Protection of crayfish, Cambarus clarkii, from white spot syndrome virus by polyclonal antibodies against a viral envelope fusion protein.

White spot syndrome virus (WSSV) is a large double-stranded DNA virus, causing considerable mortality in penaeid shrimp and other crustaceans. WSSV produces five major structural proteins, including two major envelope proteins, VP28 and VP19. To produce VP28 and VP19 as a single protein for antibody production, DNA sequences encoding both open reading frames were fused together and cloned into pET-22b(+) expression vector. The fusion protein, VP(19+28), was expressed in Escherichia coli, purified using Ni2+ His affinity chromatography and injected into a rabbit. Antiserum collected from the immunized rabbit was tested in vivo for ability to protect crayfish, Cambarus clarkii, from disease caused by WSSV. Fifteen days after challenge with WSSV, treatment with VP(19+28) antiserum gave 100% protection against disease in the ambient temperature range of 15-22 degrees C and 65% protection at a constant temperature of 26 degrees C. These results demonstrated VP(19+28) antiserum is effective in protection of crayfish from WSSV and confirmed that VP19 and VP28 play an important role in WSSV host infection. Targeting both VP19 and VP28 may be effective for the design of both immunotherapeutic medicines and reagents to detect WSSV.

A novel artificial prosthetic replacement for the proximal interphalangeal joint of the hand--from concept to prototype.

This article describes the development of a proximal interphalangeal (PIP) joint prosthesis based on the principles of replicating anatomical surface components, the use of macrolocking intramedullary stem and the use of a cobalt-chrome alloy material. The design features are intended to obtain an optimal range of motion while retaining stability and longevity. The final prototype, for which a patent has been filed, is described.

Tuberculosis post-liver transplantation: a rare but complicated disease.

INTRODUCTION: Tuberculosis is a rare but serious complication after transplantation. We report a case and discuss its presentation and management. CLINICAL PICTURE: A 60-year-old Indonesian male presented initially with fever, acute confusion and rapidly progressive right upper lobe pneumonia 3.5 months post-liver transplant, and was diagnosed with pulmonary tuberculosis by positive sputum smear for acid-fast bacilli and tuberculosis culture. TREATMENT/OUTCOME: Standard anti-tuberculosis therapy was administered but was complicated by interaction with cyclosporine and drug-induced cholestasis. CONCLUSION: A high level of suspicion, prompt antituberculosis treatment and close follow-up are essential in management of post-transplant tuberculosis.

Performance of an intensive pond system treating municipal wastewater in a cold region.

A full-scale intensive pond system (IPS) with shorter HRT was designed, constructed and operated in Jining, Inner-Mongolia for the treatment of municipal wastewater, which is a mixed domestic and industrial wastewater characterized by quite high SS and lower BOD5/COD ratio values or lower biodegradability. Therefore, the pond system was designed as an integrated intensive pond system (IIPS) consisting of settling/anaerobic pond (SAP), intensified anaerobic pond (IAP), facultative pond (FP), and polishing ponds (PPs). In order to improve the performance of the IPS, some intensified measures were made, including inlet and outlet even-distribution systems of each unit pond, package of biofilm carrier in IAP for the increase and even distribution of biomass; overflow waterfalls on the dikes between unit ponds for the increase of DO in pond water, gravel filtration dike (or dam) for removing suspended solids including algae, which have improved the performance of the IPS remarkably in terms of removal of main pollutants, such as SS, COD, BOD5, TN, NH3-N, TP and total bacteria. The final effluent from the IPS in warm seasons from May to October were SS 7.2-10.8 mg/L, BOD5 8.5-19.6 mg/L, COD 44.1 - 76.5 mg/L, and NH3-N 1.5-10.2 mg/L, which well meet Chinese national discharge standard (2nd class) of secondary municipal wastewater treatment plants, i.e. BOD5 and SS 30 mg/L respectively, COD 100 mg/L, and NH3-N 25 mg/L.

