Tyrosine replacement in P-selectin glycoprotein ligand-1 affects distinct kinetic and mechanical properties of bonds with P- and L-selectin.

Selectins are adhesion molecules that initiate tethering and rolling of leukocytes on the vessel wall. Rolling requires rapid formation and breakage of selectin-ligand bonds that must have mechanical strength to resist premature dissociation by the forces applied in shear flow. P- and L-selectin bind to the N-terminal region of P-selectin glycoprotein ligand-1 (PSGL-1), a mucin on leukocytes. To define determinants on PSGL-1 that contribute to the kinetic and mechanical properties of bonds with selectins, we compared rolling of transfected preB cells expressing P- or L-selectin on transfected cell monolayers expressing wild-type PSGL-1 or PSGL-1 constructs with substitutions in targeted N-terminal residues. Rolling through P- or L-selectin required a Thr or Ser at a specific position on PSGL-1, the attachment site for an essential O-glycan, but required only one of three nearby Tyr residues, which are sites for Tyr-SO(3) formation. The adhesive strengths and numbers of cells rolling through P- or L-selectin were similar on wild-type PSGL-1 and on each of the three PSGL-1 constructs containing only a single Tyr. However, the cells rolled more irregularly on the single-Tyr forms of PSGL-1. Analysis of the lifetimes of transient tethers on limiting densities of PSGL-1 revealed that L-selectin dissociated faster from single-Tyr than wild-type PSGL-1 at all shears examined. In sharp contrast, P-selectin dissociated faster from single-Tyr than wild-type PSGL-1 at higher shear but not at lower shear. Thus, tyrosine replacements in PSGL-1 affect distinct kinetic and mechanical properties of bonds with P- and L-selectin.

Binding of ovine uterine serpin to lymphocytes.

PROBLEM: The endometrium of the sheep produces a progesterone-induced member of the serpin superfamily of serine proteinase inhibitors that can inhibit lymphocyte proliferation and reduce natural killer cell activity. Present results indicate that this molecule, called ovine uterine serpin (OvUS), can bind specifically to lymphocytes. METHOD OF STUDY/RESULTS: Biotinylated OvUS bound to peripheral blood lymphocytes in a dose-dependent and saturable manner. Binding was inhibited by OvUS, but not by several other proteins, including serpin-enzyme complex (alpha 1-antitrypsin-trypsin). Heparin blocked binding when added to the binding reaction or when used to pretreat lymphocytes. Both lymphocytes and Madin-Darby bovine kidney (MDBK) cells also bound fluorescein isothiocyanate-labeled OvUS. CONCLUSIONS: Results suggest that OvUS can interact with lymphocytes and other cells through binding to a cell surface molecule. Such binding may indicate that inhibition of lymphocyte activation by OvUS involves 1) binding of OvUS to a cell surface receptor or 2) competitive inhibition of binding between OvUS and a co-activation molecule required for lymphocyte activation.

Papillomavirus capsid protein expression level depends on the match between codon usage and tRNA availability.

Translation of mRNA encoding the L1 and L2 capsid proteins of papillomavirus (PV) is restricted in vivo to differentiated epithelial cells, although transcription of the L1 and L2 late genes occurs more widely. The codon composition of PV late genes is quite different from that of most mammalian genes. To test the possibility that PV late gene codon composition determines the efficiency of PV late gene expression in some cell types, synthetic bovine papillomavirus type 1 (BPV1) late genes were constructed with codon composition modified to resemble the typical mammalian gene. Expression of these genes from a strong promoter in Cos-1 cells was compared with expression of wild-type BPV1 late genes from the same promoter. Both unmodified and modified PV late genes were transcribed in Cos-1 cells, but only the codon-modified genes were translated. In vitro translation of wild-type but not synthetic BPV1 L1 mRNA was markedly enhanced by addition of aminoacyl-tRNAs. Codon composition thus limits BPV1 late gene translation in Cos-1 cells, and this limitation can be overcome by modification of the codon composition of the genes or by provision of excess tRNA. Replacement of codons in the green fluorescent protein (gfp) gene with those frequently used in PV late genes did not alter gfp transcription in Cos-1 cells but almost abolished translation, supporting the hypothesis that the observed differences in efficiency of translation of modified and unmodified PV capsid genes were related to codon usage rather than mRNA structure. As tRNA populations vary within and between tissues in the same eukaryotic organism, we speculate that matching of tRNA availability to codon usage may be one determinant of the restriction of expression of PV late genes to differentiated epithelium.

Sequence close to the N-terminus of L2 protein is displayed on the surface of bovine papillomavirus type 1 virions.

