Assessment of plasma profile of pregnancy-associated glycoprotein (PAG) in sheep with a heterologous (anti-caPAG55+59) RIA and its potential for diagnosing pregnancy.

The purpose of the present investigation was to generate pregnancy associated glycoprotein (PAG)-profiles throughout pregnancy in a heterogenous sample of sheep using a radioimmunoassay with a heterologous antibody (anti-caPAG(55+59), #708) and utilize them for the purpose of pregnancy detection. From 2 weeks after the introduction of males into the breeding herd until 4 weeks after parturition, weekly blood samples were collected from 66 pregnant and 25 non-pregnant ewes of various breeds. Between 3 and 5 weeks after conception, plasma PAG levels increased, remained almost stable until week 17, then continued to increase, culminating in a drastic surge during the last 2 weeks of pregnancy. By 4 weeks of gestation, the plasma PAG level exceeded the level typical for non-pregnant ewes by five standard deviations, permitting a reliable pregnancy diagnosis. Plasma PAG levels were higher in twin-bearing ewes than in ewes carrying a single lamb, differences getting more evident as pregnancy proceeded. Neither breed and parity of the mother nor sex and weight of lambs borne exerted a significant effect. The heterologous assay system utilizing a caprine antibody proved to deliver results that are more consistent and less depending on various variables than those used in other studies. It may be concluded that, at the present state of development, the assay provides a reliable means of diagnosing pregnancy in sheep from the 4th week after they have been bred onward.

Technical Note: Transcervical deep cornual insemination of goats.

A newly developed technique for trans-cervical deep uterine insemination of goats is described. An experiment was conducted to compare this transcervical method with the laparoscopic intrauterine insemination using frozen-thawed semen. Of the 46 primiparous and 21 multiparous estrus-synchronized Boer goat does inseminated transcervically, 71% (22/31) kidded compared with 53% (19/36) of does inseminated laparoscopically (P < 0.025). The corresponding litter sizes were 2.27 and 1.89 (P < 0.01). In another 34 parous does inseminated transcervically without preceding hormone treatment, kidding rate and litter size were 71% and 1.76, respectively. We conclude that transcervically conducted bilateral deep cornual insemination may be considered a viable alternative to laparoscopic insemination.

Cloning and characterization of boar epididymal secretory proteins by homology to the human.

Northern blot analysis suggested that the boar epididymis produces closely related counterparts to human epididymal proteins HE1, HE3, HE4, HE5 and HE12. 'Full-length' cloning by nucleic acid and amino acid sequence similarity was achieved by RT-PCR methods in the case of the porcine counterparts of HE3 and HE4, while the homologues of HE5 and HE12, despite their cross-hybridization during Northern blot analysis, have not yet been cloned. The two novel porcine cDNAs were derived from moderately abundant epididymal mRNAs that were 75 and 83% identical to HE3 and HE4 cDNAs, respectively. To emphasize their relationship to the corresponding HEs, they were named Se3 and Se4 cDNAs. Their open reading frames predicted small secretory proteins with 55% (Se3) and 76% (Se4) conserved amino acids. Monospecific antipeptide antibodies to HE secretory proteins identified He3- and HE12-related proteins on Western blots of porcine epididymal fluid and semen. Both Northern and Western analyses indicated that the Se proteins were produced in a regionalized pattern and accumulated in the cauda fluid.

Embryo collection in prepubertal gilts and attempts to develop an improved embryo transfer technique.