Modelling and simulation of the intervertebral movements of the lumbar spine using an inverse kinematic algorithm.

An inverse kinematic model is presented that was employed to determine the optimum intervertebral joint configuration for a given forward-bending posture of the human trunk. The lumbar spine was modelled as an open-end, kinematic chain of five links that represented the five vertebrae (L 1-L5). An optimisation equation with physiological constraints was employed to determine the intervertebral joint configuration. Intervertebral movements were measured from sagittal X-ray films of 22 subjects. The mean difference between the X-ray measurements of intervertebral rotations in the sagittal plane and the values predicted by the kinematic model was less than 1.6 degrees. Pearson product-moment correlation R was used to measure the relationship between the measured and predicted values. The R-values were found to be high, ranging from 0.83 to 0.97, for prediction of intervertebral rotation, but poor for intervertebral translation (R= 0.08-0.67). It is concluded that the inverse kinematic model will be clinically useful for predicting intervertebral rotation when X-ray or invasive measurements are undesirable. It will also be useful to biomechanical modelling, which requires accurate kinematic information as model input data.

Interleukin-7 and suppression of local peritoneal immunity in ovarian carcinoma.

OBJECTIVES: To investigate the epithelial ovarian carcinoma (EOC) secretion of interleukin-7 (IL-7). METHODS: Levels of IL-7 were assayed by enzyme-linked immunoadsorbent assay and IL-7 mRNA, and protein expression in tissues and cell lines were detected by RT-PCR and immunohistochemistry. RESULTS: The median serum IL-7 level in patients with EOC (32 cases; 32.49 pg/ml) was significantly higher than that of patients with benign tumors (16 cases; 7.59 pg/ml) and healthy women (16 cases; 10.64 pg/ml) (P<0.05). The median peritoneal fluid IL-7 level in patients with EOC (17.39 pg/ml) was slightly higher than that of patients with benign tumors (14.09 pg/ml), but not significantly so (P>0.05). There were positive correlations between the serum and peritoneal fluid IL-7 levels in both ovarian cancer and benign group (P<0.05, both). Only two EOC specimens expressed IL-7 mRNA, and no IL-7 protein positive was found in any specimens. CONCLUSIONS: Epithelial ovarian carcinoma cells rarely express IL-7, and IL-7 levels are decreased in the ascitic fluid of patients with EOC.

Voltage- and Ca2+-dependent burst generation in neuroendocrine Dahlgren cells in the teleost Platichthys flesus.

The neuroendocrine Type 1 Dahlgren cells of the caudal neurosecretory system of the flounder display characteristic bursting activity, which may increase secretion efficiency. The firing activity pattern in these cells was voltage-dependent; when progressively depolarized, cells moved from silent (approximately -70 mV), through bursting and phasic to tonic firing (< -65 mV). Brief (10 s) evoked bursts of spikes were followed by a slow after-depolarization (ADP; amplitude up to 10 mV, duration 10-200 s), which was also voltage-dependent and could trigger a prolonged burst. The ADP was significantly reduced in the absence of external Ca(2+) ions or the presence of the L-type Ca(2+) channel blocker, nifedipine. BayK 8644 (which increases L-type channel open times) significantly increased ADP duration, whereas the Ca(2+)-activated nonselective cation channel blocker, flufenamic acid, had no effect. Pharmacological blockade of Ca(2+)-activated K(+) channels, using apamin and charybdotoxin, increased the duration of both ADP and evoked bursts. However, action potential waveform was unaffected by either apamin/charybdotoxin, nifedipine, BayK 8644 or removal of external Ca(2+). The short duration (approximately 100 ms), hyperpolarization-activated, postspike depolarizing afterpotentials (DAP), were significantly reduced by nifedipine. We propose that long duration ADPs underlie bursts and that short duration DAPs play a role in modulation of spike frequency.

