Different protective effects of dimercapto-butanediol stereoisomers in experimental gastric ulceration.

The erythro version of 1, 4,-dimercapto-2, 3,-butanediol provided a significant protection against experimental gastric ulcer induced by indomethacin, while it was ineffective in stress ulcer of rats. The threo version of the same compound showed a complete ineffectiveness. It seems that most probably there is a specific receptor system in the living organism where only the erythro version is effective. Moreover, there was a remarkable difference between indomethacin-induced and stress ulcer as far as their protection is concerned. Sulfhydryl containing substances inhibited indomethacin ulcer but they were ineffective against stress ulcer. This experimental result provides further data for the difference of the pathomechanism of various experimental ulcers and thus their direct extrapolation to the human disease and to its treatment needs extreme caution.

Pentagastrin analogues containing alpha-aminooxy acids. VII. Catabolism in vivo and in vitro of a pentagastrin analogue containing N-terminal aminooxyacetyl residue.

In rat, BOC-NHO-1-14C-Ac-Trp-Met-Asp-Phe-NH2 (14C-I) is metabolised more slowly than BOC-1-14C-Gly-Trp-Met-Asp-Phe-NH2 (14C-II). Of the cytosol fractions of the rat organs the lung and pancreas exhibited a lower activity in the catabolism of BOC-NHOAc-Trp-Met-Asp-Phe-NH2 (I) than in that of BOC-Gly-Trp-Met-Asp-Phe-NH2 (II). In contrast, the cytosol fractions of the dog's lung, small intestine and pancreas hydrolysed I at a faster rate than they hydrolysed II.

[Effect of decreased blood flow in the liver on blood pressure and on the renin-angiotensin system of the rat].

The effect of reduced blood flow in the liver was studied in acute and chronic experiments on rats. 2 hours after partial constriction of the portal vein in anesthetized rats the mean blood pressure was lower and the plasma renin activity higher than in the control group. There was no relation between the two parameters. 20 weeks after partial constriction of the portal vein in unanesthetized animals the body weight, liver weight and serum albumin fraction were decreased, while the kidney weight was increased significantly and the heart weight insignificantly. The systolic blood pressure of these animals was found to be lower than that of the controls, and their blood pressure response to the i. v. administration of angiotensin II was also lower. In comparison with the control no significant differences were found in the blood renin and substrate levels. The natriuresis proved considerably lower in the animals with the partial constriction of the portal vein.

Modification of the cytoprotective effect of prostacyclin on gastric epithelial cells of rats by colchicine.

The effect of prostacyclin (PGI2) and colchicine on fundic gastric mucosa of sexually mature female Wistar rats was investigated histochemically after restraint stress. Under PGI2 treatment the ulcer index decreased significantly; this effect was moderately inhibited by colchicine. The mucus content of epithelial cells was decreased during restraint and was increased by PGI2 treatment. This protective action was also inhibited by colchicine. The nuclear volume of fundic epithelial cells increased significantly in restraint and decreased following colchicine application. These phenomena were not influenced by PGI2. According to our results the cytoprotective effect of PGI2 is not accompanied by nuclear volume changes.

Measurement of gastric mucosal blood flow in dogs by the 99mTc-4-methylaminophenazone clearance technique.

In anesthetized dogs, correlation was found between the 99mTc-4-methylaminophenazone and aminopyrine clearance measured in the same animal after administration of the gastric secretory stimulants vasopressin or glucagon. Similar correlation was observed between the 99mTc-4-methylaminophenazone and [14C]aminopyrine clearance during histamine administration. The clearance values were always corrected by the actual degree of dissociation of the radioactive molecules. The results suggest that 99mTc-4-methylaminophenazone is suitable to study circulatory changes in human gastric mucosa. Advantages of this compound include a much lower dose of radiation absorbed by the patient and his surroundings, a simplified measurement technique, and considerably reduced expenses.

On the cytoprotective action of sulfhydryl-containing substances.

Sulfhydryl substances (cysteine, glutathione, cysteamine and BAL) provide protection against experimental gastric ulcer induced by indomethacin while being ulcerogenous in stress-ulcer. Experimental data indicate the necessity to distinguish between "real" and "occasional" cytoprotective drugs.

[Dysfunction of Oddi's sphincter, a problem diagnosis].

Problems of the diagnosis of Oddi's sphincter dysfunction are discussed based on experiences gained mostly in cholecystectomized patients. In every case the existence of an organic lesion interfering with free bile flow was ruled out. The functional nature of the lesion was verified by using the morphine-choleretic test combined with either dynamic hepatobiliary scintigraphy or percutaneous transhepatic cholangiography. Illustrative cases are demonstrated.

Trophic effect of cholecystokinin-octapeptide in man - a new way in the treatment of chronic pancreatitis?

The effect of 3-week CCK-OP treatment on test meal stimulated pancreatic secretion was investigated in chronic pancreatitis patients. One drop of a 1 mg/ml CCK-OP solution applied intranasally, three times daily during 3 weeks resulted in a significant increase in volume, trypsin, lipase and amylase secretion in response to the Lundh test meal. Augmentation of trypsin secretion was the most pronounced. Functional capacity of pancreatic enzyme secretion remained elevated for 3 months after treatment. Nearly all patients became symptom free during and for some time after treatment. The results were attributed to a trophic effect of CCK-OP on human pancreas.
