Genetic analysis of the gene for N-acetylglucosaminidase in Dictyostelium discoideum.

Three independent mutations affecting N-acetylglucosaminidase in Dictyostelium discoideum were mapped by the parasexual system and found to lie on linkage group IV. These mutations as well as two others were found to be recessive and noncomplementing in heterozygous diploids. Thus they all appear to affect the nagA locus. Since two of the mutations give rise to thermolabile enzyme, this defines the structural gene for N-acetylglucosaminidase. The enzyme is a homodimer of a 68,000 dalton subunit and thus would be expected to be determined by a single locus. The expression of this gene is regulated by the stages of development; however, it should be mentioned that none of the mutations fell in a separate locus that might determine a specific positive regulatory proteins.

The effect of divalent cations on aggregation of Dictyostelium discoideum.

Following the initiation of development, amoebae of Dictyostelium discoideum aggregate chemotactically toward cyclic AMP (cAMP). Adenyl cyclase, cAMP phosphodiesterase, and cAMP binding sites all increase 20--40 fold during the first few hours of development. It has been shown that addition of 1 mM EDTA and 5 mM MgCl2 accelerates the aggregation process. Likewise, the calcium ionophore, A23187, leads to precocious aggregation while 4 X 10(-5) M progesterone considerably delays it. These treatments have now been shown to result in increased accumulation of adenyl cyclase in the case of EDTA and Mg2+ or the ionophore and greatly decreased accumulation in the case of the steroid. Treatment with EDTA and Mg2+ or the ionophore has been shown not only to accelerate aggregation in wild-type amoebae but to overcome complete blocks to aggregation in certain mutant strains. We have found that addition of Mn2+ will also permit aggregation of mutant cells otherwise unable to aggregate. This divalent ion, unlike EDTA and Mg2+ or the ionophore, was shown to directly stimulate adenyl cyclase. Calcium ions were also found to affect the enzyme such that at Ca2+ concentrations found within the cells the great majority of the activity is inhibited. Manganese ions can overcome the inhibition by Ca2+. These findings show that conditions which stimulate aggregation result in increased activity of adenyl cyclase either by increased accumulation of the enzyme or by increased activity of the available enzyme, and support the proposed central role of adenyl cyclase in aggregation.

Isolation and characterization of a component of the surface sheath of Dictyostelium discoideum.

Aggregates of Dictyostelium discoideum are surrounded by a surface sheath which functions to maintain polarity and integrity during development. We have isolated and partially characterized a component of the surface sheath. It is composed of 60% cellulose, 15% protein, 3% heteropolysaccharide (heteropolymer), 5% lipid, and 1% sulfate when isolated from migrating slugs. The sheath, isolated from aggregates prior to tip formation, has less protein, a different heteropolymer, and cellulose of a lower crystallinity than the sheath of migrating slugs. The increase in crystallinity of the cellulose during development may be important in determining the strength of the surface sheath.

Kinetic analysis of biochemical differentiation in Dictyostelium discoideum.

The metabolic network leading to accumulation of cellulose, trehalose, and mucopolysaccharide during development of Dictyostelium discoideum was simulated on a computer. The program consists of a metabolic map, the measured specific activity of the enzymes involved at each stage in development, and the substrate and inhibitor affinities. The Km values of four enzymes, amylase, UDP-galactose polysaccharide transferase, UDP-galactose epimerase, and cellulose synthetase, were determined for this study. At each iteration (1 min) during the period simulated (1500 min), the in vivo activity was calculated for each enzyme using Michaelis-Menten equations and new values for metabolites and end products were generated. The computed values for the concentration of both metabolites and polysaccharides were in close agreement with the measured values at all stages of development. We conclude that the in vitro measured values correlate well with the measured in vivo rates when treated in this manner. The program was modified to simulate the alterations in carbohydrate metabolism which might be expected in mutant strains with reduced activity of various enzymes. Trehalose was found to overaccumulate when either the peak value of the developmentally controlled increase in the specific activity of UDPGlc pyrophosphorylase was reduced. Trehalose accumulation was decreased in simulations of mutants lacking glycogen phosphorylase or glycogen synthetase. The interaction of these metabolic pathways is discussed.

Structure and function of beta-blucosidases in Dictyostelium discoideum.

There are two isozymes of beta-glucosidase in developing cells of Dictyostelium discoideum. A procedure for screening large numbers of clones for beta-glucosidase activity was utilized to obtain mutations which directly affect the activity. We recovered seven strains which lack both isozymes and four strains with residual activity in which enzymatic and physical properties of both isozymes are altered. Beta-Glucosidase appears to act as a block to selfing in macrocyst formation as shown by the fact that ssite mating type to form macrocyst-like structures. Immunological evidence utilizing antisera prepared against purified beta-glucosidase-1 demonstrates that most of the glycosidases in Dictyostelium discoideum share a common antigenic determinant which appears to be added post-translationally. The two isozymes of beta-glucosidase share common protein subunits but the antigenic determinant is either lacking or masked in beta-glucosidase-2. This may account for some of the enzymatic and physical differences between the two isozymes.

Quantitation of the spatial distribution of 'prespore vacuoles' in pseudoplasmodia of Dictyostelium discoideum.

The axial distribution of an organelle, the prespore vacuole (PV), previously reported absent from the prestalk region, was determined in pseudoplasmodia of varying sizes, under differing conditions of photostimulation of migration. The distribution of these organelles, determined quantitatively by electron microscopy of sections from known axial locations, was found to have a spatial pattern which varied with pseudoplasmodial size. The total complement of these organelles appeared constant for any size of pseudoplasmodium under similar conditions of illumination. Increased illumination decreased the total number of the organelles. The spatial distribution of PV varies with total cell number, and the size of the region with no PV bears no relationship to the proportion of the cell mass which would form stalk cells. Similarly, the number of cells containing PV bears no fixed relationship to the number of cells which will form spores. On these grounds, the reported role of PV, that of directing or reflecting spore differentiation, appears unlikely.
