Simultaneous high-pressure liquid chromatographic determination of niacin and niacinamide in multivitamin preparations: reversed-phase, ion-pairing approach.

A high-pressure liquid chromatographic procedure for the simultaneous determination of niacin and niacinamide in multivitamin preparations was developed and evaluated. Reversed-phase liquid chromatography with dioctyl sodium sulfosuccinate in the mobile phase is utilized for the analysis. The accuracy and precision of the procedure were compared with those of the currently official procedure.

Synthesis and structure-activity relationships of selected isomeric oxime O-ethers as anticholinergic agents.

A series of isomeric (Z)-and (E)-oxime O-beta-dimethylaminoethyl ether methylhalide derivatives was synthesized, and their (Z)-and (E)-assignments were made on the basis of chemical and spectral data. The respective (Z)-and (E)-isomers were evaluated as anticholinergic agents on the rat ileum. The antimuscarinic potencies of the respective (Z)-and (E)-isomers were compared to determine the effect upon potency of this type of geometric isomerism. Three general structure-activity relationships and discernible among the synthesized compounds: (a) among oxime O-ethers derived from aromatic aldehydes, the higher potency consistently resides in the isomer where the aryl substituent is (E) to the ammonium ether substituent; (B) among oxime O-ethers derived from diaryl ketones, the (Z)-and (E)-isomers are approximately equipotent; and (c) oxime O-ethers derived from diaryl ketones are the most potent of the synthesized compounds.

Simultaneous analysis of hydrocortisone and hydrocortisone phosphate by high-pressure liquid chromatography: reversed-phase, ion-pairing approach.

The reversed-phase, ion-pairing approach to high-pressure liquid chromatography was applied to the simultaneous analysis of hydrocortisone and its phosphate ester in laboratory-prepared samples and injectable solutions. Results of this technique were evaluated and compared with results of the official procedure.

Development and evaluation of method for simultaneous determination of phenobarbital and diphenylhydantoin in plasma by high-pressure liquid chromatography.

A procedure for the simultaneous analysis of diphenylhydantoin and phenobarbital in plasma by high-pressure liquid chromatography was developed and evaluated. Separation is effected on a porous particle silicic acid column with chloroform dioxane-isopropanol-acetic acid (310:9.7:1.0:0.1 by volume) and is monitored at 254 nm. Results of the procedure were compared with results of a GLC assay.

Characterization of hydrogen bonding between selected barbiturates and polyethylene glycol 4000 by IR spectral analysis.

Several barbiturates and primidone were equilibrated with polyethylene glycol 4000 in pyridine. IR spectral properties of these samples indicate that seven disubstituted barbiturates complex with polyethylene glycol 4000 while five disubstituted barbiturates and two trisubstituted barbiturates as well as primidone do not. Forces responsible for complexation of barbiturates with polyethylene glycol 4000, as inferred from spectral data, consist of hydrogen bonds formed between N1 and N3 hydrogens of the barbiturate ring and two oxygen atoms of the --O--CH2CH2--O--moiety. Also, there appear to be three configurations of intermolecular hydrogen bonding sites between disubstituted barbiturates. Several factors affect the barbiturate-polyethylene glycol 4000 interaction, including the nature of the solvent, C5 substituents, the number of hydrogen bonds formed between reactants, and the 2-carbonyl group of the barbiturate ring. Complexes of polyethylene glycol 4000 with phenobarbital, butabarbital, and cyclobarbital are stable in water at 26 degrees or below, but complexes of polyethylene glycol 4000 with butethal, cyclopentenyl allylbarbituric acid, pentobarbital, and probarbital are not.

Quantitative extraction of tubocurarine, gallamine, and decamethonium from biological materials.

A procedure is described for quantitative extraction of tubocurarine, gallamine, and decamethonium from blood, urine, and tissue. Ion pairs of the drugs are formed with picric acid and are extracted into a dichloromethane/1-pentanol solvent. Qualitative analysis of the extract is effected using thin-layer chromatography. Quantitative analysis is accomplished by elution of TLC areas containing the drug, formation of the ion pair with sodium 9,10-dimethoxyanthracene-2-sulfonate, and analysis of the ion pair by spectrophotofluorometry.
