Adaptive response to pneumonectomy in puppies.

When left pneumonectomy was performed on 9-week-old puppies, the right lung increased in weight, volume, surface area, and number of alveoli so that at age 20 weeks these variables were the same as those of both lungs of control animals and significantly larger than those of the right lung of control animals. The adaptive response of the right lung after pneumonectomy was greater in the lower lobe than in the middle or cardiac lobes. The number of alveoli per ml and the average interalveolar wall distance, hence the surface to volume ratio, was the same in both lungs of control animals and the same in the cardiac, middle, and lower lobes of their right lungs.

Static lung function in puppies after pneumonectomy.

We performed left pneumonectomy on eight puppies at 9 wk of age; seven littermates underwent a sham operation and functioned as controls. All were studied at 20 wk of age, at which time the remaining lung in the animals with pneumonectomy had the same volume, weight, and surface areas as both lungs of the controls. Total lung capacity (TLC) was the same in controls and postpneumonectomy dogs, but in the latter, the ratio of functional residual capacity to TLC (FRC/TLC) and that of residual volume to TLC (RV/TLC) were larger. Lung pressure-volume curves also differed, postpneumonectomy dogs demonstrating slightly greater recoil at lung volumes below TLC. Therefore, the increased FRC and RV of postpneumonectomy animals could not be ascribed to changes in lung recoil and must have been caused by increased chest wall recoil perhaps related to differences in shape between lung and chest wall. Studies of regional lung function revealed reduced aerated lung in the left hemithorax of the postpneumonectomy dogs and increased FRC/TLC and RV/TLC at the left base. The abnormal distribution of regional volumes did not account entirely for the increase in overall FRC and RV. Comparison of excised lung pressure-volume curves in those measured in intact animals revealed better agreement in postpneumonectomy dogs than in controls.

Serum from pneumonectomized rabbits stimulates alveolar type II cell proliferation in vitro.

Serum samples collected from pneumonectomized rabbits 9 and 21 days after surgery significantly enhanced the incorporation of [3H]thymidine into DNA by alveolar Type II cells in vitro, but they had no effect on lung or skin fibroblasts. Activity was absent from serums collected 140 days after surgery. Active serums showed a dose-response effect at concentrations between 1% and 10% in the alveolar Type II cell culture system and the activity was heat stable. The active factor eluted from columns of Sephadex G75 at the void volume at neutral pH, but after acid-ethanol extraction and chromatography under acidic conditions, it eluted with a Kd of approximately 0.65. We propose that the active substance is a somatomedin or somatomedin-like peptide.