The bovine papillomavirus type 1 (BPV1) L2 protein purified from Escherichia coli was used as an antigen to produce monoclonal antibodies (MAbs). A total of 26 individual clones which recognized the BPV1 L2 protein were obtained. Using infectious BPV1 virus particles, 3 of the MAbs were found to interact with BPV1 virus particles. Binding of the MAbs to BPV1 was confirmed by immunoelectron microscopy. A set of 92 13-mer peptides overlapping by 8 amino acids spanning the entire BPV1 L2 protein was synthesized on a membrane and used to map the epitopes recognized by these antibodies. Seventeen linear epitopes were identified. Our results revealed that a sequence toward the N-terminus of the L2 protein (aa 61-123) is displayed on the virus surface, while the remaining L2 sequences are hidden inside the virus capsid. Although the polyclonal antisera raised against BPV1 L2 neutralized the BPV1 virus, we failed to detect any neutralizing activity for the 3 L2-specific monoclonal antibodies which bound to the BPV1 particles. This suggests that extra binding sites may be needed for neutralization. This study prompted us to propose a model about how L1 and L2 proteins may interact during infectious papillomavirus assembly.

Increased expression of cell surface markers on endometrial gamma delta T-cell receptor+ intraepithelial lymphocytes induced by the local presence of the sheep conceptus.

PROBLEM: In sheep, gamma delta T-cell receptor (TCR)+ cells are a major lymphocyte subpopulation in the luminal epithelium of the uterine endometrium. During late pregnancy, this population of T cells increases in number and becomes more granulated. This study was performed to determine whether this apparent activation was induced by local effects of the conceptus or systemic effects of pregnancy. METHODS: The unilaterally-pregnant ewe, in which the conceptus is surgically confined to one uterine horn, was used to distinguish between systemic and local effects of pregnancy on function of endometrial gamma delta TCR+ intraepithelial lymphocytes. Lymphocytes collected from peripheral blood, and from the endometrial luminal epithelium of cyclic and unilaterally-pregnant ewes (day 140 of gestation) were analyzed by flow cytometry. RESULTS: As compared to gamma delta TCR+ lymphocytes in peripheral blood, gamma delta TCR+ intraepithelial lymphocytes from non-pregnant ewes had lower percentage of cells staining positive for CD25, CD44, and L-selectin. There was no effect of pregnancy on the percentage of gamma delta TCR+ peripheral blood lymphocytes staining positive for CD25, CD44, CD29, or L-selectin. Similarly, the percentage of intraepithelial lymphocytes staining positive for these antigens was similar for cells collected from cyclic ewes and cells from the nonpregnant uterine horn of unilaterally-pregnant ewes. In contrast, there was a higher percentage of CD25, CD44, CD29, and L-selectin+ cells for gamma delta TCR+ intraepithelial lymphocytes from the conceptus-containing uterine horn of pregnant ewes than for gamma delta TCR+ intraepithelial lymphocytes from the endometrium of cyclic ewes or from the nonpregnant uterine horn of pregnant ewes. CONCLUSION: The local presence of the conceptus causes changes in cell surface marker expression on endometrial gamma delta TCR+ intraepithelial lymphocytes during pregnancy. These changes may reflect conceptus-induced activation of this population of cells.

Mechanisms of the human intestinal H+-coupled oligopeptide transporter hPEPT1.

The hPEPT1 cDNA cloned from human intestine (Liang, R., Fei, Y.-J., Prasad, P. D., Ramamoorthy, S., Han, H., Yang-Feng, T. L., Hediger, M. A., Ganapathy, V., and Leibach, F. H. (1995) J. Biol. Chem. 270, 6456-6463) encodes a H+/oligopeptide cotransporter. Using two-microelectrode voltage-clamp in Xenopus oocytes expressing hPEPT1, we have investigated the transport mechanisms of hPEPT1 with regard to voltage dependence, steady-state kinetics, and transient charge movements. The currents evoked by 20 mM glycyl-sarcosine (Gly-Sar) at pH 5.0 were dependent upon membrane potential (Vm) between -150 mV and +50 mV. Gly-Sar-evoked currents increased hyperbolically with increasing extracellular [H+], with Hill coefficient approximately 1, and the apparent affinity constant (K0.5H) for H+ was in the range of 0.05 1 microM. K0.5 for Gly-Sar (K0.5GS) was dependent upon Vm and pH; at -50 mV, K0.5H was minimal (approximately 0.7 mM) at pH 6.0. Following step-changes in Vm, in the absence of Gly-Sar, hPEPT1 exhibited H+-dependent transient currents with characteristics similar to those of Na+-coupled transporters. These charge movements (which relaxed with time constants of 2-10 ms) were fitted to Boltzmann relations with maximal charge (Qmax) of up to 12 nC; the apparent valence was determined to be approximately 1. Qmax is an index of the level of transporter expression which for hPEPT1 was in the order of 1011/oocyte. In general our data are consistent with an ordered, simultaneous transport model for hPEPT1 in which H+ binds first.