Prepubertal gilts were treated with 1,500 iu equine chorionic gonadotrophin, followed 72 hours later by 500 iu human chorionic gonadotrophin (hCG), and inseminated 36 and 48 hours later. Embryos were collected at slaughter 168 hours after the hCG treatment. Blastocysts classified as 'good' or 'fair' were transferred to synchronised recipients, either by conventional surgical means or by a 'semi-endoscopic' approach, and the recipients were slaughtered four weeks later. Of 238 donor gilts, 98.4 per cent had responded with a mean (se) 23.5 (1.0) ovulations and 19.1 (1.0) ova or embryos, of which 47 per cent were considered morphologically intact and transferable. The large proportion of non-transferable embryos was not associated with the age or weight of the gilts, the season or with their housing conditions. Conventional surgical transfer of 15 to 20 (mean 17.4) blastocysts to synchronised recipients yielded 88 percent (14 of 16) pregnancies with between seven and 14 (mean 8.2) viable fetuses, and an embryo survival rate of 47 per cent in the pregnant recipients and 41 per cent in all the recipients. The corresponding data for the semi-endoscopic transfers were 16 to 20 (mean 17.7) blastocysts transferred, 47 per cent (eight of 17) pregnancies, four to 12 (mean 7.3) viable fetuses per pregnant recipient and an embryo survival rate of 41 per cent in the pregnant recipients and 19 per cent in all the recipients. Significantly fewer of these recipients became pregnant and a significantly smaller proportion of the embryos survived (P<0.05).

Immunocytochemical analysis of vimentin expression patterns in porcine embryos suggests mesodermal differentiation from day 9 after conception.

The expression of the intermediate filament proteins vimentin and keratin in porcine embryos was studied by whole-mount immunocytochemistry between day 7 and day 11 after conception. Expression of vimentin was first detected in the inner cell mass of about 50% of the 9-day-old embryos. In elongated 11-day-old embryos, cells expressing vimentin were observed in the epiblast (after disappearance of Rauber's membrane) and in cells migrating from the epiblast between the trophoblast and the underlying hypoblast layer. A keratin-positive response was observed in trophectoderm cells at all stages. These findings suggest that inner cell mass cells in the pig start differentiating into mesodermal cells not later than day 9 after conception. While the delamination of the mesodermal germ layer is known to correlate with the loss of pluripotency of the inner cell mass cells, the early onset of mesodermal differentiation in the porcine embryo, characterized by vimentin expression and in contrast to the mouse, could in part be responsible for the lack of success in establishing pluripotent embryonic stem cell lines in this species. Our results suggest that further attempts to isolate inner cell mass-derived pluripotent cells should be attempted well before day 9 after conception.

Hormonal response of female goats to active immunization against a recombinant human inhibin alpha-subunit, and establishment of an enzyme-linked immunosorbent assay for caprine follicle-stimulating hormone.

The effect of selective immunosuppression of endogenous inhibin in goats on FSH, LH, progesterone and estradiol-17beta profiles was studied during the breeding and nonbreeding seasons. Eighteen adult female Boer goats were immunized against the recombinant human inhibin alpha-subunit (hINH-alpha). With the exception of estradiol, which was determined by radio-immunoassay (RIA), all plasma hormone concentrations were determined by ELISA. The ELISA for FSH presented in this paper was established in the authors' laboratory, based on an existing RIA. Mean basal concentrations of FSH were not affected by immunosuppression of endogenous inhibin, nor was there a difference in the amplitude of the pre-ovulatory FSH surge. Immunization against inhibin appears to eliminate the slight secondary rise of FSH occurring 12-20 h after the major surge associated with ovulation. The LH profiles of the immunized goats were characterized by lower basal concentrations both before and after the pre-ovulatory LH surge which itself was reduced by 50% in immunized does. By contrast, concentrations of circulating estradiol were significantly elevated after inhibin-immunization. Progesterone profiles were not affected. Extending immunization into the anoestrous season by a booster injection of hINH-alpha, implicating oestrus induction with a progestagen and eCG, produced no discernible differences in FSH and LH profiles in comparison with nonimmunized control goats. The findings suggest that in goats, paracrine factors may play a more significant role in controlling follicular activity than a feedback mechanism acting via the pituitary.

Technical note: Vitrification of goat embryos by the open pulled-straw method.

