cDNA cloning of a major allergen from timothy grass (Phleum pratense) pollen; characterization of the recombinant Phl pV allergen.

We isolated a cDNA encoding a major grass pollen allergen from a timothy grass (Phleum pratense) pollen expression cDNA library using allergic patients' IgE. The complete cDNA encoded an allergen that binds IgE from about 80% of grass pollen-allergic patients. Significant sequence homology was found to other major grass pollen allergens from Kentucky bluegrass (Poa pratense) as well as from rye grass (Lolium perenne) which originally were believed to form different identities. Using different monoclonal and polyclonal antibodies raised against group V allergens we identified the recombinant protein as a group V allergen from timothy grass, Phl p V. In IgE-binding studies it is demonstrated that the rPhl p V allergen can be used to block binding of patients' IgE to natural group V isoallergens on two-dimensional immunoblots. IgE inhibition experiments show that up to 60% of grass pollenspecific IgE can be preadsorbed with the rPhl p V allergen from patients sera. The purified rPhl p V induced specific histamine release of blood basophils from grass pollen-allergic patients. This emphasizes the usefulness of the rPhl p V for diagnostic and therapeutic purposes and corroborates the view that specific diagnosis and therapy of type l allergy could be performed with a limited panel of relevant recombinant allergens.

Demonstration of membrane-associated phospholipase A2 in cultivated heart muscle cells by immunogold-technique in surface replicas.

Recently we produced mAb's to phospholipase A2 (PIA2) from bee venom for different purposes (allergen-identification and -standardization). Obviously one of these monoclonal antibodies "5D5" binds to a conserved (cross reactive) epitope of membrane PIA2 of intact cells. In this study we demonstrate binding of 5D5 to membrane associated phospholipase A2 of rat heart muscle cells by means of two-step immunogold technique in combination with replica technique.

Monoclonal antibody immunoassay for quantitative analysis of group V allergens in grass pollen extracts.

A two-site monoclonal antibody (MoAb) ELISA has been developed for the quantification of the Phleum pratense major allergen, Phl p V. The assay is based on two MoAbs which recognize different non-overlapping epitopes on the Phl p V molecule; one antibody (1D11) was immobilized on the solid phase and the other (3B2) was biotinylated. An affinity-purified Phl p V preparation (purity of 95%) was used as standard. The assay has a sensitivity of 10 ng/ml of allergen and is suitable for the detection of group V allergen in aqueous grass extracts. The specificity of the assay was investigated with 14 grass pollen and five non-grass pollen extracts. Different levels of group V allergen were detected in extracts of grasses, but not in non-grasses. The assay gives a good correspondence with allergenic activity of extracts as determined by ELISA inhibition using serum pool of allergic patients. The results indicate that the two-site MoAb ELISA could be very useful in the standardization of allergenic extracts from grass pollen.

Detection of crossreactive determinants in grass pollen extracts using monoclonal antibodies against group IV and group V allergens.

Monoclonal antibodies (MoAb) to defined allergens of P. pratense were raised. Five of them were selected for detailed studies by means of immunoblotting after SDS-PAGE and IEF of extracts from P. pratense and L. perenne. Three antibodies (1D11, 3B2, 2D1) recognize structures with mol. wt of 29 and 34 kD and pI of 4.8-7.6, corresponding to group V allergens. Two other MoAbs (2D8, 3C4) are directed against strong basic structures with a mol. wt of 50 kD and pI of 7.8-9.5 according to group IV allergens. The specificity of antibodies was supported by direct ELISA with purified group V and IV allergens. The isolated allergens were characterized before by SDS-PAGE and CIE and that allergenicity was detected with sera of patients with allergic rhinitis. Using our selected MoAbs crossreactive epitopes on group V and IV allergens have been excluded. Our antibodies have been used to detect crossreactivity in 14 grass pollen extracts. The evaluation of the pollen extracts has been performed by enzyme immunoassay (EIA) inhibition. One MoAb (3C4) is able to recognize group IV allergens in all grass species analysed whereas the MoAb 2D8 seems to identify group IV structures in selected grasses only. Binding to conserved structures of group V has been proved for MoAb 1D11. Other group V specific MoAbs (2D1, 3B2) identify similar, however incomplete, spectra. These results have been confirmed also by the dot immunobinding assay.

Characterization of murine monoclonal antibodies to phospholipase A2 and mellitin from bee venom.

9 murine monoclonal antibodies (MoAbs), 7 to phospholipase A2 (Pla2; Api m I) and two to mellitin (Api m III) have been produced. Specificities were demonstrated using immunoblotting and enzyme immunoassays (ELISA). Antibodies specific for Pla2 were characterized in detail. Epitope mapping of Pla2-specific MoAbs identified three independent binding regions. 5 selected MoAbs bound to one of these sites compete with specific human IgE. Two of MoAbs characterized are suitable tools to quantify Pla2 in a two-site binding assay.

Correlation of ventricular size and head circumference after severe intra-periventricular haemorrhage in preterm infants.

No detailed information is available about the timing and correlation of objectively gauged ventricular width (VW) and the rate of growth of the occipitofrontal circumference in premature infants with intraventricular haemorrhage preceding progressive ventricular dilation. For this study, two groups were selected according to clinical course: group A (n = 6) had ventriculomegaly with no signs of raised intracranial pressure, while patients in group B (n = 7) developed progressive ventricular dilation after a period of latency. A VW between 0.9 and 1.4 cm (group A) did not affect normal head growth, whereas a VW greater than 1.5 cm was always associated with the development of hydrocephalus. Our data further suggest that the widely used criterion of a head growth rate of more than 2 cm/week is a relatively poor criterion for the definition of post-haemorrhagic hydrocephalus.

