Uptake of liposomes and sheep red blood cells by the liver and spleen of rats with normal or decreased function of the reticuloendothelial system.

The distribution of negatively charged liposomes in rats with normal or depressed function of the liver RES was examined. RES activity was determined by the uptake of the sheep red blood cells (SRBC). Whereas pretreatment with colloidal carbon or dextran sulphate drastically diminishes the SRBC uptake by the liver, the liposome uptake is decreased by 12-15% only. In the spleen, such pretreatment boosts the SRBC uptake five- to sixfold, whereas liposome uptake was decreased by about 50%. This indicates that phagocytosis by the RES is only of several liposome-cell interactions. Consequently, the suppression of the RES function is of no practical use when attempting to suppress the preferential liposome uptake by the liver and spleen.

[Influence of procaine and nicotinoyl-procaine on enzyme activities, nucleic acid and protein synthesis in primary cultures of heart and skeletal muscle cells of rats (author's transl)].

The comparative studies are continued on the influence of two tertiary amines, procaine and nicotinoyl-procaine, on isolated heart and skeletal muscle cells in culture. A daily addition of these substances to cell cultures produces an increase in the specific activities of glutamate dehydrogenase and glutamate-oxaloacetate transaminase and an inhibition of alteration in the isoenzyme pattern of lactate dehydrogenase. The presence of procaine or nicotinoyl-procaine, resp., inhibits thymidine incorporation by cells in culture, whereas the incorporation of thymidine is increased after pretreatment of cells with these substances. These results support the findings of an influence of said tertiary amines on the metabolism of cells in culture.

[Influence of procaine and nicotinoyl-procaine on primary cultures of cerebellum and kidney cells from neonatal rats (author's transl)].

A comparative study was performed on the influence of two similar tertiary amines, procaine and nicotinoyl-procaine, on isolated kidney and cerebellum cells in culture. Shortly after the addition of the test substances vesicles are found in the cells and the cells are more broadly attached to the underground. But the attachment of microfilaments is scarcely adhered. Daily addition of procaine or nicotinoyl-procaine, resp., to cell cultures leads to an increase of protein concentration and enzyme activities. Particularly organotypical enzymes are enhanced, as gamma-glutamyltransferase in kidney cells and creatine kinase in cerebellum cells.

"In vivo" distribution of liposomes between parenchymal and non parenchymal cells in rat liver.

The in vivo uptake of 3H-methotrexate containing, 14C-cholesterol labelled liposomes was determined for the two major liver cell populations, parenchymal cells (PC) and non-parenchymal cells (NPC) after intravenous injection of the liposomes into normal rats and rats with a portocaval shunt, followed by isolation and purification of the two cell types. One hour after injection, the purified NPC had bound 2-3 times more of the liposome-attached radioactivity per cell than the purified PC, whereas 6 hours after injection the ratio was inverted. In animals with a portocaval shunt, the PC contained 3-4 times more liposomal radioactivity per cell than NPC already 1 hour after injection, although the total uptake by the whole liver was not diminished under these conditions. Recalculating the data obtained from the isolated cells for the whole liver, it is found that 1 hour after injection the liver NPC had bound the same amount of liposomal radioactivity as liver PC, although they account for only 7% of the liver volume. After 6 hours, the PC had bound 5 times more liposomal radioactivity than the NPC; this ratio is achieved in animals with a portocaval shunt already 1 hour after injection. This shift is simultaneously caused by a gain of PC-bound radioactivity and decrease of NPC-bound radioactivity.

[Influence of local anaesthetics on isolated muscle cells in culture (author's transl)].

A comparative study was performed on the influence of a series of chemically defined local anaesthetics such as procaine, nicotinoylprocaine and butacaine on isolated skeletal muscle cells and heart cells in culture. About 30 min after addition of local anaesthetics to cell cultures vesicles appear in the cells. These formations are reversible and need a minimum concentration of 13(-3) mol/l procaine, 10(-4) mol/l nicotinoylprocaine, or 10(-4) mol/l butacaine, respectively. Soon after addition of local anaesthetics a reduction of oxygen consumption is measurable in both cell types. The degree of this reduction depends on the chemical structure and the concentration of local anaesthetics and on the kind of cells. After daily addition of local anaesthetics to cell cultures metabolic parameters were studied. The local anaesthetics, except for butacaine, produce an increase of protein concentration, creatine kinase and lactate dehydrogenase activity per sample and an increase of the specific activity of lactate dehydrogenase. These increases depend also on the concentration of local anaesthetics and on the kind of cells. These results support the hypothesis of local anaesthetics action via membrane lipids.

Predictibility of the stability constant of a radium-cryptate by means of in vivo data from radioactive alkaline earthes.

By means of a formula, developed by J. Schubert[9] and A. Catsch, H.J. Heller[3] as well as a relation postulated by A. Catsch[1] the "thermodynamic" stability constant of the Radium (222)-cryptate (KRaRa(222) was calculated from measurements of the total body retention of the total body retention of the radioactive alkaline earthes 85SR, 140Ba and 224Ra and its (222)-cryptates in rats [5-7]. From the same in vivo data a direct lineary relationship between the log of the effectiveness quotient, log EQM(222), and the log of the "thermodynamic" stability log KMM(222) was found graphically. The values from the graph correspond with those of the calculation.
