[Decolorization and isolation of recombinant hirudin expressed in the methylotrophic yeast Pichia pastoris].

Schemes for decolorization of broth and isolation of recombinant hirudin from the fermentation supernatants of recombinant yeast Pichia pastoris were developed to remove a great deal of non-desired proteins and coloring matters. Based on the stability of hirudin, heating is desirable to pre-treat the fermentation broth. Several different column chromatographies, including anion-exchange, cation-exchange, hydrokyapatite adsorption, hydrophobic interaction, gel filtration and their combination were investigated on the efficiencies of decolorization and isolation. Among these methods, the combination of cation-exchange and anion-exchange was found to be a success in both decolorization and isolation of hirudin.

[Detection of the soluble form of the Fas molecule in patients with multiple sclerosis].

We examined the levels of the soluble form of the Fas(sFas) in the cerebrospinal fluids(CSF) and the sera of patients with multiple sclerosis(MS) using an enzyme-linked immunosorbent assay (ELISA). The levels of sera and CSF sFas were significantly higher in MS patients than that in other neurological diseases (OND) group and healthy control(HC); MS patients in the active stage had more sFas molecule than that in the inactive stage. It suggests that sera and CSF sFas may be related to clinical activity in patients with MS, and that sFas may play an important role in the pathogenesis of the MS.

The effects of T cell subpopulations and recombinant interleukin (IL)-2 on peripheral B cell function in patients with myasthenia gravis.

Myasthenia gravis is an autoimmune disease mediated by antibodies to acetylcholine receptors (AChR) of skeletal muscle. The production of anti-AChR antibodies has been shown to be T cell dependent. To elucidate the mechanism(s) of anti-AChR antibody production in myasthenic patients, we studied the effects of regulatory T cells and/or IL-2 on the differentiation of AChR-primed B cells, with use of AChR stimulation for the induction of anti-AChR antibodies in vitro. Our data suggest that CD8+ T cells possess some complicated functions. CD8+ T cells could not only provide help for B cells to secrete anti-AChR antibody, but also possibly inhibit response of CD4+ T cells or kill B cells, then repress anti-AChR antibody production in MG patients. There might be some defect either in the number or function of CD8+ T cell in MG patients. Exogenous IL-2 could completely restore the suppression activity of CD8+ T cells in anti-AChR antibody production in vitro.

[Role of neural cell adhesion molecule and polysialic acid on the neuronal development and regeneration].

The formation of nervous system depends on intercellular adhesion. This review covered the role of neural cell adhesion molecule (NCAM) and its polysialic acid (PSA) moiety on the neuronal development and regeneration. Mediation of cell adhesion is the fundamental role of NCAM, while the existence of PSA on NCAM decreases cell adhesion by its specific structure. It is known that during the development of chick embryo, the expression of PSA at three critical phases determines whether motoneurons can accurately innervate muscle. Following the peripheral nerve lesions of adult rats, the expression of NCAM is regulated by the state of innervation of muscle. In the adult rat brain, the disconnection of entorhinal cortex and hippocampus results in the elevation of PSA in the outer molecular layer of the dentate gyrus, with this increased expression remains for at least 60 days after lesion. Existing data strongly suggest that the reexpression of PSA in the denervated area may promote axonal outgrowth of transplanted neurons and reconstruct synaptic connection with host.

[Fate of human fetal dopamine neurons transplanted into rhesus monkey model of Parkinson's disease: a tyrosine hydroxylase immunocytochemical study].

To predicate the value of human fetal substantia nigra transplantation in clinical treatment of Parkinson's disease (PD), dissociated cells of substantia nigra from 8-12 week old abortive human fetus were grafted into the neostriatum of 5 adult rhesus monkeys with hemiparkinsonism induced by unilateral injection of MPTP. At 2, 5 and 12 months after transplanting the monkeys were sacrificed for tyrosine hydroxylase (TH) immunocytochemistry to examine the survival and possible synaptic contact of transplanted dopamine (DA) neurons. Transplanted TH immunoreactive cells took a pattern of patches scattered in the neostriatum. Each of the cell patches consisted of 3-10 cells. The TH immunoreactive fiber network was seen in the neostriatum. Electron microscopic survey revealed that TH+ buttons arising from grafted DA neurons formed symmetric or asymmetric synapses with TH- dendritic shafts/spines, and TH+ dendrites were seen to form synapses with TH- axons of the host. Additionally, there were a few synapses formed by TH+ axonal terminals with negative buttons. The results suggest that DA neurons from 8-12 week old abortive human fetus are able to survive grafting into the neostriatum of monkey, a species phylogenetically very close to human, and to establish reciprocal synaptic connectivity with the host even at 2 months post-transplanting. It is, therefore, inferable that embryonic human DA neurons transplanted into human neostriatum may have the same fate as in monkeys.