Effects of endometrial serpin-like proteins on immune responses in sheep.

PROBLEM: The uterine milk proteins (UTMP) are a pair of related glycoproteins that are the major secretory products of the endometrium of the pregnant ewe. UTMP are members of the serpin superfamily of serine protease inhibitors but have no known antiprotease activity. One possible role for UTMP is to inhibit uterine immune responses--UTMP inhibit mitogen and mixed lymphocyte-induced proliferation of peripheral blood lymphocytes, natural killer (NK) cell activity and abortion caused by NK cell activation. Present objectives were to further evaluate the lymphocyte-inhibitory activity of UTMP and test whether UTMP modify immune responses in vivo. METHOD: One experiment demonstrated that UTMP inhibited antigen-induced lymphocyte proliferation induced by Candida albicans extract. In another experiment, ewes were immunized with OVA mixed with 3.75 mg/ml of UTMP or ovine serum albumin (OSA control). Injections of 1 mg OVA+UTMP or OSA in incomplete adjuvant were administered 6 wk later. Titers of antibody to OVA were lower (P < 0.001) for ewes administered UTMP than for ewes administered OSA. Effects of UTMP on delayed hypersensitivity reactions were evaluated in three experiments using skin-fold thickness assays. RESULTS: UTMP did not inhibit the increase in skin-fold thickness caused by PHA and Mycobacterium tuberculosis but rather tended to increase the response to PHA. CONCLUSION: Results strengthen the thesis that UTMP are physiologically relevant immunoregulatory molecules. Nonetheless, effects on skin-fold responses indicate that actions of UTMP can be more complex than would be predicted based on the proteins only having a single biological effect.

Progesterone-induced secretion of dipeptidyl peptidase-IV (cluster differentiation antigen-26) by the uterine endometrium of the ewe and cow that costimulates lymphocyte proliferation.

Dipeptidyl peptidase-IV (DPPIV) is a serine proteinase widely distributed in mammalian tissues, including lymphocytes, where it is identical to the T-cell activation antigen, cluster differentiation antigen-26. In the present study, DPPIV enzymatic activity was found in uterine secretions of unilaterally pregnant ewes in increasing amounts as gestation progressed. Progesterone increased DPPIV in uterine secretions from ovariectomized ewes and cows. DPPIV was enriched from ovine uterine secretions by a combination of cation exchange, gel filtration, lectin, and Gly-Pro-NH2 affinity chromatographies. The mol wt was 107 kilodaltons, as indicated by sodium dodecyl sulfate-polyacrylamide gel electrophoresis under reducing conditions, and 140 kilodaltons by gel filtration. The pI was estimated to be 6.8. Enzymatic activity had a pH optimum of 8.3 and a Km of 0.32 mM. The sequence of the 34 N-terminal amino acids was 77-87% homologous to an internal region of human cluster differentiation antigen-26 and rat liver DPPIV. Thus, uterine DPPIV appears to be missing the signal sequence that allows integration into the cytoplasmic membrane. DPPIV was localized immunohistochemically to lumenal and glandular endometrial epithelial cells and, in some pregnant ewes, discrete endometrial stromal cells. Highly enriched sheep uterine DPPIV costimulated proliferation of mitogen-treated sheep lymphocytes. Stimulation occurred in the presence of the uterine milk proteins, a pair of progesterone-induced endometrial secretory proteins with well characterized lymphocyte inhibitory activity. However, uterine milk proteins did not inhibit the costimulatory effect of DPPIV on phytohemagglutinin-L stimulated-lymphocyte proliferation. In conclusion, the uterine endometrium synthesizes a biologically active form of DPPIV under the influence of progesterone that is capable of enhancing mitogen-stimulated T-lymphocyte proliferation.

[Effects of arsenic on the offspring development in mice].

Effects of arsenic exposure on offspring development were studied in pregnant mice. The results showed arsenic contents of the body and brain tissue increased and the structure of neural cells in cerebral cortex became abnormal after exposure. The offspring neurobehavioral development appeared retardant and the proportion of their peripheral lymphocytes with alpha-naphthalene acetate enzyme (ANAE) declined in a mice group with exposure to 0.75 mg/kg arsenic, and the offspring body weight gain slowed after weaning, blood cholinesterase activity and serum level of haemolysin declined significantly in a group with 4.50 mg/kg. It indicated arsenic could affect embryonic and offspring development in mice through pregnant exposure.

