[Herpes virus in cell cultures of the hematopoietic organs of Hamadrymas baboons with hemoblastosis].

Five transplantable suspension lymphblastoid cell cultures from haematopoietic organs of baboons with malignant lymphoma were obtained, designated as KMPG-1, SPG-1, 2, 3, 4. The cells of these suspension cultures contain HVP virus belonging to herpes-type according to morphological (electron-microscopic) investigation, but differing from infectious viruses of this group and closely related to Epstein-Barr virus as evidenced by its characteristics. In conformity with the preliminary data the isolated HVP virus is oncogenic for M. arctoides monkeys.

The establishment of continuous lymphoblastoid suspension cell cultures from hematopoietic organs of baboon (Papio hamadryas) with malignant lymphoma.

Two continuous suspension lymphoblastoid cell cultures designated as SPG-1 and KMPG-1 have been established from the bone marrow and spleen cells of the hamadryas baboon no. 9386 with malignant lymphoma. Virus particles morphologically characterized as herpes-like have been revealed in KMPH-1 and SPG-1 cultures in 2-10% of cells. The paper describes the establishment of these suspension cultures.

Transmission of human leukaemia to nonhuman primates.

The experiments described show that inoculation of two monkey species, M. arctoides and P. hamadryas, with human leukaemic blood or its filtrates causes a viral disease with the characteristics of malignant lymphoma of mixed type. The disease was passed in subsequent passages. The virus was isolated and identified as an oncornavirus of C-type by its characteristic morphological appearance and buoyant density (1.16 g/cm3 in sucrose and 1.21 g/cm3 in caesium chloride), and by the presence of 60-70S RNA and RNA-dependent DNA polymerase. There is some evidence that it differs immunologically from other known oncornaviruses of mammals including primates. It is postulated that both horizontal and vertical transmission of this oncornavirus can occur.

[Antibodies to herpesvirus type 8 in Kaposi's sarcoma patients and controls in Russia].

The purpose of the present study was to investigate the HH-8 seroprevalence among patients with Kaposi's sarcoma (KS), melanoma and gastric carcinoma (GC) as well as among renal recipients and blood donors. The obtained data revealed a high percentage of seropositive KS patients, which ranged from 83.6% in the classical disease type to 68.8% and to 71.4% in the immunosuppressive and AIDS-associated disease types, respectively. On the whole, the positive humoral response to HHV-8 reliably correlated with the positive findings if the viral genetic information in tumor tissue samplings. An unexpectedly high percentage of seropositive persons was found among the GC patients (41.8%) and among the renal recipients (26%), which is apparently predetermined by the immunosuppressive condition of such patients. Seroprevalence was found only in 4% of blood donors. Thus, the obtained data make it possible to conclude that KS cases, as diagnosed in Russia, are tensely associated with HHV-8 in spite of a low virus spread among the healthy population. Patients with pathology concomitant with a pronounced immunosuppression are characterized by a high prevalence of HHV-8 and belong to the category of persons with a KS risk.

[The virogenic status of cultured continuous simian lymphoid cells and their immunophenotypical characteristics].

Immunophenotyping of cultured continuous simian lymphoid cultures using a panel containing monoclonal antibodies to human B- and T-cell antigens has been carried out, by employing enzyme immunoassay and flow cytofluorometry. The test cultures showed a wide variety of cells containing B- and T-cell antigens with their varying expression. The cultures were also found to comprise lymphoblasts simultaneously containing B- and T-cell markers. Simian lymphotropic viruses, such as EBV-like and STLV-1 retrovirus, detected apart or simultaneously, have been verified by polymerase chain reaction. Whether there is a possible relationship between the type of cells in the culture and the type of their replicating virus(es) is discussed in the paper.

[Immunologic study of suspension cultures prepared from hematopoietic organ cells of baboons with lymphoma].

