Tumor suppression after tumor cell-targeted tumor necrosis factor alpha gene transfer.

The tumor necrosis factor alpha (TNF-alpha) gene was introduced by retroviral gene transfer into the TNF-alpha-insensitive tumor cell line J558L. Production of 40 pg/ml TNF-alpha by clone J2T12 consistently did not change the growth rate in vitro, but drastically suppressed tumor growth when injected into syngeneic BALB/c mice. Within 2 wk, 90% of the mice inoculated with J558L cells developed a tumor, but none of the mice injected with J2T12 did so. Within the observation period (greater than 3 mo), 60% of the mice inoculated with J2T12 did not develop a tumor. In the other 40% of the mice, tumor manifestation was significantly delayed. Mice injected simultaneously with J2T12 cells and an anti-TNF-alpha monoclonal antibody developed tumors similar to parental J558L cells. Similarly, the tumor-suppressive effects of TNF-alpha were abolished, e.g., by injection of an anti-type 3 complement receptor (CR3) monoclonal antibody that is known to prevent migration of inflammatory cells. These results and the observation of tumor-infiltrating macrophages suggest that lack of tumorigenicity of J2T12 cells is due to the TNF-alpha secretion by the tumor cells and that TNF-alpha acts indirectly by a mechanism that involves chemotactic recruitment and activation of cells, predominantly of macrophages. In contrast, the tumor growth was not affected when, instead of TNF-alpha, interleukin 6 was expressed by J558L cells. Together, our results support the concept of tumor cell-targeted cytokine gene transfer as a tool for cancer treatment, and particularly demonstrate that extremely low doses of TNF-alpha produced by tumor cells are sufficient to inhibit tumor growth without detectable side effects.

Major histocompatibility complex class II hyperexpression on B cells in interleukin 4-transgenic mice does not lead to B cell proliferation and hypergammaglobulinemia.

The murine interleukin 4 (IL4)-encoding cDNA expressed under the control of the immunoglobulin heavy chain enhancer/promoter was inserted into the mouse germ line. All B cells present in the IL4 transgenic mice show a marked increase in expression of class II histocompatibility (Ia) antigens compared to B cells from control mice. In vivo 5-bromo-2'-deoxyuridine incorporation and cell cycle analysis revealed no significant increase in B cell proliferation in spleens of IL4 transgenic mice compared to littermate controls. Likewise, serum immunoglobulin concentrations in IL4 transgenic mice were not increased. From these experiments we conclude that Ia hyperexpression on B cells per se does not induce activation of autoreactive T cells leading to B cell proliferation or hypergammaglobulinemia.

Generation of long-lived B cells in germ-free mice.

In the present study we have addressed the question of whether the composition of the peripheral B cell pool is similar in mice raised under conventional, specific pathogen-free or germ-free breeding conditions. For this purpose, the representation of various B lineage subsets was quantitated by flow cytometry and the lifespan of B cells analyzed by labeling dividing cells with the thymidine analogue 5-bromo-2'-deoxyuridine in vivo. The results indicate that except for an increased fraction of CD45R (B220)dull B lineage cells in the bone marrow and a marginally faster turnover of splenic B cells in germ-free compared to conventional mice, there is no obvious difference in the representation and turnover of B cells in the different kinds of mice. The data indicate that a stable pool of peripheral B cells is generated even in the absence of microbial stimulation.

The influence of head position on the intracranial pressure in preterm infants with posthemorrhagic hydrocephalus.

In order to measure the intracranial pressure (ICP) of 8 newborn with posthemorrhagic hydrocephalus (group A) we used the noninvasive technique via the anterior fontanel (Ladd monitor). The ICP was monitored in two different head positions (30 degrees elevated, horizontal). The median measurements were: elevated head position, 13 mm Hg, horizontal head position, 16 mm Hg. For comparative reasons, we also measured the ICP of 12 healthy newborns (group B). The median results were: elevated head position, 4 mm Hg, horizontal head position, 6.5 mm Hg. Significant differences were found between groups.

Cholinergic input uncouples Ca2+ changes from K+ conductance activation and amplifies intradendritic Ca2+ changes in hippocampal neurons.

