Reversal of increased microvascular permeability associated with ischemia-reperfusion: role of cAMP.

Ischemia-reperfusion (IR) is a form of oxidant injury known to increase microvascular permeability in the lung. Agents that increase adenosine 3',5'-cyclic monophosphate (cAMP) levels have been shown to have beneficial effects in several models of oxidant lung injury associated with increased microvascular permeability. We investigated the role of adenylate cyclase activation with isoproterenol (ISO) or forskolin (FSK) in reversing the increased microvascular permeability associated with IR. ISO or FSK administered after 45 min of ischemia and 46 min of reperfusion caused a reduction in the capillary filtration coefficient (Kfc) from 1.25 +/- 0.13 to 0.53 +/- 0.08 and 0.55 +/- 0.10 ml.min-1.cmH2O-1.100 g tissue-1, respectively, at 90 min of reperfusion. This reduction in Kfc was accompanied by a rise in perfusate cAMP levels from 16.5 +/- 4.9 and 31.2 +/- 11.9 pmol/ml at 45 min of reperfusion to 444.2 +/- 147.8 and 276.1 +/- 91.0 pmol/ml at 105 min of reperfusion in lungs treated with ISO or FSK, respectively, at 46 min of reperfusion. Dibutyryl cAMP (DBcAMP), a membrane-permeable cAMP analogue, mimicked the permeability effect by reducing Kfc to 0.67 +/- 0.15 at 90 min of reperfusion. Significant hemodynamic changes occurred but were small and cannot explain the observed effect on Kfc. Photomicrographs from lungs treated with ISO or FSK revealed a reversal of the morphological manifestations of increased microvascular permeability. We conclude that the increased microvascular permeability associated with IR can be reversed by ISO, FSK, and DBcAMP and that cAMP produced by the lung contributes to the observed reversal.

A large parosteal osteosarcoma with transformation to high-grade osteosarcoma: a case report.

This case of a large (31 cm) parosteal osteosarcoma had x-ray documentation 4 1/2 years before treatment. When the tumor began enlarging rapidly, treatment was sought. The tumor histologically showed typical parosteal osteosarcoma as well as malignant cartilage, osteoid, and undifferentiated sarcoma. The case shows one course a parosteal osteosarcoma can take over a 7-year untreated period. The significance of high-grade sarcoma and intramedullary involvement is discussed. When unequivocal high-grade osteosarcoma is present, it should be treated with radical resection and adjuvant multi-drug chemotherapy.

A tridimensional view of intradermal nevi as revealed by scanning electron microscopy.

When scanning electron microscopy (SEM) was applied to the study of intradermal nevi, interesting tridimensional features were recognized. The free surface of the lesions showed ruffled keratinized cells. "Normal" hairs as well as "corkscrew" hairs emerged from the follicular openings. Nevus cells were either round or elongated and surrounded by connective tissue fibers.

Scanning electron microscopy of dermatofibromas.

The three-dimensional morphology of six dermatofibromas was studied by scanning electron microscopy. Hypertrophic epidermal ridges covered with large keratinized cells overlay each dermal lesion. Small collagenous fibers in the tumors contrasted with the bundles of much larger collagenous fibers in the normal dermis peripheral to the lesions.

Low strength anthralin in psoriasis.

Anthralin in low concentrations (0.01%--0.03%) was applied in an ointment form once per day in the hospital to 130 psoriasis patients. The treatment program included also the use of UVB light, emollients and bath oil and an antihistamine. Clearing of psoriasis was achieved in all patients in an average time of 4 weeks. The involution of the lesions was carefully studied by correlated clinical observations, light microscopy, transmission electron microscopy (TEM) and scanning electron microscopy (SEM). Striking healing of the holes in the stratum corneum and of the gaps in the basement membrane occurred. Low strength anthralin, in addition to its effectiveness, did not produce irritation, staining or side-effects.

Sexual differences among duodenal glands of hamster.

This histochemical study was undertaken to determine whether sexual differences existed in the duodenal glands of hamster. Alcian blue staining and atomic absorption spectrophotometry (for copper content of the bound Alcian blue) showed the duodenal glands of males to contain about double the acid mucosubstance of the female glands. Periodic acid Schiff staining was stronger in the females over a larger portion of the gland than in males. It was concluded that sexual differences were evident in duodenal glands of hamster.

Ultrastructural basis for the assay of topical acne treatments. Transmission and scanning electron microscopy of untreated comedones.

After two decades of using systemic antibiotics as a main treatment for acne, emphasis is again being placed on topical agents. Thus, it is highly desirable to have a procedure whereby the activity of the various compounds can be evaluated by direct visualization. Scanning electron microscopy combined with transmission electron microscopy provide the tools for such in assay. This study describes the ultrastructure of untreated comedones and provides the baseline or control data necessary for testing topical treatments. Comedones obtained by punch biopsies or comedo extractors were processed for electron microscopy and studied with an ETEC Autoscan Scanning Electron Microscope and with Philips EM 300 and EM 301 Transmission Electron Microscopes. Microorganisms, keratinized cells, sebum, and hairs interact with each other to form the comedonal mass. Corynebacterium acnes and Pityrosporum ovale proliferate abundantly in close association with sebum and penetrate the keratinized cells. Fine structural details of bacteria and yeasts as well as features of host-microbial relationship have been elucidated.

New observations on the physiology of menstruation.

Endometrium was studies histologically, histochemically, and ultrastructurally in a series of biopsies taken from 3 normal, ovulating patients on Days 1-9 of the cycle. The occurrence of ovulation and the adequacy of progesterone were determined by radioimmunoassay. The most striking feature of menstruating endometrium was its vigorous attempt to survive. This was manifested by lysosomal activity, lipid accumulation, expulsion of glycoproteins, and the uptake of stromal debris by epithelial cells for passage to the uterine cavity. Regression, rather than cell death, was the chief event of menstruation. While some cells of the spongiosa underwent necrosis, the vast majority remained viable and underwent remodeling to participate in the new cycle. These studies may lead to further understanding of the process of menstruation and the pathophysiology of anovulatory bleeding and irregular shedding of the endometrium.

[Effects of quingestanol acetate on the histology, histochemistry and ultrastructure of the endometrium].

According to studies carried out using the optical microscope and the electron microscope quingestanol acetate, which is a powerful progestagen, modified significantly the histology and the histochemistry of the endometrium. The mechanisms of transport across the cellular membranes are not changed, but probably there is competition between the oestrogens at the binding sites as a result of which the synthesis of proteins and hydrocarbons is changed. This in turn provokes the formation of aggregations of platelets in the blood vessels and cellular degeneration starting from the onset of the cycle. The contraceptive efficiency of quingestanol acetate when it is administered by itself is due to alterations in cellular biology which it brings about at the level of the endometrium as well as due to its inhibitory effect on gonadotropins.