The suitability of the open pulled-straw (OPS) method for vitrifying bovine embryos was tested for goat embryos. Of 14 does receiving OPS-vitrified embryos, all became pregnant and 13 (93%) kidded. The corresponding values for an established conventional freezing program were 58% pregnant and 50% (6/12) kidding. Overall embryo survival amounted to 64% (18/28) for OPS-vitrified and 42% (10/24) for conventionally frozen embryos. All differences were statistically significant. It is concluded that OPS vitrification is a suitable method for cryopreserving caprine d-7 blastocysts.

Birth of a piglet derived from an oocyte fertilized by intracytoplasmic sperm injection (ICSI).

This report is of a live piglet derived from an embryo produced by intracytoplasmic sperm injection and carried to term. In vivo-matured oocytes were injected with in vitro-capacitated spermatozoa and after 40 h of culture, 2 to 4-cell-stage embryos were transferred to the oviducts of oestrus induced gilts 24 h after the expected ovulation time. Pregnancy of the surrogate mothers was maintained by estrogen treatment on days 10 to 16 after transfer. The single piglet that was born to a mother that had received 32 embryos was underweight, though healthy and lively.

The effect of heparin, caffeine and calcium ionophore A23187 on in vitro induction of the acrosome reaction in frozen-thawed bovine and caprine spermatozoa.

The effect of heparin (5 IU), caffeine (5 mM) and calcium-ionophore A23187 (0.1 mM) on motility and in vitro induction of the acrosome reaction in glass wool filtered frozen-thawed bull and goat semen was studied. The motile spermatozoa fraction was obtained after glass wool filtration of frozen-thawed semen. The seminal plasma was removed from filtered semen by centrifugation, and the sperm pellet was resuspended in Sperm-TALP medium. Samples of treated and untreated control semen of both species were incubated at 37 degrees C. At 1, 15 and 30 min of incubation the proportions of progressively motile and acrosome-reacted spermatozoa were assessed. Trypan blue and Giemsa stain was used to differentiate live and dead spermatozoa having undergone acrosome reaction. Glass wool filtration enhanced the proportion of motile spermatozoa from 43% to 62% in the bovine and from 41% to 60% in the caprine. Whereas the effect of incubation with caffeine, heparin and calcium-ionophore on spermatozoan motility was negligible, the treatment of semen with calcium-ionophore resulted in a significantly improved percentage of live spermatozoa with true acrosome reaction at all stages of incubation, both in the bovine and the caprine.

Transcervical embryo collection in Boer goats.

Two experiments were conducted, aimed at improving the practicability of the method for transcervical embryo collection in Boer goats described by Pereira et al. [Pereira, R.J.T.A., Sohnrey, B., Holtz, W., 1998. J. Anim. Sci. 76, 360-363]. Invention of a hammock-like restraining device, use of a wider-bore flushing catheter and a modified flushing mode contributed toward this end. The importance of a luteolytic prostaglandin F(2alpha)-treatment [Pereira et al., 1998] was confirmed. In Experiment 1, administration of PGF(2alpha) 8h before does are flushed, increased the recovery rate from 43 to 79% (P<0.05). Advancing the PG F(2alpha)-treatment to 24h before flushing was instrumental in further enhancing embryo recovery rate. The amount of time required for flushing was reduced by about 20min (P<0.05) and the number of embryos recovered from the first 10 out of 30 flushes amounted to more than 80%, compared to 50% (P<0.05) when treating 8h before flushing. Administration of 1IU of oxytocin at the onset of flushing did not have any significant effect. When applying the findings of this investigation, the time required for flushing may be reduced from about 4h [Pereira et al., 1998] to less than 45min per doe and the required number of person involved decreased from four to two persons.

Myelin basic protein induces cell death of mature pig oligodendrocytes in vitro and produces demyelination in vivo.

Two methods prevail at present in producing demyelinated areas in the central nervous system. One uses the detergent-like effect of lysolecithin, the other is based on a cell killing effect of ethidium bromide plus x-irradiation. Unwanted side-effects are inherent in both methods. Based on the fact that myelin basic protein (MBP) kills adult pig oligodendrocytes but almost no astrocytes in vitro, we have used MBP for creating demyelinated areas in the centrum semiovale of the pig brain. These lesions are characterized by a loss of oligodendrocytes and myelin, a preservation of axons and astrocytes, and by the presence of macrophages. Thus, this type of lesion might present an alternative option for studying the fate of transplanted myelinating cells.