Synthesis and characterization of human alpha-defensins 4-6.

Human alpha-defensins are small, Cys-rich, cationic proteins expressed predominantly in neutrophils and intestinal epithelia. They play important roles in innate and adaptive immunity against infection. Progress in studying these molecules can be accelerated by access to large quantities of high-quality materials, which have been obtained mainly from natural sources. Here, we report total synthesis of human alpha-defensins 4, 5, and 6, also known as HNP4, HD5, and HD6, using the optimized N,N-diisopropylethylamine (DIEA) in situ neutralization/2-(1 H-benzotriazolyl)-1,1,3,3-tetramethyluroniumhexafluorophosphate (HBTU) activation protocol for solid-phase Boc chemistry. Oxidative folding/disulfide formation was achieved directly using crude peptides, resulting in an overall synthetic yield of 10-16% with high purity. Antimicrobial activity assays were performed with Escherichia coli ATCC 25922 and Staphylococcus aureus ATCC 29213, using colony-counting methods, and the results demonstrated differential activity against these strains. Our report describes a highly efficient synthetic approach that enables thorough structural and functional studies of these three important immunologic molecules.

Field performance of VAQTA (inactivated, purified hepatitis a vaccine) in Chinese children in Jiangsu.

In Jiangsu, 30% of children between the ages of 5 and 8 years test seropositive for hepatitis A. The safety, tolerability, and immunogenicity of a 2-dose regimen (0, 6 months) of VAQTA (0.5 ml of 25U) administered IM in 50 healthy children aged 5 to 8 years without prior serological screening was evaluated. Blood samples were collected prior to the first dose and after each additional dose of VAQTA to determine the initial anti-HAV serostatus and response rates to the vaccine. Twelve children (24%) were initially seropositive and 38 (76%) were initially seronegative. Four weeks after the primary dose of VAQTA, 34 of the 38 subjects (89.5%, 95% CI 75 to 97) were anti-HAV seropositive. The geometric mean titer was 33.1 mIU/ml (95% CI 22.4 to 49.0). After the booster dose at 6 months, all the subjects were seropositive (37/37), giving a seroconversion of 100% (95% CI: 90, 100). The geometric mean titer was 7585.8 mIU/ml (95% CI: 5623.4 to 10,471.3). Adverse experiences were generally mild and transient. Results of this study are consistent with results from a previous double-blind randomized trial of this vaccine and confirm that VAQTA is highly immunogenic, and generally well-tolerated.

Plasma wakefield acceleration in self-ionized gas or plasmas.

Tunnel ionizing neutral gas with the self-field of a charged particle beam is explored as a possible way of creating plasma sources for a plasma wakefield accelerator [Bruhwiler et al., Phys. Plasmas (to be published)]. The optimal gas density for maximizing the plasma wakefield without preionized plasma is studied using the PIC simulation code OSIRIS [R. Hemker et al., in Proceeding of the Fifth IEEE Particle Accelerator Conference (IEEE, 1999), pp. 3672-3674]. To obtain wakefields comparable to the optimal preionized case, the gas density needs to be seven times higher than the plasma density in a typical preionized case. A physical explanation is given.

Plasma-wakefield acceleration of an intense positron beam.

Plasma wakefields are both excited and probed by propagating an intense 28.5 GeV positron beam through a 1.4 m long lithium plasma. The main body of the beam loses energy in exciting this wakefield while positrons in the back of the same beam can be accelerated by the same wakefield as it changes sign. The scaling of energy loss with plasma density as well as the energy gain seen at the highest plasma density is in excellent agreement with simulations.

A comparison of quinacrine sterilization (QS) and surgical sterilization (TL) in 600 women in Guizhou Province, China.

