[Investigations on LH and FSH serum levels before and after application of synthetic LH-releasing-hormone in healthy men of different age].

50 male healthy volunteers of different age had measurements of plasma luteinizing hormone (LH) and follicle-stimulating hormone (FSH). Basal levels of LH and FSH as well as maximal release of both hormones after i.v. application of 25 mug of synthetical LH-releasing hormone have been done. Results demonstrate an increasing reserve capacity of LH and FSH with age. FSH/LH ratio is altered in favor of FSH with duration of life. Sensitivity of gonadotropin producing cells of adenohypophysis to LH-releasing hormone is diminished in old age that can be recognized from a diminution of the relative rise of serum LH and FSH after LH-RH application. Correlations and partial correlations of absolute and relative body weight with age and basal LH and FSH serum levels demonstrate that age and not body weight plays a main part for increasing gonadotropin serum values. Causative responsibility for the increased LH serum concentrations is a decrease of testosterone production with age. Data obtained in this study and reports presented in literature give no convincing explanation for the increase of FSH serum levels with age.

[Age-dependent changes of hypophysis-thyroid - regulation (author's transl)].

In health men and women different age total thyroxine, free thyroxine-binding-capacity, total triiodothyronine as well as TSH in serum before and after intravenous application of 500 mcg synthetic thyrothropine-releasing-hormone (TRH) were controlled. The investigations have shown: 1. With increasing age total thyroxine level in serum does not change. 2. With increasing age free thyroxine-binding-capacity in serum does not change. 3. With increasing age in men and women a significant decrease of total triiodothyronine in serum is to be measured. 4. Basal TSH serum levels do not change with increasing age. On the other hand in men and women with increasing age significant decreased TSH increments after TRH application were measured. The investigations have shown, that with increasing age the hypothalamus-hypophysial-thyroid-axis is shifting to a lower regulation level.

[Electron microscopic investigations in the origin of 'ghost bodies' in the aorta].

Electron microscopic studies on aortas of normal mongrel dogs revealed a special form of degeneration of vascular smooth muscle cells and the mode of origin of 'ghost bodies'. These are formed by budding off portions of cytoplasm of smooth muscle cells probably as a result of worsening of the metabolic situation of these cells with increasing age.

Effects of synthetic somatostatin on endotoxin-induced changes of growth hormone, cortisol and insulin in plasma, blood sugar and blood leukocytes in man.

In normal man synthetic somatostatin inhibits the growth hormone response to intravenous administration of endotoxin (Piromen), but exerts no influence on cortisol plasma levels. Endotoxin-induced leukocytosis in blood is significantly reduced by somatostatin. Plasma levels of insulin and the blood sugar in man receiving endotoxin are firstly reduced in a significant manner and then increasing to normal values during somatostatin infusion.

Electron microscopic study on permeability of coronary artery wall of normotensive rabbits using horseradish peroxidase as a tracer.

In order to study the pathways in the coronary artery vessel wall horseradish peroxidase (HRP) was used as a tracer. HRP was injected directly in the left ventricle of rabbit hearts. 30 seconds after intracardial injection of HRP in normotensive rabbits, peroxidase reaction products could be located in random areas of the coronary arterial system and could then be observed in all three layers of the coronary walls at these locations. HRP reaches the subendothelial space of the coronary arteries along two pathways: through intercellular junctions and by plasmalemmal vesicles. Through the fenestations of the internal elastic lamina HRP reaches the media and can be detected between the media smooth muscle cells. The influence of the methodology on the results is discussed.

Experimental results to immunomesenchymal suppression.

Wet weight of cotton-pellet granuloma and 35SO4-uptake in the acid mucopolysaccharides (AMPS) of granulation tissue of rats and the number of plaque forming cells (PFC) of mice were investigated after the treatment with antilymphocyte globulin, prednisolone, cyclophosphamide and azathioprine. In this way, the effect of these substances could be observed in a mesenchymal reaction and in an immune reaction. The substances are mesenchymosuppressive as well as immunosuppressive, but the present results show that the effective dosage is higher for mesenchymal suppression than for immunosuppression. Antilymphocyte globulin did not cause an alteration of the 35SO4-uptake in AMPS. These experimental findings confirm the clinical experience in the treatment of the so-called autoimmunological diseases which are primary localized in connective tissue. These diseases need a higher dosage of cytostatic substances, glucocorticoids and antilymphocyte globulin in times of acute inflammatory exacerbation than in the chronic phase.
