The immotile cilia syndrome: phase contrast light microscopy, scanning and transmission electron microscopy.

In the immotile cilia syndrome, transmission electron microscopy of the cilia shows abnormalities in the arrangement of the central pairs of tubules and in the dynein arms of the peripheral tubules, or in the radial spokes, We studied four nonrelated children, 9/12, 5, 6, and 6 years old, with situs inversus and a history of chronic sinusitis and bronchitis (Kartagener's syndrome) and four children in the same age group and with the same history, but without situs inversus. Under the phase contrast microscope no motile cilia were seen in the four patients with Kartagener's syndrome and in two of the four other children. Transmission electron microscopy showed aberrations in the cilia (absence of dyneim arms, random orientation of central tubules) in the patients with Kartagener's syndrome. Scanning electron microscopy revealed differences in morphology and arrangement of cilia between patients and controls. In the patients much more mucus was present on the mucosal surface. Furthermore, the cilia were in a state of disorder, with a multidirectional orientation instead of the parallel orientation seen in controls.

[Heavy chain disease: humoral and cellular findings in six patients with mu chain disease (author's transl)].

Clinical and chemical findings in 6 patients with mu-chain disease are reported. In all of these patients a lymphoproliferative disease was observed which by its symptoms resembled CLL. This similarity was underlined by the finding of an impaired response of PBL of the patients to mitogens as PHA, PWM, and Con A. A substantial result of our studies lies in the fact that the presence of mu-chains in the serum does not correlate with a clinically defined entity. Moreover, mu-chains have been observed in a case of benign lymphadenopathy. In one case a good remission of 4 years duration could be induced by the application of cytostatic agents.

A cytophotometric and flow cytofluorometric approach to the differentiation of T lymphocytes in the thymus.

By cytophotometric and flow cytofluorometric DNA and protein determinations two main proliferating subpopulations of thymus lymphocytes with a different percentage of cells in the S phase could be distinguished. One subpopulation had a very low protein content, was cortisone sensitive and located in the cortex. Cells with comparable low protein contents were not found amongst lymphocytes of the peripheral blood. The other lymphocyte subpopulation had a higher protein content, was cortisone resistant and situated in the cortex around a group of epithelial cells and in the medulla. The protein content of these thymus lymphocytes appeared to be comparable to that of the peripheral blood lymphocytes. On the basis of the protein content per cell, it is possible to identify and isolate the more often described major subpopulation of cortisone sensitive thymus lymphocytes remaining and dying in the thymus, and the minor cortisone resistant subpopulation of thymus lymphocytes which is the source of the peripheral T lymphocyte.

An improved method in rabbit intubation and thymectomy.

A procedure is developed for endotracheal intubation followed by positive pressure ventilation anaesthesia, allowing major thoracic surgery, including thymectomy, to be performed in the young and adult rabbit. A method is described to bring the animal in a position favourable for thymectomy. A detailed description is given of a thymus ablation technique in which the thoracic wall is left intact.

Early host cell-Molluscum contagiosum virus interactions.

Basal and suprabasal layers of human epidermis infected with the poxvirus Molluscum contagiosum have been examined with the technique of serial sectioning. Phagocytic vacuoles, formerly not observed in human epidermis, were found exclusively in the basal region. They did not fuse with other virus-containing vacuoles or with lysosomes to form digestive vacuoles. Various stages of uncoating, preceding ejection of the virus core into the cytoplasm, were observed in the virus-containing vacuole. Clusters of cores were commonly found close to or even associated with centriolar structures. Their possible interference with mitosis is discussed in relation to alterations observed in the plasma membrane. It is assumed that excision of gap junction elements precedes the induction of mitosis.

An improved method of tracheal intubation in the rabbit.

A rabbit to be intubated was anesthetized and placed in a supine position with its head back at a 45 degree angle. A speculum made of spring steel and cord was used to hold the mouth open. The tongue was held to one side, an infant size laryngoscope was inserted in the direction of the larynx, and an uncuffed nasal/oral tube was introduced into the mouth. The tube was then passed into the larynx during inspiration and held in place in the mouth and a specially designed bit.
