Following single, high dose (3 mg/kg) DDP therapy seven patients with head and neck cancer developed significant serum electrolyte disturbances. Hypocalcemia, hypomagnesemia, hypokalemia, and hypophosphatemia were the most clinically significant electrolyte abnormalities. Renal clearance studies support a pathologic mechanism of a diffuse renal tubular leakage not associated with renal failure.
The rate of recurrent duodenal ulcer following selective proximal vagotomy (SPV) has been assessed in two different groups of patients. Completion of vagotomy was left to the surgeons judgement in the first 50 consecutive cases, while in the following 50 cases a vagomotor electrostimulation device ("Vagorec") was used intraoperatively for control. The failure rate of 14% in the first group could be reduced to 2% in the second one. We believe that technique and success of SPV can be checked easily using this device. It is an educational remedy for the learning surgeon in order to avoid an incomplete SPV as well as it is a necessary tool for the experienced one.
The parents of a child with the clinical symptoms of Ehlers-Danlos syndrome type VI were identified as third-degree cousins. Biochemical analysis of the dermis of the patient revealed a complete lack of hydroxylysine in the dermal collagen. The dermis of both parents contained only half the amount of hydroxylysine found in healthy individuals. Hydroxylation of prolyl residues was normal in the skin of the patient and his parents. Investigation of the collagen synthesized by fibroblasts derived from the skin of the patient showed a normal proportion of type I and type III collagen. However, while hydroxylation of prolyl residues was normal in type I and type III collagen, hydroxylation of lysyl residues was markedly lower than normal in both type I and type III collagen.

A variety of Sepharose derivatives containing DL-O-phosphorylserine or adenosine nucleotides with different points of attachment, has been synthesized and tested for affinity to phosphoglycerate kinase. The most effective gels contained periodate-oxidized ATP or ADP bound via the ribose by hydrazone formation to adipoyl-dihydrazo-Sepharose. The effect of pH, magnesium and buffer ions on the binding capacity of the ATP derivative of Sepharose has been examined. Optimal elution of phosphoglycerate kinase was investigated using different combinations of adenosine nucleotides, 3-phosphogylcerate and magnesium ions. A method is presented giving conditions for the purification of phosphoglycerate kinase from different sources (spinach, human erythrocytes, human, rabbit and trout muscle). It includes extract preparation, affinity chromatography and gel filtration. The method is greatly superior to known isolation procedures by virtue of its technical simplicity, excellent yield (85-100%) and reproducability. The capacity of the ATP-ribosyl-adipoyl-dihydrazo-Sepharose was 5 mg phosphoglycerate kinase per 1 g of matrix. Polyacrylamide gel electrophoresis in the presence of sodium dodecylsulfate indicated that the final products are homogeneous. The phosphoglycerate kinases from different sources appear to have the same affinity for this ATP derivative of Sepharose, the same molecular weight and the same specific activity.
Liposome formulations were studied to achieve an efficient entrapment procedure for the production of liposomes of 99mTc-pentetic acid. The entrapment efficiency was studied by separation of the product using column chromatography. The particle-size range of the prepared liposomes was evaluated using electron microscopy. Entrapment techniques and separation procedures led to a liposome preparation with particles in the colloidal size range (0.001-0.5 micron). Dramatic differences in the organ distribution of the liposome preparation in mice were produced when different particle-size ranges were injected. Liposomes eluted in the first fraction after the void volume led to a maximum uptake by the liver and spleen 10 min after intravenous injection. Liposomes from pooled fractions provided less than half of the activity in the liver, as did the narrow size range liposome preparation.