Effect of insemination of estrus-induced prepuberal gilts on ensuing reproductive performance and body weight.

Prepuberal gilts reared and managed to 85-90 kg live weight in a common system were allocated at random to one of three first-mating treatments in an experiment conducted over a period of more than 5 years. In two of the treatments, gilts received a single i.m. injection of 400 IU equine chorionic gonadotropin (eCG) and 200 IU human chorionic gonadotropin (hCG) (PG600; Intervet) and were either inseminated 4 and 5 days later on a fixed-time basis regardless of oestrus (treatment A), or at the second oestrus following treatment (treatment B). The third group of gilts remained untreated and was inseminated on the first spontaneous oestrus (treatment C). Thereafter, all gilts were managed in the same way and those observed in oestrus were re-inseminated. Significantly more gilts returned to oestrus after the first service in treatment A (35%) than in treatment B and C (12 and 17%, respectively; P<0.01). Gilts farrowed to the first or repeat inseminations at a significantly younger age (P<0.01) in treatment A (304 days) than treatment B (324 days) and C (320 days). The age difference at farrowing remained in surviving gilts at the end of their third parity. The first farrowing performance of the gilts was significantly affected by treatment in terms of litter size at birth (A 7.0, B 8.4 and C 8.3 live piglets per gilt; P<0.01), litter size at weaning (A 6.2, B 7.2 and C 7.2 live piglets per gilt, P<0.05), and piglet birth weight (A 1.4, B 1.3 and C 1.3 kg; P<0.05) but piglet survival rate and weaning weight were not affected by treatment. The live weights of the gilts were significantly different between the treatments at first insemination (A 95.7, B 106.5 and C 109.2 kg; P<0.01) but not when the first litter was weaned (A 133.6, B 135.1 and C 136.6; P>0.05). After the first farrowing there were no differences between the treatments in terms of the survival rate, productive or reproductive performance of the gilts/sows and their offspring. Without conducting a detailed cost-benefit-calculation it was deduced that, from an economical point of view, differences between treatment A and treatments B and C are negligible because the savings associated with farrowing at a younger age on this treatment just about compensated for any additional costs associated with the treatment and the lower number of piglets born at the first farrowing.

Development of a sensitive enzymeimmunoassay (EIA) for FSH determination in bovine plasma.

A highly sensitive enzymeimmunoassay (EIA) procedure for FSH determination in bovine plasma on microtiterplates using the biotin-streptavidin amplification system and the second antibody coating was developed. Biotin was coupled to FSH and used to bridge between streptavidin-peroxidase and the immobilized antiserum in the competitive assay. The EIA was carried out directly in 50 microl of bovine plasma and compared with an established radioimmunoassay (RIA) employing 100 microl plasma. Same FSH standards and FSH specific antiserum were used in both procedures. FSH standards prepared in hormone free plasma were used. The sensitivity of the EIA procedure was 6.25 pg/well FSH which corresponded to 125 pg/ml plasma; the 50% relative binding sensitivity was seen at 200 pg/well. In comparison to RIA, the EIA was at least four times more sensitive besides requiring 6 times less FSH specific antiserum. Plasma volumes for the EIA ranging from 12.5 to 50 microl did not influence the shape of the standard curve even though a slight drop in the OD450 was seen with higher plasma volumes. When both EIA and RIA methods were used to measure FSH in cows, the levels were detectable only by the EIA procedure. The assay detects high and low plasma FSH levels within the physiological variation as well as changes in plasma FSH after stimulation with a GnRH analog. In conclusion, in addition to being non-radioactive and low cost in nature, the method offers several advantages over the conventional FSH RIA procedure; these are (a) higher sensitivity, (b) less labour and time saving, (c) more economical use of precious FSH antiserum and (d) long shelf-life of the biotinyl-FSH label (in contrast to the short half life of iodinated FSH in RIA).

