[Stresses and health risks of farm women].

Women farmers have a specific profile both of illness and of complaints, exhibiting a higher frequency of diseases than women of other occupations but, at the same time, registering fewer complaints. Women farmers tend to consider their own state of health less satisfactory than do other women but demand medical attention less readily. This seemingly contradictory picture is discussed and interpreted, taking into consideration the special situation of these women and the stress factors to which they are subjected. The results were taken from an study conducted in southern Wuerttemberg in 1990 during the course of which 775 farmer's women were questioned about their state of health.

Effects of human thyroxine-binding globulin and prealbumin on the reverse flow of thyroid hormones from extravascular space into the bloodstream in rabbits.

A plasma fraction rich in thyroid hormone-binding globulin (hTBG, human thyropexin) was injected iv into rabbits in order to see whether thyroid hormone concentrations in plasma would increase by return of T3 and T4 from the extravascular space. For this purpose, both [125I]T3 and [131I]T4 were simultaneously injected. After 1 h, or after 16 h in another series of experiments, 50 mg hTBG were injected iv. Thereafter, the mean radioactivity of both [125I]T3 and [131I]T4 in the plasma rose, and reached its peak 20-30 min after hTBG injection; [125I]T3 and [131I]T4 returned to the preinjection value slowly, after more than 3 h. When hTBG was injected 15-16 h after the radioactive hormones, the mean radioactivity of [125I]T3 reached its peak about 1 h after hTBG injection and returned to the base value after approximately 5.5 h, [131I]T4 reached its peak about 1 h after hTBG injection and returned to the base value within 12 h. After injection of hTBG, total T4 and T3 concentrations in plasma increased about 3- to 5-fold over the base values. At the same time, the percentage of both, free T4 and free T3 dropped instantly whereas absolute free T4 and free T3 values remained almost constant. After injection of 500 mg transthyretin (hTBPA), a similar flux of [125I]T3 and [131I]T4 was observed, whereas 500 mg human serum albumin were ineffective. These marked effects of injected hTBG and hTBPA on the serum levels of [125I]T3, [131I]T4, and total T3 indicate that reentry of T3 and T4 into the intravascular compartment is an important component of thyroid hormone distribution and transport. As can be anticipated from the animal experiments, the efficiency of plasmapheresis or hemofiltration methods may be improved by previous application of large doses of hTBPA or hTBG in cases of thyrotoxicosis.

Differentiation changes in cord blood T lymphocytes induced by synthetic C-terminal peptides of thymosin alpha 1.

Thymosin fraction 5 and its component thymosin alpha 1 has been reported to play a regulatory role in the later stages of T lymphocyte differentiation. Previous studies from our group have also demonstrated that thymosin fraction 5 can induce changes in purine degradative enzymes and in surface antigenic markers of cord blood lymphocytes, which have been shown to be immature compared with normal adult lymphocytes. Birr et al., have shown that the C-terminal region of thymosin alpha 1 is essential for the biological activity. In this study, the effects of these synthetic peptide fragments on cord blood T lymphocytes were studied. After incubation with different peptide sequences, cord blood lymphocytes were analysed for surface antigenic markers (OKT4/OKT8) and for purine degradative enzymes (PNP, 5 NT). In the range of 1.3 X 10 to 1.3 X 10(-5)M, one of the eleven peptides examined (sequence 13-19) exhibited activity approximately equal to that of the parent peptide thymosin alpha 1: increase in activity of 5'NT (p less than 0.01) and reduction of cells positive for OKT4 (p less than 0.01). Another sequence (20-28) was able to induce an increase in 5'NT only and a third one (20-25) a reduction of cells positive for OKT4. The results obtained in this study confirm that the C-terminal region contains molecular signals for T cell differentiation.

Immunoadsorbents: non-specific binding of proteins to albumin-sepharose.

Human serum albumin-Sepharose was prepared by coupling human serum albumin to cyanogen bromide activated Sepharose 4B. This immunoadsorbent showed considerable non-specific protein adsorption. The adsorbed proteins were mainly immunoglobulins which could not be separated from required antibody. It is suggested that basic groups formed in the preparation of the albumin-Sepharose are responsible for this non-specific protein adsorption. Non-specific protein adsorption could be completely eliminated by neutralizing the basic groups of the albumin-Sepharose with the anionic dye blue dextran. The resultant blue dextran-albumin-Sepharose allowed purification of anti-albumin-antibody with high yield. It is shown that the isolated antibody is pure and retains its native properties.