OBJECTIVES: Compare the safety and efficacy of quinacrine sterilization (QS) and surgical sterilization, also known as tubal ligation (TL). METHODS: 300 women accepted QS in Guiyang, China during the period from July 1995 to September 1997. Each patient was scheduled for follow-up at 3, 6, 12 and 24 months. In March 1998, a comparison group of 300 women electing TL during the same time period was systematically chosen. Researchers visited the village of every woman and conducted a structured interview. Each candidate was given a general health and pelvic exam at a clinic in her village. All interviews and exams were completed in August 1998. RESULTS: Of the 289 QS patients interviewed (a follow-up rate of 96.3%), 265 had had 2 insertions. There were 3 pregnancy failures for a cumulative life table failure rate of 1.2 per 100 women at 24 months. The 299 TL patients (a follow-up rate of 99.7%) had a similar rate of 0.7. There were no life-threatening side effects or deaths in either groups. QS was less disruptive, more easily tolerated, required fewer resources and was viewed more favorably than TL by women and their spouses. CONCLUSIONS: Both methods were found safe and very effective. However, QS was considered to be more acceptable than TL.

From pro defensins to defensins: synthesis and characterization of human neutrophil pro alpha-defensin-1 and its mature domain.

Human neutrophil alpha-defensins (HNPs) are small, cationic, Cys-rich antimicrobial proteins that play important roles in innate immunity against infectious microbes such as bacteria, fungi and enveloped viruses. Synthesized as inactive precursors in vivo (pre-proHNPs), HNPs are activated through proteolytic removal of the inhibitory pro-peptide required for subcellular sorting and correct folding. We seek to understand the molecular basis for the recognition between the 45-residue pro-peptide and the C-terminal functional domain. Here we described, total chemical synthesis of the 75-residue human neutrophil pro alpha-defensin-1 (proHNP1) via native chemical ligation. After oxidative folding, proHNP1 is cleaved by cyanogen bromide at the Met45-Ala46 peptide bond to release the mature form. The native disulfide connectivity in HNP1, i.e. Cys1-Cys6, Cys2-Cys4 and Cys3-Cys5, is verified by mass mapping of peptide fragments generated by proteolytic digestion and Edman degradation. Fluorescence spectroscopy studies and antimicrobial activity assays further support that synthetic proHNP1 and HNP1 are correctly folded. While largely unstructured in aqueous solution, the pro-peptide binds to HNP1 intermolecularly with an apparent Kd value of 6.2 microM at pH 7.4, confirming the mode of intramolecular inactivation of human alpha-defensin precursors.

Electrical activity of caudal neurosecretory neurons in seawater- and freshwater-adapted flounder: responses to cholinergic agonists.

The caudal neurosecretory system (CNSS) of the euryhaline flounder is involved in osmoregulatory responses underlying adaptation to seawater and freshwater. This study compared electrophysiological activity and responses to cholinergic agonists in the neuroendocrine Dahlgren cells in an in vitro preparation taken from fully seawater- (SWA) or freshwater-adapted (FWA) fish. Resting membrane and action potential parameters showed few differences between SWA and FWA cells. The hyperpolarisation-activated sag potential and depolarising afterpotential were present under both conditions; however, amplitude of the latter was significantly greater in SWA cells. The proportions of cells within the population exhibiting different firing patterns were similar in both adaptation states. However, bursting parameters were more variable in FWA cells, suggesting that bursting activity was less robust. The muscarinic agonist, oxotremorine, was largely inhibitory in Dahlgren cells, but increased activity in a non-Dahlgren cell population, alpha neurons. Nicotine promoted bursting activity in SWA Dahlgren cells, whereas it inhibited over half of FWA cells.

Superior mesenteric arterial embolism: a retrospective study of local thrombolytic treatment with urokinase in West China.

