Distribution and persistence of pyrethroids in runoff sediments.

Pyrethroids are commonly used insecticides in both agricultural and urban environments. Recent studies showed that surface runoff facilitated transport of pyrethroids to surface streams, probably by sediment movement. Sediment contamination by pyrethroids is of concern due to their wide-spectrum aquatic toxicity. In this study, we characterized the spatial distribution and persistence of bifenthrin [BF; (2-methyl(1,1'-biphenyl)-3-yl)methyl 3-(2-chloro-3,3,3-trifluoro-1-propenyl)-2,2-dimethylcyclopropanecarboxylate] and permethrin [PM; 3-(2,2-dichloroethenyl)-2,2-dimethylcyclopropanecarboxylic acid (3-phenoxyphenyl)methyl ester] in the sediment along a 260-m runoff path. Residues of BF and PM were significantly enriched in the eroded sediment, and the magnitude of enrichment was proportional to the downstream distance. At 145 m from the sedimentation pond, BF was enriched by >25 times, while PM isomers were enriched by >3.5 times. Pesticide enrichment along the runoff path coincided with enrichment of organic carbon and clay fractions in the sediment, as well as increases in adsorption coefficient K(d), suggesting that the runoff flow caused selective transport of organic matter and chemical-rich fine particles. Long persistence was observed for BF under both aerobic and anaerobic conditions, and the half-life ranged from 8 to 17 mo at 20 degrees C. The long persistence was probably caused by the strong pesticide adsorption to the solid phase. The significant enrichment, along with the prolonged persistence, suggests that movement of pyrethroids to the surface water may be caused predominantly by the chemically rich fine particles. It is therefore important to understand the fate of sediment-borne pyrethroids and devise mitigation strategies to reduce offsite movement of fine sediment.

Evaluation of Kd underestimation using solid phase microextraction.

Many important environmental pollutants are strongly adsorbing hydrophobic compounds. Because of their potential to adsorb to dissolved organic matter (DOM), their partition coefficient Kd may be underestimated by the conventional approach due to incomplete phase separation. In this study, solid-phase microextraction (SPME) was compared with liquid-liquid partition (LLP) for measuring Kd of bifenthrin and permethrin isomers on sediments. Due to its selective detection of the freely dissolved concentration, SPME gave Kd values 0.6-4.4-fold greater than those obtained by LLP in creek and field sediments and 3.6-21.7-fold greater in nursery runoff sediments. Underestimation by the conventional method was attributed to adsorption to DOM that was not excluded from the aqueous phase by centrifugation. The degree of underestimation was dependent on the source and amount of DOM and may be generally significant for compounds that have DOM adsorption coefficient (KDOM) > 10(4). This study provides evidence that the existing Kd values for many hydrophobic pollutants may be underestimated, and SPME may be a viable, efficacious tool for evaluating the underestimation.

[Study of delayed encephalopathy after acute carbon monoxide poisoning by clinical characteristic, CT and MRI features].

OBJECTIVE: To explore the diagnosis value of CT and MRI in delayed encephalopathy after acute carbon monoxide poisoning (DEACMP). METHODS: The clinical, CT and MRI data of 20 patients with DEACMP were analysed. RESULTS: The main clinical manifestations of the disease were intellectual disturbances and personality disorders. CT scan in DEACMP showed that some characteristic changes (such as diffuse low density) was observed within bilateral cerebral white matter, prominently within bilateral or unilateral globus pallidus or basal ganglia areas. The MRI showed that brain lesions in DEACMP were bilateral symmetric punctate, spotty or confluent foci within periventricular white matter and centrum semioval, which showed high signal intensity in T2-weighted and low signal intensity in T1-weighted. The chronic ischemia changes were found in the globus pallidus or basal ganglia areas. Cerebral cortex lesions were observed occasionally. CONCLUSIONS: The diagnosis of DEACMP mainly depends on case history, clinical features and neuroradiological presence. MRI is more valuable in the diagnosis and the differential diagnosis than CT.

Expression of HLA class I, beta(2)-microglobulin, TAP1 and IL-10 in Epstein-Barr virus-associated nasal NK/T-cell lymphoma: Implications for tumor immune escape mechanism.

