Quantitative ultrastructural evaluation of satellite cells in soleus muscle from rats kept in hypokinesia.

The aim of the present study is a morphometric, ultrastructural evaluation of satellite cells (SC) derived from the soleus muscle (SOL) of rats exposed to conditions of hypokinesia for a period of 7 or 21 days. Qualitative and quantitative analysis of 320 electron micrographs of SC from each group was carried out. After 7 and 21 days of immobilization, profiles of the SC, in contrast to the control group, had a lower mean surface area, had a cylindrical shape, and exhibited more folded membrane. Analysis of electron micrographs of SC showed that after immobilization, a lower number of SC contained profiles of mitochondria, rough endoplasmic reticulum (RER), and Golgi. Volume fractions of RER were twofold lower after 7 days of hypokinesia and fivefold lower after 21 days compared with the control group. The SC of SOL of rats subjected to hypokinesia differed from the control group by a markedly decreased number of ribosomes and RER profiles. After 21 days of immobilization the ultrastructural characteristics of SC were typical for cells in an inactive state showing various degrees of degeneration. The results of our study presented here permit the conclusion that 21 days of hypokinesia induce a depression of SC activity, whereas subtle changes in SC appear only after 7 days of immobilization.

Morphometric analysis of satellite cells in rat skeletal muscles: soleus and extensor digitorum longus.

Satellite cells (SC) are present in all types of skeletal muscles. They can exhibit DNA replication and mitotic divisions throughout their life. Depending on the kind of muscle, a different number of SC is present. The aim of this study was to establish whether the differences in muscle metabolism are reflected in the quantitative ultrastructural evaluation of their SC. Two skeletal muscles of rats, soleus (SOL) with predominant slow fibers and extensor digitorum longus (EDL) with predominant fast ones, were studied. Morphometric analysis was performed according to the modified method of Weibel [Stereological Methods, vol 1: Practical Methods for Biological Morphometry. London, Academic Press, 1979]. In the SC of EDL mitochondria, rough endoplasmic reticulum and Golgi apparatus were less frequent as compared with the SOL. The volume fractions of these organelles were much higher in SC of SOL than in EDL. The ribosomes and polyribosomes were more densely packed in SC of SOL than in EDL. Ultrastructural morphometric evaluation demonstrated significant differences between the SC components of two different muscles: a slow (SOL) and a fast (EDL) one.

Morphometric ultrastructural evaluation of satellite cells in musculus gastrocnemius and musculus latissimus dorsi anterior of rats subjected to 7 day hypokinesia.

The investigations were undertaken to evaluate morphometrically the satellite cells (SC) of two muscles: the slow, musclus latissimus dorsi and the fast, musculus gastrocnemius from rats kept in hypokinetic conditions for seven days. The changes in both muscle were of similar character, they concerned a decrease in the Golgi apparatus, rough endoplasmic reticulum fractions and the number of ribosomes, and an increase in the relative volume of heterochromatin content. These findings seem to indicate a reduced activity of the SC. Differences were also demonstrated between the satellite cells in both examined muscles in the control group. They also suggest that the SC are not a homogeneous population as regards parameters evaluated in the present study.

Morphometric ultrastructural study of satellite cells of rats during postnatal development (1st and 30th day of life).

Ultrastructural morphometric analysis of satellite cells of the diaphragm from one and 30-day-old rats was performed. The nuclear and cytoplasmic components of the cells were evaluated. The results obtained indicate that the satellite cells of one-day-old female rats are in a state of low activity. In 30-day-old animals, on the contrary, the cells are excited and active. This may be connected with their intensive participation in the process of development of the diaphragm muscle.

Morphometric ultrastructural evaluation of secondary oocytes of mice receiving vincristine.

Secondary oocytes from mice receiving vincristine in amounts corresponding to therapeutic doses in human, in neoplasm treatment, were evaluated morphometrically and ultrastructurally. In the group of animals treated with vincristine an increase of the volume fraction of vesicular complexes and a decrease of the volume fraction of fibrous material were observed. It has also been demonstrated that, in secondary oocytes from mice receiving vincristine, the number of both multivesicular bodies and vesicles falling to one vesicular complex profile was higher than in the control group. The present results suggest that vincristine applied to females, even in therapeutic doses, may affect egg cells.

Influence of vincristine on the Golgi apparatus in preimplantation development of the mouse embryo.

The aim of our study was the evaluation of the action of vincristine (VCR) on the Golgi apparatus of mouse embryos from females receiving this drug. Quantitative and qualitative changes were investigated in 1-, 2-, 4-, and 8-cell embryos, morulae, and blastocysts from female mice treated with VCR in the amount of 0.075 mg/kg body weight 5 times once weekly. The number of embryos was decreased in all the examined developmental stages of preimplantation development. Electron microscopic investigation demonstrated translocation and dispersion of the Golgi apparatus components and changes in their relative volume as compared with that of control animals. The widest changes were noted on the 4-, 8-cell embryos and morulae in the experimental group. In the blastocyst stage, statistically significant differences in the Golgi apparatus were not demonstrated between the experimental and the control group. The present results seem to suggest the existence of remote effects of VCR which may influence the development of the progeny of females treated with this drug.

