Isolated hepatocytes from newborn rats are able to produce ketone bodies from added medium-chain and long-chain fatty acids. Carnitine enhances the rate of ketone body synthesis from palmitate as well as from caprinoate. The 3-OHB/AcAc ratios indicate a highly reduced state of the mitochondrial redox carriers in the presence of both fatty acids and carnitine. Ketogenesis from palmitate accounts for about 90% of the total beta-oxidation. At recovery of 95% of the radioactivity two thirds of totally fatty acid uptake are channeled into esterification, whereas the remainder is oxidized. alpha- and beta-agonists stimulate glycogen degradation and glucose release and reduce net lactate production in hepatocytes from newborn rats. The (1-14C)-palmitate uptake is not altered by alpha- and beta-agonists. Phenylephrine significantly enhances 14CO2 production from (1-14C)-palmitate. Neither of the agonists affects the rate of esterification or of ketone body production with palmitate as substrate. Isoproterenol, however, stimulates ketogenesis from caprinoate even in the presence of optimal carnitine concentrations.
A well reproducible method for preparing isolated parenchymal cells from fetal rat liver on the 22nd day of gestation by tryptic digestion is described including criteria for assessing their viability. The content of parenchymal liver cells was higher than 90% as could be demonstrated by light microscopic analysis of thin sections of the pelleted hepatocytes. Electron microscopic examination of the freshly prepared suspension revealed a well preserved ultrastructure of the hepatocytes. The metabolic competence of the isolated fetal hepatocytes was tested by alpha- and beta-adrenergic stimulation of glycogenolysis and glycogenolytic glucose release.
Using the results of the chemical amino acid analyses of the unconventional proteins blood, collagen and wool and the conventional proteins soya, maize and casein, protein mixtures were developed. The quality of the protein mixtures was estimated in feeding experiments with growing male Sprague-Dawley rats and expressed as protein efficiency ratio (PER). Different blood parameters and the weight of some organs were also recorded. Blood, collagen and wool protein are very poor in different essential amino acids, therefore their quality is very low, it can however be significantly improved when they are fed in mixtures with other animal or plant proteins of higher quality. In mixtures of three components only containing blood (10--40%), collagen (10--25%) and casein (50 to 70%) the deficit in the content of essential amino acids cannot be compensated. Rats fed with such diets showed, however, decreased food intake and depressed growth. The relative PER values of those mixtures were 44--69% of that of casein. The best supplementing effect on the imbalanced amino acid patterns was achieved when the protein mixture was developed from 4 or 5 different proteins. The quality of these five-component-mixtures amounted to 94--100% of that of casein. In these mixtures the percentage of casein in the protein mixture amounted to only 35% and soya or maize to 15%. Exactly 50% of the protein supply could be derived from hydrolysates of the unconventional proteins blood, collagen and wool.
The quality of protein mixtures on the basis of blood protein, collagen, wool and other proteins was tested using different chemical and biological criteria of protein evaluation. Before mixing the single proteins were hydrolysed enzymatically by means of acid or alkaline proteinases. In this study the influence of both hydrolysis processes on the bioavailability of the protein mixtures was also considered. The N-balance experiments were carried out with weanling Sprague-Dawley rats in groups of n = 6 animals each. The estimated BV's showed that the protein quality was significantly affected by both hydrolysis procedures especially when the hydrolysates were compared with the non-hydrolysed protein components. The BV's also showed that the quality of all mixtures of protein hydrolysates was less than that of casein (reference protein) and the availability of the alkaline hydrolysed proteins better than that with acid proteinase treated proteins. This result could be confirmed by the calculated chemical criteria E/T- and E/N-ratios, EAA-Index and Arnould-Index. The results of the growth-criteria PER, NPR and RNV did neither correspond with each other nor they were of good agreement with N-balance based BV and NPU.
Fetal hepatocytes isolated at day 22 of gestation by trypsin digestion of rat liver retain their responsiveness to hormones. Glucagon significantly stimulates glycogenolysis and glucose release to 115 and 124%, respectively. The effect of alpha- and beta-agonists is more pronounced enhancing glycogen breakdown and glucose release to 133 and 147% (L-phenylephrine) and 183 and 202% (isoproterenol), respectively. The isolated fetal hepatocytes obtained by trypsin digestion are a useful tool for studying the hormonal control of liver metabolism during the perinatal stage without limitations met otherwise with extrahepatic factors and the effect of altered blood flow.
By means of the "multivariate variance analysis" ("MANOVA"; Friedrich and Olfert, 1977), we tried to differentiate the histologic types of malignant melanomas also by quantifiable criteria. The investigation depended upon data from 411 patients. Difficulties arose from incomplete data material. But in principle, the indication of this method is given in such problems. If the number of cases could be augmented, the method would be helpful as a support for diagnostic quality control.
Histologic findings and clinical data of 411 patients with malignant melanomas treated in 1952 up to june 1977 reveal an improvement of diagnostic accuracy. Within the last years, fewer patients were referred to the clinic with extensive tumor growth or with metastases. A change from the levels of invasion V and IV to III is evident. Further improvement to level II and I with a corresponding smaller tumor thickness is appearing in outlines.
The 5'-nucleotidase considered as a marker enzyme of the plasma membranes shows a highly specific acitvity in both nuclear and mitochondrial-lysosomal fractions of the rat yolk-sac. In isopycnic sucrose gradient fractionation of the mitchondrial-lysosomal fraction a relative accumulation of this enzyme could be shown in the density level of the phagolysosomes (d = 1,16--1,20).

A case of a severe Candida sepsis is reported, which was treated successfully by a combination therapy of flucytosine with fluconazole. After an extensive abdominal operation, a 70-year-old man developed a syndrome of fulminant sepsis due to Candida albicans with the beginnings of renal failure. The latter fact forced us to search for a therapeutic alternative to the classical amphotericin B plus flucytosine combination therapy.
