Assay and purity analysis of diatrizoate sodium in drug product by LC.

An LC procedure was developed to separate diatrizoate sodium from three known impurities. These impurities are 2,4- and 2,6-diiodo-3,5-diacetamidobenzoic acid (DDZA), and the free amine (5-acetamido-3-amino-2,4,6-triiodobenzoic acid). The separation was achieved using a Hamilton, PRP-X100, anion exchange column. The retention of diatrizoate sodium and the impurities was dependent on pH, potassium chloride concentration and phosphate concentration. Increasing any of these mobile phase modifiers decreased the retention time of all of the components. The eluent for assay and purity determination of drug product consisted of 0.1 M potassium chloride and 0.05 M potassium phosphate dibasic in water/acetonitrile (900:100). The mean concentration of diatrizoate sodium in Hypaque Sodium 50% determined over 3 days was 102.3% of label claim with an R.S.D. of 1.3. The accuracy of the purity method, determined by spiking known amounts of the impurities at five concentrations ranging from 0.025 to 0.06% (w/w) into drug product, was 100.1% for DDZA and 94.2% for the free amine. The decomposition of diatrizoate sodium in 0.1 N potassium hydroxide at 85 degrees C followed pseudo first-order kinetics. The calculated half-life was 2 days.

The effects of acidification on the geochemistry of Al, Cd, Pb and Hg in freshwater environments: a literature review.

Acidification of freshwater environments (terrestrial, surface water, and freshwater sediment) can significantly affect the geochemistry of Al, Cd, Pb and Hg; for example, metal mobility within soils (Al, Cd), the relative distribution of dissolved metal species (Al, Cd, Pb, Hg), and the sedimentation rate of metals in standing water bodies (Cd, Pb) can be altered by acidification. In this critical review of the literature over the last decade concerned with the interaction of acidification with these four metals, we have attempted both to provide an assessment of the current state of knowledge in this field and to emphasize those areas where significant progress has been made since the possible implications of environmental acidification on metal geochemistry became widely appreciated in the late 1970s. We have also indicated those areas which we feel are most in need of further research.

Screening for neonatal isohemolytic anemia in calves.

Anti-erythrocytic immunoglobulins in serum and colostrum of 124 anaplasmosis-vaccinated cows were detected with a saline agglutination test. Positive results were correlated with the occurrence of neonatal isohemolytic anemia (NIA) in calves and were used to predict the occurrence of NIA. The disease was prevented by withholding colostrum from calves with a high potential for NIA.

Comparisons of serotests with calf inoculations for detection of carriers in anaplasmosis- vaccinated cattle.

Fifteen months after discontinuing vaccination for anaplasmosis in a herd of 131 cattle, 126 of the animals were reactive to the complement-fixation or card-agglutination tests. Inoculation of splenectomized calves proved that 9 animals were carriers and 117 animals were free of anaplasmosis. Anaplasmosis vaccination induced titers detectable by the standard serotests that persisted for at least 15 months.