Intracytoplasmic injection (ICSI) of in vivo or in vitro matured oocytes with fresh ejaculated or frozen-thawed epididymal spermatozoa and additional calcium-ionophore activation in the pig.

In Experiment 1, we performed intracytoplasmic sperm injection (ICSI) of frozen-thawed epididymal and fresh ejaculated in vitro-capacitated spermatozoa into in vivo and in vitro-matured porcine oocytes. Within each group, oocytes were sperm-injected, sham-injected or served as handling controls. After subsequent in vitro-culture for 48 h the number of unchanged, fragmented und cleaved oocytes was recorded. The best result (14% cleaved after ICSI vs 2 and 0% with the sham injection and handling controls; P < 0.01) was achieved with fresh in vitro-capacitated spermatozoa injected into in vivo-matured oocytes. In vitro-matured oocytes displayed high fragmentation rates. In Experiment 2, in vitro matured oocytes were injected with freshly ejaculated in vitro-capacitated spermatozoa, followed by a 5 min-exposure to 0 (control), 50 or 100 microM calcium-ionophore. Comparable groups were sham injected or served as handling controls. It became apparent that Ca-ionophore treatment after injection of spermatozoa was ineffective at 100 microM, where at 50 microM a significant reduction in cleavage rate was observed (6 vs 26% with untreated controls, P < 0.01). Fluorescence staining with Hoechst 33342 revealed that in most cases of sperm-injected oocytes that remained unchanged after 48 h of in vitro-culture, sperm heads had not decondensed. Only few oocytes had continued to the pronucleus stage. In this context no favorable effect of Ca-ionophore was to be observed.

Transfer of pig embryos to different uterine sites.

Embryo transfer in pigs normally involves surgery. In connection with the development of nonsurgical or endoscopic transfer techniques, it is important to know whether the uterine site to which embryos are transferred has an effect on the success rate. In the present investigation, prepubertal donor gilts were treated with 1,500 IU of PMSG and, 72 h later, with 500 IU of hCG. Gilts were artificially inseminated 24 and 36 h after hCG injection. Embryos at the expanded blastocyst stage were collected from donor gilts. Recipient gilts were treated synchronous with the donors, using 1,000 IU of PMSG followed, 72 h later, with 500 IU of hCG. After a maximum of 3 h in vitro, embryos (n = 15 to 20, mean = 17.3) were transferred surgically to the middle of the uterine horn, to the caudal quarter of the uterine horn, or to the uterine body. Recipients were slaughtered between 28 and 34 d after transfer. The pregnancy rate of the recipients was low when the embryos were deposited in the uterine body (12%), compared with the middle (88%) or the caudal quarter of the uterine horn (81%) (P < .01). The corresponding average number of viable fetuses per pregnant recipient was 8.2 in the uterine body, 5.6 in the middle, and 4.5 in the caudal quarter. Average survival rate of embryos after transfer to the middle of the uterine horn was 41% vs 29 and 3% after transfer to the caudal quarter or the uterine body, respectively (P < .01). Hence, the uterine body seems to be an unsuitable site for embryo transfer in pigs. These results may explain the unsatisfactory results achieved with nonsurgical embryo transfer in the past.

Comparison of ultrasound-guided vs laparoscopic transvaginal ovum pick-up (OPU) in simmental heifers.

