Biochemical properties of CO2-dependent streptococci.

A total of 153 clinical isolates and 10 reference strains were employed in an investigation of CO2-dependent streptococci. Their selection was based on a lactic acid homofermentative end product. This group of organisms grew best in 5 to 10% CO2, and several species, including Streptococcus mutans, S. intermedius, Streptococcus MG, S. anginosus, and S. constellatus, required increased CO2 for primary recovery. A basal medium of thioglycolate with 0.1% Tween 80 and phenol red was prepared and used with selected carbohydrates. This media provided luxuriant growth. Serological testing failed to give any definitive correlation with species identification. A shortened differentiation scheme, combined from previous studies, was proposed.

Recurrence of group B streptococcal infection.

It is clear that the group B streptococcus has become a major pathogen of young infants within the comparatively recent past. Further it is clear that, as with other endemic and epidemic pathogens, increasing clinical and laboratory experience brings to light variations not initially evident. In addition, therapy presumably effective in initial cases may not continue to be so. The following papers by coincidence were received over a relatively brief period of time. Hence, for emphasis of some of the problems related to this organism, they are presented as a group.

The effect of polymethylmethacrylate and antibiotic combinations on bacterial viability. An in vitro and preliminary in vivo study.

Polymethylmethacrylate itself has a slight, long-term bacteriostatic effect on the growth of staphylococci, but while polymerizing the heat of polymerization and possibly the elaboration of monomer can be bactericidal to many pathogenic organisms. Twelve antibiotics incorporated into polymethylmethacrylate cement retained their antibacterial activity for long periods in dry storage, but the antibiotics leached out of polymethylmethacrylate quite rapidly in vitro and in vivo. No antibiotic could be found after thirty-seven days in treated cement pellets placed in a rabbit thigh.

In vitro correlates of delayed hypersensitivity in man: ambiguity of polymorphonuclear neutrophils as indicator cells in leukocyte migration test.

Delayed cutaneous hypersensitivity (DCH) of 12 normal adult subjects to purified protein derivative (PPD) of Mycobacterium tuberculosis, streptococcal streptokinase-streptodornase (SK-SD), and Candida albicans Dermatophytin O (DO) was assayed in vivo by skin testing and compared with such in vitro correlates of cellular immunity as lymphocyte transformation (LT) and inhibition of leukocyte migration (ILM) from microcapillary tubes or in agarose gel. LT was shown to be the best in vitro correlate of specific lymphocyte sensitization with all antigens. In the ILM assays, PPD showed good correlation with in vivo DCH and in vitro LT; SK-SD showed partial correlation; DO showed no correlation, not being active in any of the ILM tests. Cell distribution and morphology of stained migration patterns, ILM tests performed on separated populations of lymphocytes and polymorphonuclear leukocytes (PMN), as well as the ability of test antigens to stimulate PMN cells to reduce nitroblue-tetrazolium dye, indicated that in ILM tests mononuclear cells were not inhibited in their migration, whereas migration of PMN cells appeared to depend on their direct reaction with the test antigens.

Auxotypes and antibiotic susceptibilities of Neisseria gonorrhoeae from women with acute salpingitis. Comparison with gonococci causing uncomplicated genital tract infections in women.

An investigation of the MICs of various antibiotics and the nutritional requirements (auxotypes) of Neisseria gonorrhoeae recovered from the cervix, fallopian tubes, and peritoneal cavity of women with acute salpingitis was done. These MICs and auxotypes were compared to those of gonococci isolated from the cervix of women with uncomplicated or asymptomatic genital tract gonorrhea. The MICs of minocycline, penicillin G, ampicillin, cefoxitin, and cefaclor for gonococci isolated from women with acute salpingitis were significantly higher. Significant differences in auxotype patterns were identified between isolates from salpingitis cases and uncomplicated cases. The prototrophic form was the most common (75%) among salpingitis strains. No strains requiring arginine, hypoxanthine, and uracil were identified among the salpingitis isolates. In contrast, strains that required these nutrients were the most frequent auxotype among isolated from women with uncomplicated genital tract gonorrhea. Discriminant analysis revealed that the MIC of minocycline and the auxotype were the most powerful discriminators between groups of patients with different manifestations of gonorrhea.
