine) on the biosynthesis of avermectins in the cultures of two mutants of Streptomyces avermitilis was studied. It was shown that an excess of the nitrogen compounds in the fermentation medium induced a decrease in the antibiotic biosynthesis and the relative content of the group B avermectins.

[Optimization of the medium for the yeast synthesis of biotin].

In order to increase the yield of biotin produced by the culture Sporobolomyces pararoseus, the medium containing sucrose, asparagine, MgSO4 (NH4)2SO4, KH2PO4, vitamin complex and trace elements was optimized. With the aid of a fractional factor experiment (2(5-1)) and a complete factor experiment (2(4)), the proportion of constituents was chosen in such a way as to double biotin yield, i.e. to increase it to 55.25 micrograms/l. An enrichment of the medium with yeast autolysate, casein hydrolysate and peptone in the presence of adenine increased biotin yield to 105.7 micrograms/l and cell productivity from 6.1 to 8.0 micrograms/l dry biomass.

[Effect of the nutrient medium composition on biotin biosynthesis by an Actinomyces species 313-152 culture].

The effect of the composition of the nutrient medium on the biosynthesis of biotin by the culture Actinomyces species 313-152 was investigated. This microorganism can synthesize biotin on simple and complex media. The biotin synthesis is stimulated by oleic acid and Tween-80 utilized by the culture as a carbon source. The exogenous biotin added at concentrations of 1 x 10(-8) M to 1 x 10(-6) M to the medium containing fodder yeast hydrolyzate and Tween-80 did not inhibit the synthesis by the culture Actinomyces species 313-152.