[A study on retinal light sensitivity of normal human visual fields].

The full threshold program of QZS-2 automated perimetry made in China was used to measure the retinal light sensitivity of the central 30 degrees and peripheral 30 degrees-60 degrees visual fields of 111 eyes of 59 normal persons. Their ages ranged from 15-68 years and the average age was 38.4 years. From the corresponding respective comparisons of the results, it is shown that the mean light sensitivity of the macular, central 30 degrees and peripheral 30 degrees-60 degrees visual fields between the left and right eyes has no statistical significant differences (P > 0.05), and there are also no statistical significant differences (P > 0.05) in the mean light sensitivity of the central and peripheral fields between the male and female subjects. The mean light sensitivity declines linearly with the increase of age and eccentricity. The mean light sensitivity of the central and peripheral visual fields decreases 0.6 dB and 0.7 dB, respectively, with each one decade increase of age. The mean light sensitivity also decreases by 2.6 dB per 10 degrees eccentricity. The mean short-term fluctuation in the normal subjects is 1.39 +/- 0.56 dB.

[A randomized controlled study of the anti-cicatricial effect of preoperative subconjunctival triamcinolone on the filtering bleb].

A randomized controlled study of the anticicatricial effect of preoperative subconjunctival triamcinolone acetonide (TA) on the filtering bleb in 89 eyes (61 cases) of refractory glaucoma showed that the rates of surgical success in the TA groups were 77.3-89.0% and that in the control group 48.0%. The varying dosages of TA produced insignificant differences; however, the proportional distribution of the types of filtering blebs differed significantly among the various dosage groups, and complications occurred mostly in the high dosage group of 12 mg. The study indicated that TA raised the surgical efficacy in treating refractory glaucoma and the dosages of choice were 4-8mg.

[Traumatic lens dislocation-related glaucoma].

Among 106 patients of traumatic lens dislocation, comprising 70 males and 36 females aged 3-90 years, secondary glaucoma occurred in 93 cases (87.7%). The authors are of the opinion that lenses dislocated into the anterior chamber should be removed as soon as possible, and dislocated lens in the vitreous cavity without adhesion to the retina should also be extracted with vitrectomy. For patients with pupil block glaucoma that indicate peripheral iridectomy, the locations of operation should be carefully selected.

[Stages of primary pupil-block chronic angle-closure glaucoma].

In China, primary pupil-block chronic angle-closure glaucoma is a blinding eye disease of high incidence in primary glaucoma, with different clinical course and features from acute angle-closure glaucoma. Based on the conditions of the anterior chamber angle, the level of IOP, the defects in optic disc and visual field, and the mechanism of IOP elevation, this morbid entity was classified into 5 stages, i.e., the preclinical, the early, the progressive, the advanced and the absolute stages, and the clinical manifestations of each stage were described. This classification may be of referential use to ophthalmologists in clinical and research work.

[Comparison of optic disc damages and fluorescein filling defect of the disc between low tension and primary open angle glaucoma].

The neuroretinal rim area (NRA), the cup area and the ratio of absolute fluorescein filling defect of the disc were measured in 23 cases of low tension glaucoma (LTG) and 25 cases of primary open angle glaucoma (POAG) with fluorescein angiography and computerized image analysis. The results showed that the inferior and temporal quarters of NRA in eyes of LTG were significantly smaller than in eyes of POAG, while the ratio of absolute fluorescein filling defect of the disc in LTG was larger than in POAG, and the cup of LTG was larger than that of POAG, indicating that there might be different mechanisms of optic disc damages in the two types of glaucoma.

[The anterior chamber depth after trabeculectomy].