[Infrared casting temperature control of the induction centrifuge casting exemplary in the non-precious dental alloy Wiron 88].

A temperature-dependent, IR-controlled casting technology for the non-precious dental alloy Wiron 88 (Bego) is presented and compared with a traditional procedure. The new casting allows the production of cast objects with favourable and reproducible material properties. Compared with the commonly used technology hardness, grain size and susceptibility to corrosion could be reduced. Texture after casting was found to be homogeneous. The procedure presented meets the requirements for a modern casting technology and be adapted to older automatic casating devices.

[Production and characterization of a high affinity monoclonal antibody with digoxin and digitoxin specificity].

A monoclonal antibody with a high affinity for digoxin (KA = 5.5 x 10(10) M-1) and digitoxin KA = 5.0 x 10(10) M-1) was produced by somatic cell fusion. This antibody, designated 2A3, displayed little cross reactivity with other glucosides and no cross reactivity with endogenous steroids. It was shown that 2A3 is a suitable tool in an enzyme immunoassay for digoxin and digitoxin.

[The microstructure and corrosive properties of the nonferrous casting alloys Gisadent NCA, NCS 1 and KCM 83 before and after casting].

Nonprecious alloys (NEM) applied in dental practice often show widely differing material properties after casting. This is caused by subjective and objective differences in manufacturing of these alloys with a given technology as well as by varying composition and structure of the alloys within and in between different lots. For homogeneous composition of crude specimens technologically caused variance of casting condition itself leads to considerable differentiation of casting results. If the original state of crude alloy forms is very different then there will be a levelling out and/or increase of scattering of material parameters after casting. Optimized computed-controlled casting technology in combination with defined muffle ring size could result in reproducible casting results.

[Persistent neonatal hypoglycemia in nesidioblastosis of the pancreas].

A now 10 month old female infant suffered from persistent non ketotic neonatal hypoglycemia despite continuous intravenous application of glucose (greater than 10 mg/kg/min). There was only a transient response of blood glucose after intravenous administration of glucagon and prednisolon. Biochemical findings indicated hyperinsulinismus (insulin level of 26 mE/ml during hypoglycemia). Oral diazoxid treatment in high doses (22 mg/kg) stopped hypoglycemia episodes for several days but the newborn remained glucose infusion depended. Finally the treatment had to be interrupted because of vomiting. At the age of 4 1/2 weeks a subtotal pancreatectomy was performed. The histological examination of the pancreas confirmed the clinically suspected diagnosis of nesidioblastosis. After pancreatectomy the infant required insulin therapy. Since six months the girl is without insulin in a good condition. Despite periods of arrested head growth before pancreatectomy the psychomotoric development is normal.

Isolation of timothy (Phleum pratense) allergens using affinity chromatography with monoclonal antibodies.

The monoclonal antibody 1 D11, directed against the timothy pollen allergens I, II, III, and a further, anodically migrating component, is suitable for the isolation of these allergens by means of affinity chromatography. The purified allergens consist of glycoproteins with many different isoelectric points. Their molecular weights were estimated as approximately the same as those of the allergens in the whole timothy pollen extract. Their allergenic activity (IgE binding) was not lost during the purification steps.

[Quantitative determination of LP-X in the differential diagnosis and treatment of direct hyperbilirubinemia in infancy].

Quantitative measurements of serum concentrations of LP-X were performed in 45 newborn and infants. The changes of LP-X concentrations before and after a 2-3 weeks' course of cholestyramine therapy differentiated extrahepatic biliary atresia (n = 6) from other causes of neonatal liver disease with a sensitivity of 100%, a specificity of 78.6 to 84.6% and an efficiency of 81.3 to 86.7%. The efficiency was decreased in children with alcoholic stools (75-80%). Cholestyramine treatment of 3-7 days did not allow to diagnose all children with extrahepatic biliary atresia. However, the test was superior to a combination of single measurements of LP-X and GGT. The changes of LP-X concentrations in serum were influenced by the individual course of the disease but not by the synthetic function of the liver (as indicated by CHE activities) or by parenteral nutrition or bacterial infections. LP-X was a valuable parameter in the management of cholestyramine therapy in infants with liver diseases.

[Perinatal risk factors and intraventricular hemorrhage in newborn infants of birth weight less than or equal to 1,500 g].

In a retrospective study in 113 preterm newborns (birth weight less than or equal to 1500 g, gestational age less than 35 weeks) intraventricular hemorrhage (IVH I-IV) determined by serial sonographic examinations was correlated to a number of maternal and neonatal risk factors. While there was no significant correlation to bleeding during the gestation, premature rupture of the membranes, mode of delivery, and age of the mother, incidence of IVH was significantly lower in newborns born to mothers with EPH-gestoses (p less than 0.05). A significant high incidence of IVH was found in very premature newborns (p less than 0.025) and in newborns with hyaline membrane disease (p less than 0.005). No single risk factor for a high incidence of IVH was found. IVH in very low birth weight infants probably is caused by summation of several different perinatal risk factors.
