Laser microdissection and laser pressure catapulting for the generation of chromosome-specific paint probes.

Chromosome-specific paint probes provide a powerful tool with wide applications in cytogenetic analysis. Here, we present a new approach using UV-laser microbeam microdissection in combination with laser-pressure catapulting, which allows the fast isolation of single chromosomes for the generation of chromosome-specific paint probes. To demonstrate the feasibility of this approach, single chromosomes were collected and amplified with degenerate oligonucleotide-primed PCR, hapten-labeled and hybridized onto normal metaphase spreads. Fluorescence in situ hybridization signals revealed specific painting of the respective chromosomes.

[Long-term results and histologic studies following surgical correction of congenital penis deviation].

Between 1982 and 1988, 11 patients with congenital penile curvature without hypospadia or chordee were operated according to the technique originally described by Nesbit. In all these patients the operative results were reevaluated after a period of 1 1/2 to 7 years (average 3 3/4 years). Normal sexual intercourse was possible for all 11 men, and good long-term cosmetic results were noted. In contrast to formerly published results, histological examination of the segments resected from the tunica albuginea showed fibrotic tissue in some cases, which could play an important role in the etiology and/or pathogenesis of congenital penile curvature.

Characterization of a newly established human urinary bladder cancer cell line (BT-1).

A new human transitonal cell carcinoma cell line has been established in long term tissue culture. The BT-1 cells were derived from a poorly differentiated human bladder cancer. The tumor cell line produces tumors in nude mice. BT-1 has been characterized by cytogenetic and flow cytometric analysis and by isoenzyme typing. As in direct preparations of human bladder tumors, an isochromosome 5p is a consistent marker of the newly established line. The BT-1 cells produce transforming growth factors but do not respond to exogeneous EGF.

Interphase cell flow cytometry as a means of monitoring genomic size in normal and neoplastoid cell cultures.

The DNA specific fluorescence of mass cultures and clones derived from human skin and bladder tumor tissue was assayed by flow cytometry. In order to detect and quantitate small fluorescence intensity changes, cytogenetically defined triploid or diploid human fibroblast strains were cocultivated, harvested, and stained with the cell strain of unknown karyotype. The triploid standard (derived from human abortus tissue) proved chromosomally unstable at high passage level. Fifteen male, female, and 45,X strains displayed target-to-standard cell fluorescence ratios commensurate with their respective chromosome constitutions. Interstrain variation was highest among the 45,X strains, although mosaicism could not be detected by conventional cytogenetics. Interclonal fluorescence variation was two- to ten-fold higher among the tumor-derived clones tested. Chromosome counts and subcloning experiments indicate that this increased fluorescence variation is due to genome size variation. The clonal evolution of genome size differences was observed in subclones of chromosomally divergent parental clones. These observations suggest that well controlled flow cytometry can adequately resolve subtle degrees of genome size variation in cultivated human cells. The technique is especially suited for monitoring genome size changes in cultivated tumor cells.

Diagnosis and treatment of renal angiomyolipoma (based on 15 cases).

Fifteen cases of renal angiomyolipoma not associated with tuberous sclerosis are presented. The clinical and radiological features, the pathology and the management of these rare but well-documented tumors are discussed. They pose problems in diagnosis, and controversy still exists regarding their management. Additional techniques such as ultrasonography and computerized tomography may help in the diagnosis. When conservative surgery is not possible, or when a solid tumor cannot be confidently excluded, nephrectomy should be performed.

Identification of inhibitory growth factors from a N-[4-(5-nitro-2-furyl)-2-thiazolyl]formamide-induced murine bladder cancer cell line.

The N-[4-(5-nitro-2-furyl)-2-thiazolyl]formamide (FANFT)-induced murine bladder carcinoma cell line MBT-683 contains growth-stimulating (TGF) and growth-inhibiting factors (GI). These activities coeluted on BioGel P-30 between molecular weights 6,000 and 29,000 daltons. They can be separated by high performance liquid chromatography. Both activities were destroyed by trypsin and dithiothreitol. However, they were resistant to acid and heat. Pool 1 (TGF) stimulated the growth of normal rat kidney cells, whereas pool 2 (GI) inhibited the colony formation of the cell lines MBT-683 and RBTCC-8.

Are antireflux procedures indicated in patients with small pyelonephritic kidneys?

Antireflux surgery was successfully performed in 16 adults and children with bilateral or unilateral vesicoureteral reflux associated with a small pyelonephritic kidney. Although in 14 cases of the preoperative 131I-Hippuran clearance of the affected unit was less than 100 ml/min/1.25 m2, in 10 cases renal function was stabilized or slightly improved postoperatively. Only 1 child showed a severe deterioration of renal function. All 16 patients were cured of their acute pyelonephritis. Surgical correction of the reflux had no beneficial effect on renal size or renal scars. Urinary tract infection was eliminated in 12 of the 16 cases. It can be concluded that a small pyelonephritic kidney associated with a vesicoureteral reflux should not be removed even if its isotopic clearance is less than 100 ml/min.

[Behavior of modern suture materials in infected urine].

The recently developed absorbable materials demonstrate a constant and reproducible high quality when compared with the resorbable suture materials. However, based on in vitro studies by different authors the use of these materials in infected urine is not recommended. We investigated in vivo in infected urines the stability of the suture materials composed of polyglycolic acid, and compared the results obtained in vitro and in vivo. In a controlled study the stability of the absorbable suture materials polyglycolic acid, polyglactin 910 and polydioxanon 4/0 USP were tested in infected urine in vivo and in vitro. In 21 patients with infected urine the suture materials were placed into the bladder for 7 days. In another experiment these materials were incubated up to 7 days in culture flasks with infected urine. After that the suture materials were examined by densitometry and electrone microscopy. When using the results obtained in vitro in clinical work caution is recommended. However, the in vivo tests show that the stability of the newly developed absorbable materials is not influenced dramatically by the infected urine.

Human lactic dehydrogenase as a marker for monitoring the growth of prostatic carcinoma in nude mice.

Human lactate dehydrogenase (1,1,1,27-LDH) isoenzymes were determined in human prostatic carcinoma cell lines. The isoenzymes LDH-4/LDH-5 were identified and quantified in the serum of athymic mice bearing the human prostatic carcinoma cell lines PC-3, PC-3/M and DU 145 subcutaneously. The amounts of the slowly migrating isoenzymes released by the solid tumors correlated with increasing tumor volume and tumor weight. These correlations indicate that the human LDH-4/LDH-5 isoenzymes are useful parameters for monitoring the presence and growth of prostatic cancer in the athymic animal model.

[Ureteral injuries in intervertebral lumbar disk surgery].

In lumbar disc surgery ureteral transection is a rare complication and only nine such cases have been reported so far. They are analyzed and two more cases are described. The complication in the first case was recognized immediately and the patient was treated successfully by end-to-end anastomosis of the ureter. The diagnosis in the second case was delayed and this lead to a septic course and finally required nephrectomy.

[Ureteral polyposis].

In connection with a case report a review is given on the clinical aspects of ureteral polyposis. The symptomatology of this condition differs considerably. The most frequent symptom is gross hematuria. Ureteral polyposis will be verified in most cases by a retrograde ureteropyelogram. Resection of the tumor bearing ureteral segment should be performed in order to exclude a malignant tumor.