The anterior chamber depth was measured with the Haag-Streit Pachymeter in 26 cases (33 eyes) of primary angle-closure glaucoma preoperatively and postoperatively on days 1, 2, 3, 4, 5, 6, 8, 10, 12, 14, 30, 45 and 60. After successful trabeculectomy, the anterior chamber depth became shallower, particularly on days 2 to 4 postoperative, and then gradually deepened after day 5. By day 14 after surgery, the anterior chamber depth was recovered up to 91% of the preoperative level, and by day 30 up to 93%, to remain essentially stable thereafter.

[The neuroretinal rim area and the rim disc areal ratio in normal eyes].

A study of the neuroretinal rim area (NRA) and the rim disc areal ratio (R/D) in normal, suspected low tension glaucoma and low tension glaucoma eyes indicated that NRA had greater variation than did R/D in normal eyes, and NRA was positively correlated with the disc area whereas R/D was not. Meanwhile, R/D was more sensitive to glaucoma than was NRA or the cup and disc ratio, suggesting that R/D was useful in early diagnosis of glaucoma.

[Observations on the corneal endothelium in glaucoma].

The corneal endothelia in 87 cases of unilateral glaucoma were studied with the TOCEB-160 IV specular photomicroscope, and the healthy fellow eyes served as controls. The endothelial cell density was found decreased in majority of the morbid eyes by an average 12.7% in acute angle-closure glaucoma (34 cases), by 5.34% in chronic angle-closure glaucoma (23 cases), and by 12.4% in glaucoma cyclitis syndrome (30 cases). The average diminution of endothelial cell densities after filtering operations in 16 eyes was 10.2%, and after Nd:YAG laser iridotomy it was 5.11%. The authors are of the opinion that estimation of endothelial cells before operations and precautions against endothelial damage during operations invading the anterior chamber are necessary, especially for cases with risk factors such as trauma, uveitis, persistent high IOP, old age, or history of intra-ocular surgery.

[Preparation and evaluation of anti-human growth hormone immunoserum].

Using small dosage of human growth hormone to immunize rabbit or guinea pig, it is able to induce anti-hGH antibody formation with high titre and high affinity that could be applied to hGH RIA. In the present study five rabbits and three guinea pigs were immunized with 125-200 micrograms and 250-285 micrograms per animal of hGH respectively, followed by boosters of 10-20 or 160-250 micrograms of hGH at 2-4 week intervals for 6 or 3 months. Blood was drown 1-2 weeks before each booster for determination of antibody formation. Antibody titre and affinity were successively observed and specificity of antibody was determined for the final bleeding. It was shown that titres of immune sera from guinea pigs were much higher than those of rabbit immune sera, but vice versa for antibody affinity. This might be due to larger immunogen dose used for guinea pigs than for rabbits. Fourteen different peptide hormones were tested in reference to cross-immunoreactivity to anti-hGH antibody. It could be demonstrated that the major cross-reactive hormones are hFSH and hLH, and hTSH also reacts to rabbit anti-hGH immune sera at a lesser degree. These cross reactivities are obviously owing to the molecular homogeneities between hGH and these hormones especially of their alpha-subunits.

[Observation of oral glucose-insulin releasing test in liver cirrhosis].

In order to investigate the disorders of carbohydrate metabolism and insulin secretion as well as their correlation in patients with liver cirrhosis, we performed an oral glucose tolerance test on 30 patients with liver cirrhosis proven by history, clinical findings, liver function test, radioisotope liver scanning, ultrasonic examination, gastroscopy, barium esophagogram and liver biopsy, compared with 20 healthy controls. Blood glucose and immunoreactive insulin were determined in both groups at 60 min intervals for 180 min. Results showed marked glucose intolerance with peak value 60 min after glucose load in cirrhotic patients with normal fasting blood glucose. Plasma IRI levels were significantly higher in cirrhotic patients than in normal subjects after glucose load (P less than 0.05), especially 180 min after (P less than 0.01). Twelve of 30 cases (40%) showed an abnormal OGTT curve. Of the 12 cases seven (23.3%) showed a diabetic OGTT curve, five (16.7%) an impaired OGTT curve. While eighteen of 30 cases (60%) showed an abnormal OGIRT curve. Among the 18 cases one (3.3%) presented hypersecretic OGIRT curve, twelve (40.0%) with delayed and prolonged peak, and five (16.7%) with hypersecretic OGIRT curve and delayed, prolonged peak.