Effect of the progesterone-induced serpin-like proteins of the sheep endometrium on natural-killer cell activity in sheep and mice.

Natural killer (NK)-like cells have been implicated as an important component of the endometrial lymphoid cell population and have been suggested to play roles in resistance to viral infection, in abortion, and in fetal growth. Inhibition of endometrial NK cell activity may be an important aspect of successful pregnancy because of the potential for actions directed against the conceptus. The aim of the study reported here was to test the hypothesis that the uterine milk proteins (UTM-proteins) inhibit activity of NK cells. Physiological concentrations of enriched preparations of UTM-proteins, which are a pair of related proteins that represent the major lymphocyte-inhibitory molecules in uterine fluid of pregnant and progesterone-treated ewes, were found to inhibit cytotoxic activity of cultured sheep and mouse NK cells. Furthermore, injection of the proteins into mice reduced activity of basal and poly(I).poly(C)-induced NK activity in spleen and prevented a slight increase in fetal loss caused by injection of poly(I).poly(C). These results further implicate the UTM-proteins as important regulators of uterine immune function.

The effects of Chinese herbs on improving survival and inhibiting anti-ds DNA antibody production in lupus mice.

Systemic lupus erythematosus (SLE) is an important autoimmune disease with multiple organ system involvement. From preliminary studies, we have found that six Chinese herbs: Atractylodes ovata, Anqelica sinensis, Cordyceps sinensis, Liqustrum lucidum, Codonopsis pilosula and Homo sapiens can improve defective in vitro interleukin-2 (IL-2) production in patients with SLE. In order to investigate the in vivo effects of these herbs, we used NZB/NZW F1 mice, a typical lupus animal model used to test these herbs. It was found that C. pilosula, H. sapiens and C. sinensis could prolong the life span of female NZB/NZW F1 mice and inhibited anti-ds DNA production. Although A. sinensis could prolong the life span of experimental mice, it did not inhibit the production of anti-ds DNA antibody. These herbs may have great potential for the management of human SLE in the future.

[Pregnancy induced hypertension complicated with cerebrovascular accidents].

Fourteen patients suffering from pregnancy induced hypertension (PIH) complicated with cerebrovascular accidents were admitted for treatment from 1977-1990. These were 8 cases of cerebral hemorrhage, 4 cases of cerebral infarction and 2 cases of cerebral arteriovenous malformation with intracerebral hematomas. These accounted for 0.34% of all hospitalized PIH cases during the same period and three died. The mortality rate was 0.72%. The etiology, pathology, brain CT scan features, clinical manifestations and treatment of these accidents were discussed.

Alteration of amino acid in plasma and cerebrospinal fluid of children with seizure disorders.

Amino acid levels in plasma were measured by amino acid autoanalyser in 130 convulsive children. The levels of taurine, serine and tryptophan were significantly lower in convulsive children as compared to normal control; in contrast, isoleucine, homocystine, GABA, histidine, arginine and ammonia were higher. The children with paroxysmal disorders (headache, dizziness and abdominal epilepsy) had the highest levels of isoleucine, histidine and arginine and the lowest levels of glutamate and cystein. Clinical seizure activity within 6 months prior to the test seemed to have no obvious effect on the plasma amino acid pattern, except for the levels of glycine and arginine tended to return to normal, and the level of GABA was significantly increased in patients with the seizure being controlled. The patients treated with carbamazepin as a single anticonvulsant had the highest GABA level compared to those with other anticonvulsants. Hyperglycinemia and hyperammonaemia were also noted in patients who took valproic acid. The levels of serine, isoleucine and phenylalanine in the CSF within 6 hours after convulsion were significantly lower than the normal control; while asparagine, tyrosine, lysine and arginine were significantly higher. The concentration of ammonia in the CSF was also elevated after convulsion as compared to the normal control. Amino acids play an important role in the generation of epilepsy and recently there has been an increasing number of studies to help determine their effects during an epileptic attack. However, there still is much debate and controversy on this topic. Therefore, further studies are needed and researchers should carefully consider factors that might affect the accurate assessment of the results.

[Anti-lipid peroxidation and protective effects of phenytoin sodium on ischemic myocardium of mice].

