The cellular DNA polymerase alpha-primase is required for papillomavirus DNA replication and associates with the viral E1 helicase.

Persistent infection by papillomaviruses involves the maintenance of viral DNA as a nuclear plasmid, the replication of which requires host DNA polymerases. The role of the cellular DNA polymerase alpha-primase holoenzyme was probed by using soluble extracts from rodent cells that replicate bovine papilloma virus 1 and human papilloma virus 6b DNA in the presence of the viral E1 helicase and the E2 transcription factor. Monoclonal antibodies directed against the catalytic 180-kDa subunit of polymerase alpha inhibit DNA synthesis in this system. Addition of purified human polymerase alpha-primase holoenzyme to neutralized extracts restores their DNA synthetic activity. The amino-terminal 424 amino acids of E1 forms a specific protein complex with the p180 polymerase subunit. Immune complexes can be isolated with antibodies directed against E1 that contain a DNA polymerase activity. Moreover, this polymerase activity can be neutralized by anti-polymerase alpha antibodies. Permissivity barriers were not encountered in this in vitro system, as bovine E1 can interface with the murine and human replication apparatus. Although the large tumor antigens encoded by simian virus 40 and polyoma share limited primary sequence homology with the papillomavirus E1 proteins, the organization of functional motifs at the level of primary protein structure is remarkably similar. In addition to their origin-specific DNA-binding activity, each of these helicases may function to help recruit the cellular polymerase alpha-primase complex to the viral replication origin.

Gynecologic amyloidosis.

Amyloidosis is a generalized metabolic disorder characterized by deposition of insoluble protein in the extracellular space of various organs. We have treated a woman presenting with menorrhagia and anemia, who was subsequently diagnosed as having systemic amyloidosis. This represents the first known documentation of amyloid involvement of the female reproductive tract.

Moving a hospital. The move is the easy part.

On January 9, 1982 St. Francis Hospital in Cincinnati, Ohio, closed the doors of its old hospital and moved into a newly constructed facility to be shared with St. George Hospital, also of Cincinnati. This move was the culmination of almost ten years of planning by a Joint Long-Range Planning Committee. This article will detail the processes involved in planning the move, the move itself, and the problems encountered after the move.

Rapid assessment of fetal pulmonary maturity.

The absorbance of amniotic fluid at 653 nm has been measured in 300 specimens and found to have a significant correlation with the L/S ratio. Then 118 patients were further evaluated for neonatal outcome. RDS was absent in 99% of patients with mature absorbance values. This determination has the advantage of rapid performance without specialized equipment or training and represents a significant contribution to the assessment of fetal pulmonary maturity.

Assessment of a rapid test on amniotic fluid for estimating fetal lung maturity.

We have measured the absorbance of centrifuged amniotic fluids at 650 nm. and found reasonably good correlation between the absorbance and the L/S ratio. In 87 fluids studied, an absorbance of greater than 0.100 predicted correctly an L/S ratio of 2 or more in 98 per cent of the cases. When the absorbance was less than 0.100, 70 per cent of the fluids had an L/S ratio of less than 2.

Ready or not: assessing youths' preparedness for independent living.

This article discusses the utility of the Ansell-Casey Life Skills Assessment (ACLSA) in assessing life skills necessary for living successfully in the community upon emancipation from out-of-home care. ACLSA, completed by youths and their caregivers, identifies skills that have been mastered and those yet to be learned. Assessment information can be used for goal setting, strength identification, and relationship building, as well as to direct program planning and training in self-sufficiency services.
