Inhibition of HIV-1 protease by a boron-modified polypeptide.

Six boronated tetrapeptides with the carboxy moiety of phenylalanine replaced by dihydroxyboron were synthesized, and their activities against human immunodeficiency virus 1 (HIV-1) protease subsequently investigated. The sequences of these peptides were derived from HIV-1 protease substrates, which included the C-terminal part of the scissile bond (Phe-Pro) within the gag-pol polyprotein. Enzymatic studies showed that these compounds were competitive inhibitors of HIV-1 protease with K(i) values ranging from 5 to 18 microM when experiments were performed at high enzyme concentrations (above 5 x 10(-8) M); however, at low protease concentrations inhibition was due in part to an increase of the association constants of the protease subunits. Ac-Thr-Leu-Asn-PheB inhibited HIV-1 protease with a K(i) of 5 microM, whereas the non-boronated parental compound was inactive at concentrations up to 400 microM, which indicates the significance of boronation in enzyme inhibition. The boronated tetrapeptides were inhibitory to an HIV-1 protease variant that is resistant to several HIV-1 protease inhibitors. Finally, fluorescence analysis showed that the interactions between the boronated peptide Ac-Thr-Leu-Asn-PheB and HIV-1 protease resulted in a rapid decrease of fluorescence emission at 360 nm, which suggests the formation of a compound/enzyme complex. Boronated peptides may provide useful reagents for studying protease biochemistry and yield valuable information toward the development of protease dimerization inhibitors.

Entrapment of the index flexor digitorum profundus tendon after fracture of both forearm bones in a child.

Entrapment of the index FDP tendon in a radius fracture callus occurred after fracture of both forearm bones in a 4-year-old boy. Surgical release of the FDP tendon, three months after fracture, resulted in normal index finger motion. This clinical problem can be avoided by a detailed physical examination of children with forearm fractures, verifying full passive range-of-motion of the hand after cast immobilization. Prompt supervised active range-of-motion should be done to prevent adhesions at the fracture site.

Identification of regions in HIV-1 Nef required for efficient downregulation of cell surface CD4.

Downregulation of cell surface CD4 is a characteristic property of all lentiviral Nef proteins. We have used mutational analysis to define regions within HIV-1 Nef that are critical for this biological activity. Two discontinuous regions in Nef, extending approximately from residues 96 to 144 and from residues 175 to 186, are reported to be essential for efficient CD4 downregulation. Interestingly, these sequences coincide with two conserved regions of the Nef protein that are juxtaposed to form a single surface on the known structure of Nef. A third, more amino terminal conserved region in Nef, previously reported to be important for Nef enhancement of virion infectivity, was found to be largely dispensable for CD4 downregulation. These data raise the possibility that Nef may contain two structurally distinct functional domains, only one of which contributes to the CD4 downregulation phenotype.

Reabsorption of lung liquid produced by 2,4-dinitrophenol in in vitro lungs from fetal guinea pigs.

Lungs from near-term fetal guinea pigs (62 +/- 2 days of gestation) were supported in vitro for 3 h, and lung liquid production was measured by a dye dilution technique. Twelve untreated preparations produced fluid at 1.54 +/- 0.29 mL.kg-1 body weight.h-1 during the 1st h, with no significant changes in later hours. Twelve preparations treated with 2 x 10(-4) M 2,4-dinitrophenol (DNP) showed strong reabsorptions (significant, ANOVA and regression analysis); total loss of lactate from the preparations doubled (significant, same tests). In 12 additional preparations, increasing DNP fivefold did not abolish reabsorption; results resembled those at the lower concentration. Amiloride at 10(-6) M abolished reabsorptions after 2 x 10(-4) M DNP, although fluid production still halted (n = 6; reductions significant, same tests). Amiloride alone had no effect (n = 6); untreated controls showed no change (n = 6). Similarly, 10(-4) M sodium iodoacetate virtually abolished reabsorptions after 2 x 10(-4) M DNP, although fluid production still stopped (n = 6; reductions significant, same tests). Iodoacetate alone only reduced fluid production (n = 6; significant, same tests); untreated controls showed no change (n = 6). The results suggest that reabsorptions seen after inhibition of oxidative processes depend on amiloride-sensitive Na+ channels and glycolytic metabolism.

[Long-term results of treated bi- and trifurcations in periodontitis profunda].

In 180 patients treated between 1971 and 1989 and subjected to regular follow-up checks, a total of 1111 molar teeth have been studied. 944 of them showed furcation involvement. Primarily 828 furcation involved teeth (87.7%) could be saved. The most frequently used method of treatment was flap surgery in primary treatment, and curettage in secondary treatment. Radectomy has gained ground in the past few years in the treatment of class III furcations. A treatment concept emphasizing the discriminating use of various treatment methods allows long-term preservation of molars with advanced furcation involvement.

Remote salmonellosis: surgical masquerader.

The salmonella group of bacteria consists of at least 1700 subtypes and causes widespread gastroenteric disease throughout the world, there being approximately two million cases yearly in the United States alone. An uncommon but interesting complication is the hematogenous lodgment of organisms in other sites of the body with resultant localized abscess formation, often months or years later. We present a review of 15 cases seen in our affiliated hospitals over a nine-year period. Remote salmonellosis most often occurs in patients with underlying systemic disease, such as lupus, malignancy, diabetes, and sickle cell disease, or in association with therapy that suppresses host resistance including steroids and cancer chemotherapy. The most frequent sites of involvement are bone marrow, joints, meninges, pleura, and blood vessels, or in an area of locus minoris resistentiae. The latter include scars, hematomas, fracture sites, prosthetic devices, and neoplasms. Our experience would suggest that these infections, which can remote both in location and time from the original gastrointestinal episode, are perhaps more frequent today than is generally realized. Diagnosis is often delayed because of lack of familiarity with this entity. Treatment consist of either drainage or excision of infected tissue often combined with long-term antibiotics.

Biodegradation of phenylmercuric acetate by mercury-resistant bacteria.

Selected cultures of mercury-resistant bacteria degrade the fungicide-slimicide phenylmercuric acetate. By means of a closed system incorporating a flameless atomic absorption spectrophotometer and a vapor phase chromatograph, it was demonstrated that elemental mercury vapor and benzene were products of phenylmercuric acetate degradation.

Surveillance of acute respiratory virus infections in general practice.

The ordinary postal system provides a simple method for the transportation of specimens for virus isolation to the laboratory. This was used to carry out an epidemiological survey, in general practice, of respiratory viruses in patients with acute respiratory disease. Three distinct outbreaks of myxovirus infections were recognized and most other types of respiratory viruses were isolated sporadically. After the initial "pilot" phase of the study an isolation rate of 24% was obtained over the months from October 1968 to June 1969 (20% isolations from adults' specimens and 29% from those of children).