Muscarinic synaptic activation is known to be involved in cortical arousal as well as learning. Although simple increases in the electrical responsiveness of neurons might be the basis of arousal, the linkage of muscarinic transmission to the synaptic plasticity that might underlie learning is lacking. Most models of synaptic plasticity involve postsynaptic Ca2+ changes as a trigger for subsequent processes. We imaged muscarinic effects on free Ca2+ accumulation during intracellular recordings from CA3 pyramidal neurons in the guinea pig hippocampal slice. Muscarinic activation, either by repetitive stimulation of cholinergic fibers or by bath-applied carbachol, strongly increased intradendritic Ca2+ accumulation during directly evoked repetitive firing, in part by blocking a Ca(2+)-dependent K+ conductance. The effects of repetitive stimulation of cholinergic fibers were enhanced by the acetylcholine-esterase blocker eserine and blocked by the muscarinic antagonist atropine. These findings demonstrate a novel muscarinic reinforcement of Ca2+ changes during excitation, which are probably significant for synapse modification.

[Time resolved laser fluorescence microscopy of hematoporphyrin derivative in vivo].

Laser-assisted fluorescence microscopy proved a powerful method for quantitative in vivo measurement of HpD fluorescence. Therefore the aim of the present work was to study the distribution of HpD in tumor vs. adjacent tumor-free tissue of 10 animals over a period of 8 days. We measured additionally the differences in HpD uptake in different tumor subcompartments. In tumor tissue HpD fluorescence showed maximum values 5-9 hours after i.v. application of the drug. The relation tumor-free tissue at that time was about 3-8. The third day of examination showed nearly identical values for both tissues. Between day 5 and day 8 we found an increase of HpD contents in tumor vs. normal tissue in 50% of the examined animals. Comparative measurements in the tumor centre and the tumor edge showed a preferential uptake of HpD in the tumor edge area. Probably the higher retention of HpD in this area is due to the oedema zone surrounding the tumor and to extravasation of HpD.

[The diagnostic assessment of enlarged lymph nodes by the qualitative and semiquantitative evaluation of lymph node perfusion with color-coded duplex sonography].

Perfusion of enlarged lymph nodes by colour-coded sonography was studied prospectively in 105 benign and 115 malignant lymph nodes. The diagnosis was confirmed histologically in 158 and clinically in 62. Spectral analysis in the benign lymph nodes provided normal values for pulsatility and resistance indices. A pulsatility index greater than = 1.8, or a resistance greater than = 0.9 indicate lymph node metastases with positive prediction of 93% and specificity of 97%. In addition, subjective, semiquantitative classification of lymph node perfusion in relation to the surrounding fat or connective tissue improves the diagnosis of lymph node metastases and of malignant lymphomas.

Typing of heat-stable soluble Haemophilus parasuis antigen by means of agargel precipitation and the dot-blot procedure.

According to Morozumi's and Nicolet's (1986) investigations, a serological classification procedure for H. parasuis and to a certain extent, for H. parasuis-like strains was proposed on the basis of heat-stable cell antigens in the immunodiffusion test. It was possible to classify 72.8% of the investigated strains serologically using this procedure. 7 of 12 serotypes were described for the first time. 60.1% of the classified strains belonged to the already known serotypes SV 1 to SV 5, whereas the new serotypes SV Jena 6 to SV Jena 12 amounted to only 12.7% of the field isolates. The serotypes SV Jena 7 to 9 are represented by H. parasuis-like strains. Unencapsulated strains and isolates of serotype SV 5 dominate in animals with Glasser's disease. The serotypes SV 1, SV 5, Jena 6 and SV Jena 10 proved to be highly virulent in SPF pigs, SV 2 and SV 4 were of medium virulence. The other serotypes were non-virulent. The high specificity in AGPT was not reproducible in the more sensitive dot-blot procedure. This must be taken into account, if the dot-blot is to be used for the classification of serotypes of H. parasuis. The results point to a participation of nonimmunogenic polysaccharides in the detection reaction.

Picrotoxin- and 4-aminopyridine-induced activity in hilar neurons in the guinea pig hippocampal slice.

