Reptilian viruses: adenovirus-like agent isolated from royal python (Python regius).

An adenovirus-like agent was isolated from a moribund royal python (Python regius). The DNA containing virus replicated in IgH2-cells at 30 degrees C forming eosinophilic intranuclear inclusion bodies. The virus proved to be stabile to treatment with chloroform, pH3 und pH 12 but it was labile to heat (56 degrees C). Infected IgH2 cells revealed symmetric hexagonal virus particles measuring 67-79 nm in the nucleus. The isolate shared characteristics with the viruses of the family Adenoviridae.

Fish viruses: transmission and pathogenicity of an icosahedral cytoplasmic deoxyribovirus isolated from sheatfish (Silurus glanis).

An iridovirus-like agent which was isolated from sheatfish fry showed to be of high virulence to those fish. 100% of sheatfish fry infected by water bath died within 8 days. The agent could be transmitted by infected fish causing 100% mortality in 11 days. Moribund fry showed spiralic swimming and exhibited hemorrhages in the skin. The infection studies demonstrate that the isolated virus was responsible for a sudden epizootic in 1988 with 100% mortality occurring in a warm water recirculating aquaculture unit in Northwest Germany, regularly controlled by the State Fish Epidemics Control Service of Lower Saxony and Fish Health Service.

Fish viruses: isolation of an icosahedral cytoplasmic deoxyribovirus from sheatfish (Silurus glanis).

An icosahedral cytoplasmic deoxyvirus has been isolated from moribund sheatfish (Silurus glanis) fry of a commercial warm water recirculation aquaculture unit with cumulative mortalities of up to 100%. The agent replicated in BF-2 and in FHM cells at 20-30 degrees C producing cytoplasmatic inclusion bodies followed by lysis of the cells. The DNA containing virus proved to be labile to chloroform. Infected BF-2 cells revealed hexagonal particles in the cytoplasm measuring about 125-135 nm in diameter. The virus consisted of a central electron-dense core and a electron-translucent zone. The isolate shares characteristics with the Iridoviridae.

Prevention of infectious diseases in aquaculture.

Infectious diseases remain one of the most important limitations to the successful propagation of aquatic animals. Most of the losses caused by pathogens in aquaculture could be prevented by health inspection, adequate environment and sound management practices. Effective control measures, mainly based upon 1) avoidance of pathogens 2) modification of the environment 3) improvement of host resistance 4) vaccination and 5) chemoprophylaxis are described.

[Use of fish cell cultures for toxicity determination in order to reduce and replace the fish tests].

Dispersion of liver tissue from rainbow trout at 10 and 25 degrees C resultet in cells which were cultivated in vitro at 20 degrees C regularly. Using this method a cell line (R1) has been established useful for testing the toxicity of chemical substances and waste water. The toxicity of samples influenced the morphology, growth and the living duration of the cells. These parameters gave a clear answer about the toxicity of samples testet in 1-12 h. The tests on the vitality of cells (using trypan blue) and the release of LDH did not show an agreement with the cytotoxicity in all cases. From 106 samples of waste water were 53.8% cytotoxic but only 35% showed a toxicity for fish. 93.44% of 61 toxic samples of waste water showed cytotoxicity, only 60.65% of the samples were fish-toxic. According to the results obtained it is concluded that fish cell tissue cultures are useful tools for determination of the toxicity of chemical substances and waste water. Compared with the fish test the cytotoxicity test ist more sensitive and it reduces the material, time and money needed.