Effect of helium-oxygen mixture on myocardiac mitochondria of the rat.

Ultrastructural and stereological analysis of mitochondria of myocardiac cells from rats was performed after 2-hour breathing of helium-oxygen mixture. The following changes were observed in the cells of experimental animals as compared with the control ones: swelling of mitochondria, increase of the mean volume fraction of these structures and decrease of axial ratio of mitochondria.

Ultrastructural morphometric cell evaluation in chronic lymphocytic leukaemias.

The present study was undertaken to check whether there is a correlation between the clinical stage and the ultrastructural morphometric picture of leukaemic cells. The investigations concerned 12 cases of chronic lymphocytic leukaemia. Qualitative morphological differences could not be detected between cells originating from various clinical groups. Morphometric ultrastructural analysis, however, revealed differences in the volume fraction of cytoplasmic cell components, not only between clinical groups, but also within them. None of the clinical groups were homogeneous, but the cases classified to groups 0, I and II were uniform as regards the volume fractions of the nucleus and some cytoplasmic components. Ultrastructural morphometric studies seem to suggest that counterparts to the clinical state may be found in the morphology of leukaemic cells.

Influence of physical exercise after long-lasting hypodynamia on the morphological parameters of muscle fibers.

The purpose of this research was morphometric ultrastructure evaluation of the fibers in muscles taking part in the flight of pigeons forced to exercise after a long period of hypodynamia. It was found that following physical exercise, after 12 and 18 months of mobility limitation, there appeared marked qualitative and quantitative changes: a diminution of the volume fraction and number of mitochondria, increase of smooth sarcoplasmic reticulum and sarcoplasm, and a significant decrease of the number of glycogen granules as compared with those after 18 months hypodynamia. The above described changes were more pronounced in the supracoracoideus than in the pectoralis muscle.

Morphometric ultrastructural study of the regeneration of soleus muscle in hypokinesia.

The aim of the present study was a morphometric ultrastructural evaluation of regenerating soleus muscle fibers after denervation under conditions of hypokinesia. It was found that in rats kept under conditions of hypokinesia degenerative changes in the soleus muscle after denervation are more pronounced than in the control. On the 36th day after denervation of muscle, symptoms of regeneration of muscle fibers were observed. It was more pronounced in the control than in the experimental groups. Denervation and immobilization applied simultaneously causes more severe degeneration and atrophy of muscle fibers than does the action of only one of these factors.

Morphometric ultrastructural analysis of nuclear components in T-cell acute lymphoblastic leukaemias.

The paper deals with the morphometric ultrastructural analysis of the nuclear components performed in five cases of T-cell acute lymphoblastic leukaemia. All these cases constituted a homogeneous group in clinical and immunological evaluation. The cases analysed did not differ in cell nucleus and euchromatin volume as well as in their number of nucleoli per one cell profile.

Optical diffraction as a tool for semiautomatic, quantitative analysis of tissue specimens.

Optical diffraction was tested on electron micrographs of normal and malformed myelin sheaths as a method for semiautomatic quantitative analysis of tissue specimens. Both normal and malformed myelin sheaths were chosen for the analysis because of their characteristic internal structure and its alteration as a result of malformation. Optical diffraction patterns were obtained by means of an optical diffractometer coupled with a digital detector. The spacing and arrangement of the components of various types of myelin sheath were automatically calculated and determined and the results were verified with discriminant analysis. Out of 27 parameters of the radial and out of 25 parameters of the angular distributions of diffracted light intensity, 6 and 11, respectively, were found to have good discriminative power and were used for classification of myelin sheaths. The accuracy of automatic classification was tested by comparison with myelin sheath types of known origin. The samples visually similar by their appearance, e.g. control and regenerating myelin sheaths, were automatically classified with accuracy of 69%, whereas others were classified appropriately with 88-100% accuracy. It is believed that this kind of analysis may successfully be applied for specimens of other tissues and/or organs.

Morphometrical ultrastructural evaluation of the cytoplasmic components in acute lymphoblastic leukaemias of T-cell type.

Immunological and ultrastructural morphometric evaluation of the cytoplasmic components of cells was performed in five cases of acute lymphoblastic leukaemia of the T-cell type. All cases were a uniform group as regards clinical and immunological evaluation. On the basis of the mean cell diameter, the volume fractions and true volumes of cell organelles, cells from various cases of acute lymphoblastic leukaemia were found to be no homogeneous group. The cases examined were most homogeneous as far as the volume fraction of the Golgi apparatus and the true volumes of the cell nucleus, mitochondria, lysosomes, smooth and rough endoplasmic reticulum and Golgi apparatus were concerned.
