[Pharmacologic influencing of fetal phospholipide synthesis. Part II: Carnitin - a new way of dyspnea syndrome prophylaxis? (author's transl)].

Carnitine is a body-owned Betaine which is wide-spread in living nature in the form of a physiologic metabolit. On account of its central metabolic function, Carnitine has various effects in the intermediary metabolism. Toxicity of Carnitine is very low. DL-Carnitine hydrochloride (30 mg/kg body weight) was injected into 25 gravid Wistar rats and 24 gravid rabbits, 4 days before the Cesarean section. Compared to the group of controls, the group treated with Bromhexine metabolit VIII, as well as to the group treated with Betamethason, significantly higher values with regard to the content of total phospholipids, lecithine and palmitic acid could be found in the fetal rat lungs. Effects of Carnitine on the phospholipid metabolism are discussed.

[The combination of etomidate and fentanyl as a short acting and introduction anaesthesia in the dog in experimental medicine (author's transl)].

The effects of a combination of etomidate (1.0 mg/kg bw) and fetanyl (0.005 mg/kh bw) on haematology, blood gases, acid-base balance and haemodynamics were investigated in 8 mongrel dogs. The only significant change in haematology was leucopenia. Blood gas values and acid-base balance initially showed a depression of respiration and a tendency to slight metabolic acid dosis. Decreased heart rate caused diminution of cardiac output, while systemic and pulmonary resistance as well as stroke volume were not altered significantly.

[An experimental study on the pathogenesis of the acute pulmonary insufficiency (author's transl)].

Parameters of the lungs as Compliance PaCO2, PaO2, have been measured in connection with experimental, pathophysiological insults like hematoma, bone marrow intravasation and hemorrhage, in rabbits. Concerning the compliance quotient, a statistically proved decrease was found within the groups, which have had a hematoma. Moreover, the histological picture of the lungs from rabbits with a hematom showed perivascular and intersitial edema and considerable atelectases. Serum of animals, which have had a hematom for at least 24 h has been fractionated by gel chromatography; low molecular weight substances of the fractionated serum were injected into healthy recipients. Thereafter, measurement of the centralvenous pressure suggests the appearance of substances, pharmacodynamic similar to 5-Hydroxytryptamin, in blood of rabbits with hematom.

[Renal blood flow investigations with 133xenon and the Anger scintillation camera in the hyperacute xenograft rejection of the rabbit kidney (author's transl)].

The aim of this study was to demonstrate the advantage and validity of 133Xe-washout externally monitored by the scintillation camera. Until now there were no reports on quantitative blood flow studies in Hyperacute rejection of transplanted kidneys using a scintillation camera. Within 35 minutes after e-vivo hemoperfusion of rabbit kidneys by cats we found a simultaneous progressive decrease of renal blood flow, renal cortical blood flow as well as of the intrarenal distribution of renal cortical blood flow in all cases. The hyperacute rejection of xenografts could be verified in every case histologically. Using the scintillation camera we were able to detect regional perfusion defects caused by artifical air embolism as well as by preexisting cortical infarction.

[Acute hepatic coma. Experimental study on the predictive value of clinical-chemical findings for the prognosis of acute hepatic coma].

In order to determine the validity of clinical-chemical parameters for the prognosis of hepatic failure, 28 pigs were subjected to liver ischemia for 40--160 minutes duration. The following parameters were studied: GOT, GPT, gamma-GT, LAP, LDH, GlDH, AP and isoenzymes, total bilirubin, potassium, sodium and chloride. In a statistical comparison in the surviving animals, an unexplainable increase in GlDH activity was observed. In the other clinical-chemical parameters none was seen to be of use for the prognosis for either life or death in acute hepatic failure.

[The effect of proteinase inhibitors in the early stage after experimental myocardial infarction (author's transl)].

Our experiments have shown that in cats in the early phase after experimental myocardial infarction, the volume of extravascular fluid increases in the lungs while the intravascular volume decreases. A hemoconcentration developed with increased hematocrit and a fall in plasma volume. The thrombocyte count fell continuously during the first 4 hours after the infarction. The CPK activity in the region of the myocardial infarct was reduced in comparison to uninfarcted tissues. Intravenous injection of 50,000 KIU/kg Trasylol within 20 minutes after the infaction caused a significant reduction of the extravascular fluid in the lungs. All the other parameters were not changed by Trasylol administration, with the possible exception of the fall in thrombocytes which seemed to be slowed after Trasylol.

Experimental myocardial infarction in the cat. I. Reversible decline in contractility of noninfarcted muscle.

The contractile state of the noninfarcted myocardium was examined in adult cats after myocardial infarction produced by ligation of several branches of the left coronary artery. At 2 days, 7 days, and 6 weeks after infarction, and after determination of intracardiac pressures, papillary muscles were exicised from the noninfarcted segment of the right ventricle and attached to a myograph for analysis of contractile function. One week after infarction there was a decline in actively developed force at Lmax, caused by a decrease in the rate of force development. In addition, the response to procedures that augment myocardial contractility, such as paired stimulation and increasing the frequency of electrical stimulation, was significantly depressed. Two days after infarction, changes were less significant, although similar in direction. Six weeks after infarction, developed force at Lmax had returned to normal values. The response to procedures augmenting contractility also had returned to normal. There appears to be a distinct, reversible loss of contractility in the remaining viable myocardium in the early phase after experimental infarction.

[Contractility of the surviving myocardium following experimental infarct].

The contractile function of the non-infarcted myocardium was examined in adult cats following myocardial infarction produced by ligation of several branches of the LAD and circumflex coronary artery. Two and seven days following infarction, after determination of intracardiac pressures, papillary muscles were excised from the non-infarcted portion of the right ventricle and transferred into a myograph for analysis of contractile function. One week following infarction, force-velocity relations of the surviving myocardium showed a shift downward and to the left in comparison to sham-operated controls. At the same time, there was a decline in actively developed force at Lmax to be deserved, which appeared to be caused by a decrease in the rate of force development. Two days following infarction, similar changes were to be observed, although to a lesser extent. There appears to be a distinct loss of contractility in the remaining viable myocardium following experimental infarction.