1. Paired extra- and intracellular recording was used to study the activity of neurons in the dentate hilus and their interaction with CA3/CA4 pyramidal neurons and granule cells during picrotoxin- or 4-aminopyridine (4-AP)-induced rhythmical activity in the guinea pig hippocampal slice. 2. Picrotoxin induced synchronous repetitive population spikes in the CA3, CA4, and hilar region, but no extracellular activity in the granule cell layer. 4-AP induced rhythmically occurring positive field-potential waves in the CA3, CA4, and granular layer coincident to negative/positive field potentials in the hilus. 3. Picrotoxin-induced activity originated in the CA3 area and subsequently appeared in the CA4 and hilar region, whereas 4-AP-induced activity appeared simultaneously in all subfields. 4. Blockade of fast glutamatergic excitation by 6-cyano-7-nitroquinoxaline-2,3-dione (CNQX, 10 microM) blocked the picrotoxin-induced activity but not the 4-AP-induced activity. 5. Focal application of tetrodotoxin (TTX) between area CA3 and CA4 blocked picrotoxin-induced activity in the CA4 and hilar region but decoupled 4-AP-induced activity in the CA3 area. 6. Under intracellular recording, picrotoxin induced bursts in CA3, CA4, and hilar neurons but K-dependent slow IPSPs in granule cells. 4-AP induced rhythmically occurring burst in hilar neurons synchronous to Cl- and K-dependent IPSPs in CA3, CA4, and granule cells. 7. Comparison of picrotoxin- and 4-AP-induced rhythmical burst activity reveals that many hilar neurons are excited by CA3/CA4 pyramidal neurons in addition to the well-known excitation by granule cells and perforant path fibers, and that, in turn, many hilar neurons inhibit CA3, CA4, and granule cells.

Amplitudes of visually evoked cortical responses due to full-field stimulation and to stimulation with four half-fields.

In 30 healthy subjects the VECP following full-fields, upper, lower, nasal and temporal half-field stimulation was recorded. The full-field amplitudes is significantly larger than each of the half-field responses. For peak P120, the amplitudes following lower half-field stimulation is significantly larger than the amplitude due to upper, nasal and temporal half-field stimulation. The sum two half-field amplitude is significantly larger than the full-field response.

[Specific skin infiltration in Hodgkin's disease].

A 60 year old man consults the dermatologist because of a bean sized centrally ulcerated painless tumor in the skin of his left axilla. The axillary lymph nodes proximal to the skin tumor are firm and enlarged. The skin biopsy reveals a dense mixed infiltrate composed of lymphocytes, histiocytes, plasma cells and granulocytes and there are multiple neoplastic mononuclear and some multinuclear giant cells, resembling Hodgkin- and Reed-Sternberg-cells. Thus the diagnosis of Hodgkin's disease was considered on the grounds of the skin biopsy. Immunohistologically the atypical giant cells express the CD-30 (Ki-1)-antigen. Histology of the subsequently excised lymph nodes resembled the changes in the skin and showed classical Hodgkin's disease of mixed cellularity.

Demonstration of elastic fibres with reagents for detection of magnesium.

Investigation of elastic fibres in various human and animal tissues with the reagents quinalizarin, magneson II, and titan yellow for the detection of magnesium revealed striking positive results. After pretreatment of skin and ligamentum flavum with elastase the tests were negative. The results support the supposition that the amount of magnesium in elastic fibres is sufficient for histochemical detection. It is speculated that the marked chelate-forming property of magnesium, or its antagonistic function to calcium, is associated with the elastic property of the fibres.

Russel bodies-containing cells in the Harderian gland of the chicken in relation to different immune conditions.

The modulation of the number of Russel bodies-containing plasma cells in the Harderian gland under different experimentally induced immune conditions is reported. The findings support the conception of an immunologic function of the Harderian gland which is similar to the function of the thymus on the one hand, and to the function of the bursa of Fabricius on the other.

[Experimental studies on the effectiveness of intraligamental anesthesia].

In this study the effectiveness of the periodontal ligament injection was tested in rats. Anesthetic solutions with carbon dye were injected into the crestal periodontal ligament space under back-pressure. Vital microscopic observations showed that the solution spreads via an intraosseous path into the vessels of the pulp. These results support the contention that the periodontal ligament injection is intravascular in nature. Therefore, possible cardiovascular responses to periodontal ligament injections containing catecholamines must be considered.