Transvaginal ovum pick-up (OPU) offers several advantages over standard embryo transfer procedures. For a systematic comparison of the ultrasound-guided (U-OPU) and the laparoscopic OPU (L-OPU) method, groups of Simmental heifers were subjected to 1 of the 2 OPU-methods for 8 wk (15 sessions) followed by a treatment-free interval of 11 wk and then another 8-wk period of OPU using the alternative method. Parameters that were evaluated included the number of follicles aspirated, number and morphology of recovered cumulus-oocyte complexes (COCs), and developmental capacity of oocytes after in vitro maturation (IVM) and fertilization (IVF). Blood samples were also taken from the donors twice a wk for progesterone measurement. To evaluate effects of long-term OPU on subsequent fertility of donors, all heifers (n = 14) were inseminated during the first natural estrus after the OPU experiment. The proportion of Class I oocytes was significantly (P < 0.001) higher after U-OPU than after L-OPU (38.7% vs 21.0%). Following IVM/IVF, this difference in oocyte quality was reflected by the cleavage rate (U-OPU: 58.1%; L-OPU: 52.1%; P < 0.05) and the rate of development to morulae and blastocysts (U-OPU: 27.1%; L-OPU: 13.9%; P < 0.001). Among other factors, the greater changes in vacuum pressure during L-OPU vs U-OPU might be responsible for the difference in oocyte quality. This problem may be overcome by a more flexible system for regulating the vacuum. Progesterone levels were higher during the L-OPU than the U-OPU periods. Seven donors (50%) were diagnosed pregnant by ultrasonography on Day 28 and by palpation per rectum on Day 42.

Regional influences of the fallopian tubes on the rate of boar sperm capacitation in surgically inseminated gilts.

Aliquots of ejaculated boar semen containing known numbers of spermatozoa were deposited into the caudal isthmus or rostral ampulla of the Fallopian tubes of gilts at, or immediately after, ovulation to assess regional influences on the rate of capacitation. Eggs were recovered during a second intervention 4, 5, 6 or 7 h after surgical insemination and were examined by phase-contrast microscopy. Results were obtained from ten animals in each of the 4-, 5- and 6-h groups and from eight animals in the 7-h group. With two exceptions, fertilized eggs were not recovered until 6 h after insemination into the isthmus, the proportion (45.6%) being significantly greater than the corresponding figure (1.4%) for ampullary insemination (P < 0.001). Similarly, the proportion of fertilized eggs recovered 7 h after insemination into the isthmus (58.7%) was significantly greater than after ampullary insemination (21.9%; P < 0.01). Numbers of spermatozoa associated with the zona pellucida remained low in all these instances, with mean figures per egg ranging from 0.3 to 3.8. Insemination into the isthmus gave a 1-2 h advantage in fertilization compared with insemination into the ampulla. Although relative rates of sperm cell progression to the site of fertilization may have contributed to this, there is strong evidence that rates of capacitation differ significantly in the respective portions of the Fallopian tube. Therefore, attention was focused on: (1) the viscous glycoprotein secretion in the caudal isthmus acting to remove seminal plasma from the sperm surface; and (2) the phase of sperm head binding to the isthmus epithelium. Gradients in local endocrine modulation by the adjacent ovary offer one explanation for the functional specialization of different regions of the Fallopian tubes.

Nonsurgical embryo collection in goats treated with prostaglandin F2alpha and oxytocin.

Embryos were collected transcervically from 38 unanesthetized goats in a standing position. Goats of the Boer breed were superovulated by injecting them with 16 mg of pFSH containing 40% LH. Collection took place 6 d after the last mating. A luteolytic dose of PGF2alpha was injected 16, 8, or 0 h before flushing the uterine lumen. One half of each group received an additional injection of oxytocin just before flushing. The flushing catheter was introduced transcervically, and 24 flushings were performed, with a 2-h pause between the first 12 and the last 12 flushings. Injection of PGF2alpha 16 or 8 h before transcervical embryo collection resulted in a significant increase (P < .001, chi-square test) in embryo recovery (-16 h: 91% with oxytocin, 85% without; -8 h: 91% with oxytocin, 80% without) compared with an injection at the time of collection (52% with oxytocin, 66% without). The recovery rate which was estimated as the percentage of embryos recovered relative to the number of corpora lutea counted endoscopically, was comparable to that achieved by surgical collection. There were no differences among groups with regard to embryo morphology. Embryos were transferred, and healthy kids were born. The nonsurgical collection of caprine embryos may be considered a viable alternative to conventional surgical procedures.