Embolism of the superior mesenteric artery (SMA) is a rare disorder. To explore the selective criteria of local thrombolysis, evaluate its efficacy and discuss the therapeutic protocol of SMA embolism, a retrospective case control study was undertaken. Sixteen cases were divided into two groups: nine cases (group A) from May 1995 to April 1999 were given traditional treatment, while seven cases (group B) from November 1999 to June 2002 received comprehensive therapy including local thrombolysis. The local thrombolytic treatment was performed without procedure-related complications and the embolus was successfully lysed in four patients. The time between admission and diagnosis (or treatment) was shorter in group B than in group A (2.3 +/- 1.2 vs 12.3 +/- 9.2 hr, p = 0.013). Gangrenous bowel segments in group A were much longer than in group B (159.4 +/- 87.7 vs 45.7 +/- 61.6 cm, p = 0.009). However, mortality between the two groups was the same (p = 0.282, OR = 0.32, 95% CI = 0.039, 2.618) perhaps because of the small sample size. Angiography could shorten the duration from the onset of SMA embolism to therapy and certainly lowers the risk of mortality. Local fibrinolytic infusion may be an effective alternative to embolectomy in patients with SMA embolism but without intestinal infarction.

Detection of Fusarium head blight resistance QTL in a wheat population using bulked segregant analysis.

A population of 218 recombinant inbred lines (RILs) was developed from the cross of two wheat (Triticum aestivum L.) cultivars, 'Ning 894037' and 'Alondra'. Ning 894037 has resistance to Fusarium head blight (FHB) and Alondra is moderately susceptible. Response of the RILs and their parental lines to FHB infection was evaluated with point inoculation in four experiments both in greenhouse and in field conditions. Distribution of disease severity in the population is continuous, indicating quantitative inheritance of resistance to FHB. Bulked segregant analysis and QTL mapping based on simple sequence repeat (SSR) markers revealed three chromosome regions that are responsible for FHB resistance. A chromosome region on 3BS accounted for 42.5% of the phenotypic variation for FHB resistance. Additional QTLs were located on chromosomes 2D and 6B. These three QTLs jointly accounted for 51.6% of the phenotypic variation. SSR markers linked to the QTLs influencing resistance to FHB have potential for use in breeding programs.

Inactivation and dissociation of rice ribulose-1,5-bisphosphate carboxylase/oxygenase during denaturation by sodium dodecyl sulfate.

Ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco) is a key enzyme in photosynthesis and photorespiration. The inactivation and subsequent conformational changes and dissociation of rice Rubisco by SDS have been studied. At low SDS concentrations (0.4 mM), Rubisco completely lost its carboxylase activity and most of its sulfhydryl groups became exposed. Dissociation of small subunits and significant conformational changes occurred at higher SDS concentrations. Increasing SDS concentrations caused only slight changes in CD spectrum, indicating no significant effect of SDS on the secondary structure of the enzyme. The results prove that the active site of Rubisco is more fragile to denaturants than the protein as a whole. The results also suggest that small subunits are more liable to SDS denaturation and thus dissociate first, while the more hydrophobic large subunits remain complexed. The naturally existing hydrophobic surface of Rubisco may be an important factor in the interaction of Rubisco with other macromolecules.

Microglial activation and recruitment, but not proliferation, suffice to mediate neurodegeneration.

Microglial activation occurs during excitotoxin-induced neurodegeneration. We have reported that microglia can exhibit neurotoxic behaviors after injection of excitotoxins into the hippocampus. It is not known, however, whether microglial proliferation, which is part of the activation response, is required for neurodegeneration to be observed, or whether activation of the pre-existing resident microglia suffices. Using osteopetrotic (op/op) mice, in which injury-induced microglial proliferation does not take place, we demonstrate that only the microglia initially residing in the CNS are adequate to promote neurodegeneration. Our data suggest that there is a threshold at which a maximal microglial contribution to neurotoxicity is observed. This threshold appears to be sufficiently low, such that activation of just 40% of the microglia present in wild-type mice serves to trigger neurodegeneration. Furthermore, since the decrease in microglial numbers coincides with a decrease in tissue plasminogen activator's activity, we suggest that tissue plasminogen activator can be used as a marker for microglial proliferation.
