Altered protein patterns in brains of children with neuronal ceroid lipofuscinosis.

The neuronal ceroid lipofuscinoses (NCL) are a group of inherited neurodegenerative diseases characterized by massive intralysosomal accumulation of storage materials. We have studied the protein patterns in 5 NCL, 5 control, and one Alzheimer disease brains. When protein patterns in NCL and control brain gray matter homogenates were examined by SDS-PAGE, NCL brains showed an absence or greatly reduced amounts of the Mr 160-180 kDa component and reduced amounts of the Mr 29-36 kDa component. Concomitantly, an increase in several components with Mrs of 45-50 kDa was noted. The 180 kDa polypeptide appears to be a glycoprotein because it was bound to the lectins concanavalin A and Ulex europaeus. Recently, the abnormal processing of amyloid protein precursor (APP) and its potential role in NCL have been suggested. Possible defects in tissue proteases and protease inhibitors may be considered responsible for the presence of these amyloid beta protein precursor fragments. To examine this possibility we are using polyclonal antibodies to the C terminal 672-695 (APP) and monoclonal antibodies to inter-alpha-trypsin inhibitor. Polypeptides with molecular weights of approximately 35-38 kDa were detected in the NCL brain, but not in controls in both cases. These findings suggest abnormal protein processing in NCL brain tissue, disturbances in protein and glycoconjugate metabolism, impaired lysosomal function (i.e., metabolic enzyme and/or proteases/proteinase inhibitor abnormalities), and the involvement of improperly processed APP.

Biochemical and morphological changes in rat brain synaptosomes after exposure to normobaric hyperoxia in vivo.

Adult rats were submitted to normobaric hyperoxia for 1 to 24 h, then the brain synaptosomes were isolated and their metabolic and morphologic properties were studied. Hyperoxia lasting 1-2 h significantly increased the content of thiobarbituric acid-reactive material (TBAR) and decreased the level of protein thid groups. During the next 5-8 h of hyperoxia SH groups as well as TBAR content became almost normal, reflecting adaptation of the animals to an elevated oxygen tension. After 24 h of hyperoxia a maximal increase in the TBAR content and parallel fall in protein thiol groups were noted. Simultaneously, significant morphological differences between control synaptosomes and synaptosomes isolated from rats exposed to 24 h oxygenation were observed in electron microscopy. The high-affinity dopamine uptake in hyperoxic synaptosomes was significantly increased in all experimental groups. A specific high sensitivity of the dopamine uptake system in synaptoplasmatic membranes to the free radical modification of the membrane structure is suggested.

Stress-induced injury of pig myocardium is accompanied by increased lipid peroxidation and depletion of mitochondrial ATP.

The aim of the study was to investigate the mechanisms of myocardial lesions induced by stress in conscious, intact pigs. The animals were subjected to 24 h immobilization, controls were kept in normal conditions. The pigs were killed by electric shock and exsanguination. Lipid peroxidation products: malondialdehyde (MDA), conjugated dienes (CDB), fluorescent end products (RF) and adenine nucleotides (ATP, ADP) were measured in the left ventricular myocardium which was also subjected to histoenzymatic and electron microscopic examination. In stressed animals as opposed to the control group, alterations in ultrastructure and diminution of mitochondrial ATP have been found, together with augmented formation of MDA and CDB reflecting increased free radical generation. These changes may be the component of stress-induced myocardial injury.

Altered amyloid beta-protein precursor processing in brains of patients with neuronal ceroid lipofuscinosis.

Our previous study disclosed strong immunostaining of brain tissue in neuronal ceroid lipofuscinosis (NCL) with antibodies against amyloid beta-protein and the presence of 31-kDa polypeptide in the storage material. In the present study, we investigated the immunoreactivity of the NCL brain tissue with anti-serum (anti-GID) raised against a synthetic peptide, based on the amyloid beta-protein precursor, with the 175-186 amino acid sequence. Immunocytochemistry was performed on autopsy brain material collected from 15 NCL cases, and from 8 age-matched normal controls. The results showed strong immunoreactivity of nerve cells in the NCL cases, which according to densitometry was 5 times more intense than in the control group (P less than 0.0001 by Student's t-test). Western blot analysis revealed that in protein fractions of NCL brain homogenates anti-GID recognized the protein band of 35 kDa. Thus our present and previously performed studies supplied for the first time data pointing to abnormal processing of amyloid beta-protein precursor in NCL. Moreover, the accumulation of both 31- and 35-kDa polypeptides that was demonstrated provides further support for postulated defective protein metabolism in this disorder.

