Polysaccharide expression in Lactococci.

Lactococcus lactis produces polysaccharide under defined environmental conditions. Three approaches are being used to identify regulators of polysaccharide synthesis in the organism. i) Two new lactococcal vectors, each of which contains a promoterless reporter gene, have been developed. They are being used to select for chromosomal insertions that affect expression of polysaccharide ii) Genetic complementation with lactococcal genomic libraries identified two classes of lactococcal genomic activities, both of which affect capsule expression in Escherichia coli iii) Lon, a highly conserved protease, is a negative regulator of polysaccharide synthesis in E. coli. Antiserum specific to Lon protease reacted with a lactococcal protein similar in size to E. coli Lon. This reaction was strongest in lactococcal strains which could not be induced to express polysaccharide.

[Diagnosis and prevalence of latent hepatic encephalopathy].

To compare the efficacy of various psychometric and neurophysiological tests in the detection of latent hepatic encephalopathy (LHE) cerebral functions were studied in 146 patients with liver cirrhosis but without overt encephalopathy and in 146 matched controls. Patients with liver cirrhosis scored significantly worse than controls in 8 out of 11 tests. Best discrimination between patients with cirrhosis and controls was obtained by testing for reaction time to white and colored light with a reaction time apparatus (DTG), and with the digit symbol (UT1) and block design test (UT4), i.e. with two Wechsler adult intelligence scale performance tests. Thirtyseven out of 146 (25%) patients with cirrhosis reveiled an abnormal result with the DTG alone. A combination of the DTG, UT1 and UT4 yielded the diagnosis in 44 (30%) patients. LHE correlated with the severity of the disease (Child-Pugh classification) but not with its etiology or with portasystemic shunting. In the Federal Republic of Germany about 300,000 subjects suffer from liver cirrhosis. Based on our results 100,000 of them may have LHE.

Carrier facilitated diffusion.

The concept of a mobile carrier combining reversibly with a substrate is considered as a possible mechanism for facilitated transport across biological membranes. The mathematical model is a system of three reaction diffusion equations with certain boundary conditions. Two limiting cases are discussed in detail: The case of a "thin" membrane where the diffusion of bound and unbound carrier from one surface to the other may be simulated by a single jump. If the diffusion rate of the substance to be transported is small, then an approximate stationary solution is derived using singular perturbation theory. Finally, the results of numerical simulations are presented for a wide range of parameters.

Biological Transport

Net albumin transport across the wall of the rabbit common carotid artery perfused in situ.

We have studied the transport of radioactively labelled albumin in the rabbit common carotid artery perfused in situ at 100 cm H2O luminal pressure in the anaesthetized living animal, assessing the distribution of concentration across the wall by means of sequential frozen sectioning. We have compared the findings with those of experiments in which we have attempted to saturate the wall with label. Our findings support the belief that there is a net transport of macromolecules across the arterial wall. They show in addition that the wall is inhomogeneous. The distribution volume for label is greater in the adventitia than the media, which appears to offer a larger resistance. The transport process is seemingly dominantly diffusional.

[Acute Hedolite poisoning in humans].

Severe intoxication was contracted by an agrochemist who had handled Dintro-O-kresol (DNOC) over several days. The symptoms are described in this paper. The half-life calculated from the blood level course amounte to 148 hours. The particular case of intoxication is evaluated, and reference is made to literature, before the reader's attention is drawn to certain major and crucial aspects that should be duly considered in any use of DNOC, such as limitation of exposure, blood level monitoring, and availability of adequate safety clothing.