Isoproterenol (Iso, 20 mg.kg-1 x d-1 x 2 d) induced widespread and severe myocardial damages at ultrastructural level, decreased the myocardial Se-glutathione peroxidase (Se-GSH-Px) and superoxide dismutase (SOD) activities, increased the serum creatine phosphokinase (CPK) concentration and myocardial malondialdehyde (MDA) content. Phenytoin sodium (Phe) 15 or 30 mg.kg-1 ip pretreatment diminished the CPK release and MDA production, protected the Se-GSH-Px activity in the Iso-induced damage of mouse heart. The pretreatment with 30 mg.kg-1 abated the reduction of SOD activity. However, Phe 15 mg.kg-1 did not show such an effect. Phe (15 or 30 mg.kg-1) reduced the ultrastructural cardiotoxicity of Iso, and the membrane structure of ischemic myocardium was protected. The protective effects of verapamil pretreatment 3 mg.kg-1 ip were weaker than those of Phe on ultrastructural changes, but biochemical changes were similar to those of Phe. The results suggested that Phe possessed anti-lipid peroxidation and protective effects on ischemic myocardium.

[Observation on ultrastructure of sterile cysts of Echinococcus granulosus in mice].

The fine structure of sterile cysts of Echinococcus granulosus in mice was studied by TEM. The cyst wall consists of laminated layer and germinal membrane. At ultrastructural level, the laminated layer contains microfibrillate matrix and irregularly shaped electron-dense granules. The germinal membrane comprise of tegument and cell region. Plasmalemma invagination resembling the "pinocytotic vesicle" are seen between the microtriches of the tegument, and mitochondria are seen in the basal portion of the tegument. In the cell region, there are tegumental cells, muscle cells, and glycogen-containing cells. "Nucleolar channel system" can be observed in some of the glycogen-containing cells.

The significance of immunosuppression and hormonal alteration in neurotraumatology.

Thirty head trauma patients receiving no steroid treatment were investigated for a period of 20 days. The patients were divided into three groups according to their Glasgow Coma Scale (GCS) scores: GCS = 4-8 and GCS = 9-13. We studied their immunological and hormonal status with special reference to the relationship among severity of trauma, immunosuppression and hormonal alteration. The immunity tests included lymphocyte transformation, lymphocyte phenotyping and delayed hypersensitivity (DH) skin tests. The hormone study consisted of serum cortisol and ACTH level determinations. The following results were obtained: (1) The entire group suffered from significant suppression in lymphocyte responses to mitogens PHA and Con A on day 1 and day 10 after injury, but later they improved; (2) There was a significant fall (P less than 0.05) in OKT3 OKT4 and active T-cell on day 10, especially in patients with GCS less than or equal to 8; (3) A significant increase in OKT8 and decrease in OKT4 was concurrently noted causing an inversion in the ratio of OKT4 to OKT8 on the 10th day (patients 0.96:1, normal 1.6:1, P less than 0.01); (4) No significant change was observed in the number of B-cells, suggesting that cell-mediated immunity may be affected earlier and more severely than antibody producing capacity; (5) Delayed hypersensitivity skin test showed a high incidence of anergy in the severe (GCS less than or equal to 8) and fetal cases. A high mortality in anergic cases was also noted; (6) Elevated ACTH and cortisol levels detected upon arrival rapidly declined to normal value at day 5. In addition, we found no correlation between the curve of serum ACTH and cortisol alteration and the biphasic suppression curve of lymphocyte transformation in later study period. These findings suggest that the severity of natural resistance impairment seems to be related to the gravity of clinical manifestations and to the serum hormonal alteration only in the initial stages. Also, we find that the development of anergy may be an index of poor prognosis.

[Brain damage in asphyxiated newborn infants: early diagnosis. A follow-up study].

Cranial computerized tomographic (CT) scan and serum creatine phosphokinase-BB(CPK-BB) determination were done in 85 asphyxiated newborn infants, in some of them, long-term follow-up study including neurodevelopmental assessment and CT scan were also carried out. The types of early brain damage after perinatal asphyxia included intraparenchymal hemorrhage (IPH), periventricular-intraventricular hemorrhage (PV-IVH), subarachnoid hemorrhage (SAH) and brain edema (BE). Serum CPK-BB levels were elevated within 24 hours after the events of asphyxia, and correlated to the severity of brain damage. These findings were helpful in the early diagnosis of brain damage and estimating its severity and prognosis. The types and severity of brain damage were obviously related to prognosis. The prognosis of infants after SAH and BE were better than those of infants with PV-IVH, and infants with IPH showed the poor results. Follow-up CT scans were performed in 30 infants with brain damage. Abnormal changes included ventricular sulcus dilation, leukomalacia, cyst formation (pseudocyst) and porencephalic cavitation (porencephaly).