Nisoldipine inhibits lipid peroxidation induced by coronary occlusion in pig myocardium.

STUDY OBJECTIVE: The aim was to investigate the effect of the Ca++ channel blocker nisoldipine on the content of lipid peroxidation products in pig myocardium after acute coronary occlusion. DESIGN: Open chest pigs were subjected to the occlusion of the left anterior coronary artery (LAD) with or without nisoldipine. After 30 min ischaemia, myocardial samples were taken from the ischaemic area and from the non-ischaemic posterior wall of the left ventricle for determination of lipid peroxidation products. SUBJECTS: Subjects were farm pigs of either sex. In 10 pigs, the LAD was occluded without drug pretreatment; 11 pigs were infused with nisoldipine (10 micrograms.kg-1) 30 min before the LAD occlusion. Sham operated controls received no drug (n = 7) or nisoldipine (n = 9). MEASUREMENTS AND MAIN RESULTS: Myocardial samples were assayed for the content of lipid peroxidation products: malondialdehyde, conjugated double bonds, and fluorescent end products. Plasma nisoldipine concentration was measured in some experiments. Following the LAD occlusion, the content of lipid peroxidation products increased in both ischaemic and non-ischaemic myocardial regions as compared to the hearts of sham operated pigs. Pretreatment with nisoldipine completely prevented these increases. At a time of the coronary occlusion, plasma nisoldipine concentrations were within the therapeutic range. CONCLUSION: Ca++ antagonist prevents the excessive peroxidation of myocardial membrane lipids which affects the whole myocardium when there is acute local ischaemia. Prevention of myocardial damage in the non-ischaemic region may determine survival of the infarcted heart.

Effect of nisoldipine on stress-induced myocardial damage in the conscious pig.

1. The effect of the calcium channel blocker nisoldipine on the myocardial content of lipid peroxidation products (malondialdehyde (MDA), conjugated double bonds (CDB), fluorescent end-products (RF) and mitochondrial adenine nucleotides) was investigated in conscious pigs (n = 14) subjected to 24 h of immobilization stress. Histoenzymatic and electron microscopic studies of the myocardium were also performed. Nisoldipine was given orally in a twice daily dose of 20 mg for 2 days before and on the day of the experiment. Results were compared with those obtained in immobilized untreated pigs (n = 10) and in non-stressed treated controls (n = 8). 2. Pretreatment with nisoldipine significantly attenuated stress-induced increase in myocardial contents of CDB and RF and prevented decline of mitochondrial adenine nucleotides. Stress-induced myocardial histoenzymatic changes (decrease of succinic dehydrogenase, ATPase, acid phosphatase activity) and ultrastructural alterations (mitochondrial damage, lysis of myofibrils, dilatation of sarcoplasmic reticulum and endothelial swelling) were also diminished. 3. It is concluded that treatment with a Ca2(+)-antagonist is beneficial to the heart exposed to environmental stress.

Evidence for increased lipid peroxidation in the non-ischaemic portion of the heart with coronary occlusion.

Lipid peroxidation products, malondialdehyde (MDA), conjugated double bonds (CDB), and fluorescent end products (RF), were measured in porcine left ventricular myocardium. Myocardial samples were taken 15-20 min after left anterior descending coronary artery (LAD) ligation from ischaemic and non-ischaemic regions. Products of lipid peroxidation were also measured in the left ventricular myocardium of sham operated pigs. Non-ischaemic regions of the hearts with LAD occlusion showed a significantly higher content of lipid peroxidation products than intact hearts of sham operated pigs. The results suggest that enhancement of lipid peroxidation reflecting increased free radical generation may contribute to myocardial damage in the non-ischaemic, apparently "healthy" portion of the heart with acute coronary occlusion.

Rabbit brain lipids during short-term hyperthermia.