The cytoplasmic antigen was determined by the indirect immunofluorescence test using serum from a Burkitt lymphoma patient with a titer of antibody to Epstein-Barr virus capsid antigen of 1.320, in the cytoplasm of 2--6% of cells of suspension cultures obtained from baboons with lymphoma. There was a correlation between the number of antigen-containing cells in this cultures and the number of cells in which herpes virus was determined by electron microscope studies. Sera from baboons with lymphoma from which the suspensions cultures had been prepared contained high titers of antibody to herpes virus of baboons (HVB) and to Epstein-Barr virus (EBV). Specific differences between the antigens produced in the cultures by HVB and EBV were minimal, indicating their immunological similarity. Lymphoblastoid cultures prepared from hemopoietic organs of baboons with lymphoma belong to B cell type.

[The isolation and characteristics of continuous virus-producing lymphoid suspension cultures of African green monkey cells].

Five continuous lymphoid suspension cell cultures were established by cultivation of lymphocytes from African green monkeys. The immunological typing revealed the mixed pattern of cultures consisting of B and T cell types. Two types of viruses: herpesvirus and retrovirus C-type were demonstrated by electron microscopy. Indirect immunofluorescence and molecular hybridization studies showed one of the viruses to be an EBV-related herpesvirus of green monkeys designated HVGM and the other to belong to H (S) TLV-1 group.

[Chromosomes of 7 transplantable lines of macaque and pavian hematopoietic cells producing herpesvirus type EBV and oncornavirus type C].

Karyotypes of Su-1, Su-2, KMMA-1, and KMMA-2 (producing oncornavirus C-type), and KMPG-1, SPF-2, and SPG-3 (producing HSV, a virus of EBV type) cell lines were studied in the time course. The lines were derived from hemopoietic cells of brown macaques and baboons bearing lymphomas. Some of the lines had a paradiploid-pseudodiploid karyotype, another part aneuploid type. The selectivity of structural and numerical abnormalities of chromosomes 1, 9, 10, 11, 12, 13, 15, 18, and 20 was detected indicating a certain specificity of karyotype changes upon malignization and transformation. The number of poorly and moderately differentiating chromosomes in aneuploidy was confirmed by greater variability as compared with well-differentiating chromosomes. No parallelism between the karyotype change pattern and the type of virus produced (C or EBV) could be found thus far.

[Foamy viruses in baboons and macaques].

The occurrence of foamy viruses among baboons and macaca monkeys in Sukhumi monkey farm was investigate;. The monkeys were shown to be carriers mainly of foamy virus type II. The frequency of virus isolation increased with the age of the animals and reached 88.8-100% in adult specimens. The virus behaviour in cell cultures is described. The morphological characteristics of the virus are based on electron microscopic examinations of ultrathin sections of primary and inoculated cell cultures.

[Monkey B-lymphocyte subpopulations transformed by baboon herpes virus in vivo and in tissue cultures].

Baboon herpes virus (BHV) similarly to human Epstein-Barr virus shows tropism to B cell subpopulation. In lymphoblastoid cultures transformed in vitro by BHV no E-RFC (T cells) are found but there are B cells with surface and cytoplasmic immunoglobulins. In a number of cultures IgM could be detected only in the cell cytoplasm. E-RFC are detectable in cultures in early passages but not after 17 passages which may be due to their elimination. The time course of cell-mediated immunity values was studied in Macaca arctoides monkeys inoculated with BHV. An increase in the relative and absolute number of B cells with surface IgM and IgG in the peripheral blood was noted. Two months after inoculation the values returned to the initial level. The number of E-RFC did not change. Changes in the number of B cells may be associated with the development of immune response, or BHV influence on the process of B cell circulation in the host.

[Biological characteristics of lymphoid cell lines producing lymphotropic primate herpesviruses].

Lymphoid cell lines of monkey and human origin were characterized by proliferative activity, electrophoretic mobility, and percentage of colony-forming cells. Papio hamadryas cell lines were divided into two types similar to those of humans. One of them includes cell lines of tumor origin with high proliferative activity, decreased electrophoretic mobility, and a high percentage of colony-forming cells. The other type includes lymphoblastic lines of non-tumor origin with low proliferative activity, increased electrophoretic mobility, and low percentage of colony-forming cells.