Rabbits were subjected to 3 h of hyperthermia at 40 degrees C. The phospholipid content of the brain was unchanged, whereas the free fatty acids increased by about 73% over control levels. Hyperthermia also induced inhibition of fatty acid peroxidation processes. The last products of fatty acid peroxidation, the thiobarbituric acid reactive substances, diminished significantly during hyperthermia, whereas the level of the first intermediate of this pathway, the conjugated double bonds, remained unchanged. Simultaneously, the levels of lipid-soluble antioxidants decreased significantly. The content of free fatty acids, malondialdehyde, and lipid-soluble anti-oxidant returned toward control levels during 3 h of recovery. The content of gangliosides was 10-20% above control values in the groups of animals examined immediately and 3 h after hyperthermia. The ganglioside-specific enzymes, neuraminidase and sialyltransferase, both directed toward endogenous lipid substrates, were activated by hyperthermia, suggesting the stimulation of turnover of the gangliosides during the course of hyperthermia. Lipid alterations resulting from short-term hyperthermia may influence the physicochemical properties of neuronal membranes.

Consequences of exposure of rats to hyperoxygenation for actomyosin-like protein from synaptosomes of brain.

This paper describes the changes caused by in vivo hyperoxygenation of rats in the brain synaptosomal actomyosin-like protein preparation. We demonstrate that this preparation is composed of protein and phospholipid components attached to each other. Upon hyperoxygenation, a peroxidation of phospholipids in the lipid component proceeds. Simultaneously, with the increase of malondialdehyde, conjugated double bonds, and fluorescence intensity levels, a decrease of protein -SH groups levels, and Mg2+-dependent ATPase activity is observed. The above changes reduce the ability of the protein to superprecipitate, which reflects a contractile reaction in vivo.

Attenuation by prostacyclin of adrenaline-stimulated lipid peroxidation in the myocardium.

The study aimed to investigate the effect of adrenaline upon lipid peroxidation processes in the myocardium and to evaluate the effect of prostacyclin upon the content of lipid peroxidation products in adrenaline-treated hearts. Anaesthetized rabbits were infused for 2 hours with adrenaline /1 microgram/kg/min/, adrenaline and prostacyclin /2 ng/kg/min/ and prostacyclin alone; controls received saline. After termination of infusion lipid peroxidation products /conjugated double bonds CDB, malondialdehyde MDA, lipid-soluble fluorescent material, FM/ and SH groups were measured in the myocardial tissue. Adrenaline significantly increased the content of all three indices of lipid peroxidation and decreased the concentration of soluble SH groups. PGI2 infused simultaneously significantly decreased the concentration of final metabolites of peroxidation /MDA, FM/ and prevented decrease of SH groups. The content of intermediate products of peroxidation, CDB was elevated. In conclusion, enhanced free radical activity, resulting in increased peroxidation of myocardial lipids may contribute to known injurious effect of adrenaline upon myocardium. The finding that PGI2 prevents increased formation of final peroxidation products gives support for the hypothesis that anti-free radical activity may be a component of cytoprotective action of prostacyclin.

Dopamine uptake in striatal synaptosomes exposed to peroxidation "in vitro".

The uptake of [3H]dopamine by synaptosomes from rat brain striatum was biphasic, the rapid phase requiring less than 1 min for completion and the slower phase occurring over a few minutes. This uptake of labeled dopamine was enhanced by prior exposure of the synaptosomes to Fe2+, which gives rise to peroxidation of the synaptosomal lipids. Both lipid peroxidation and enhancement of dopamine uptake were unaffected by the presence of inhibitors of monoamine oxidase but were blocked by inclusion of omicron-phenanthroline in the preincubation medium. It is concluded that lipid peroxidation may be involved in certain neuropsychiatric disorders such as seizures evoked by exposure to high oxygen pressures.

Effects of lipid peroxidation on neurotransmitters uptake by rat synaptosomes.

This paper described the effect of in vitro peroxidation achieved by 60 s or 5 min exposure to 60 microM Fe2+ with 200 microM ascorbic acid, on selected properties of rat brain synaptosomes reflecting some steps of chemical neurotransmission. The studies have revealed dramatic differences between dopamine, GABA and choline high affinity uptake systems in response to peroxidation. The uptake of calcium by synaptosomes submitted to free radical oxidation, mostly its K+-depolarization-dependent portion, was significantly suppressed. In contrast, peroxidation appeared not to influence the transmembrane synaptosomal potential. It is concluded, that peroxidation of synaptic endings modifies the lipid content of synaptoplasmatic membranes and consequently leads to severe disturbances in the function of neurotransmitter uptake systems and depolarization-dependent calcium channels.