Several mechanisms of immune escape might be in operation in Epstein-Barr virus (EBV)-associated nasal NK/T-cell lymphoma. We have previously shown the downregulation of the immunogenic EBV nuclear antigens by alternative promoter usage and the preferential selection of the deletion genotype of latent membrane protein 1 in nasal lymphoma. To understand further the strategies used for immune escape by this tumor, we examined by immunohistochemistry HLA class I expression in 15 cases using frozen sections, along with beta(2)-microglobulin and transporter associated with antigen processing 1 (TAP1) expression in 39 cases using paraffin sections. All nasal NK/T-cell lymphomas showed positive staining for HLA class I, beta(2)-microglobulin and TAP1 on most tumor cells, except for two cases (5%) in which most of the tumor cells lacked beta(2)-microglobulin staining. We next immunostained for interleukin-10 on frozen sections in 13 cases, all of which showed strong expression by most tumor cells. Transcription of human interleukin-10 but not EBV BCRF1 (viral interleukin-10) was identified by reverse transcriptase-polymerase chain reaction in these nasal NK/T-cell lymphomas. Overall, our data suggest that global downregulation of HLA class I or TAP1 rarely accounts for the ability of nasal NK/T-cell lymphoma to evade immunosurveillance and that other immune escape mechanisms may be operating in nasal NK/T-cell lymphoma, such as production of interleukin-10 to suppress the local immune response.

Adsorption of chloroacetanilide herbicides on soil (I). Structural influence of chloroacetanilide herbicide for their adsorption on soils and its components.

Adsorption of chloroacetanilide herbicide acetochlor, alachlor, metolachlor and propachlor on soils and soil components was determined, and the structural differences of these herbicides were used to explain the order of sorptivity. Adsorption isotherms for all herbicide-soil combinations conformed to the Freundlich equation, and Ki increased with increasing soil organic carbon content. Kd on soil humic acid was greater than that on clay, but association of humic acid with clay reduced the overall adsorption. On all soils and soil humic acids, herbicide adsorption decreased in the order: metolachlor > acetochlor > propachlor > alachlor. On Ca(2+)-montmorrilonite, the order changed to metolachlor > acetochlor > alachlor > propachlor. FT-IR spectra of herbicide-clay or herbicide-humic acid-clay mixtures showed that H-bonding and charge transfer were the primary interaction pathways between these compounds and the surface of clay or humic acids. The different moieties attached to 2-chloro-acetanilide and their unique arrangement may have influenced the binding mechanisms and thus the sorptivity of these herbicides. This study indicates that the structural difference of pesticides in the same classes may be used as a molecular probe to obtain a better understanding of sorption mechanisms of pesticides on soil.

Adsorption of acetanilide herbicides on soil and its components. II. Adsorption and catalytic hydrolysis of diethatyl-ethyl on saturated Na(+)-, K(+)-, Ca(2+)-, and Mg(2+)-montmorillonite.

Adsorption and catalytic hydrolysis of the herbicide diethatyl-ethyl [N-chloroacetyl-N-(2,6-diethylphenyl)glycine ethyl ester] on homoionic Na(+)-, K(+)-, Ca(2+)-, and Mg(2+)-montmorillonite clays were investigated in water solution. The Freundlich adsorption coefficient, Ki, got from isotherms on clay followed the order of Na+ approximately K+ > Mg2+ approximately Ca2+. Analysis of FT-IR spectra of diethatyl-ethyl adsorbed on clay suggests probable bonding at the carboxyl and amide carbonyl groups of the herbicide. The rate of herbicide hydrolysis in homoionic clay suspensions followed the same order as that for adsorption, indicating that adsorption may have preceded and thus caused hydrolysis. Preliminary product identification showed that hydrolysis occurred via nucleophilic substitution at the carboxyl carbon, causing the cleavage of the ester bond and formation of diethatyl and its dechlorinated derivative, and at the amide carbon, yielding an ethyl ester derivative and its acid. These pathways also suggest that hydrolysis of diethatyl-ethyl was catalyzed by adsorption on the clay surface.

Photodegradation of alpha-naphthaleneacetic acid in aqueous solution.

Kinetic processes of alpha-naphthaleneacetic acid (NAA) photolysis were studied under different conditions. The results showed that the ultraviolet light was more effective than fluorescent light in promoting degradation, and the degradation of NAA under ultraviolet light followed the first order kinetics with the photolysis rate constant of 1.15 x 10(-2) min-1 and half-life time (t1/2) of 60 min. Further, it was proved that the photolysis rate was higher in the presence of oxygen, titanium dioxide (TiO2), and low pH (acidic solution). At last, two photolysis intermediates were identified by GC-MS and possible photolysis pathways were proposed.

Effect of copper aspirinate on contraction of isolated rabbit aortic strips.

AIM: To investigate the effect of copper aspirinate on contraction of vascular smooth muscle. METHODS: Isolated rabbit aortic strips, including intact endothelium strips and endothelial cell-denuded aortic strips, were suspended in modified Krebs'solution to determine effects of copper aspirinate on the contraction induced by norepinephrine (NE), KCl, and CaCl2, while CuSO4, aspirin, and vehicle were used as controls. RESULTS: Copper aspirinate possessed antagonistic effect on contraction of rabbit aortic strips induced by NE with an IC50 value of 31 nmol/L, while CuSO4 had much l ess antagonistic effect with an IC50 value of 0.29 micromol/L, and aspirin did not work in the same preparation. No effect of copper aspirinate were found on the contraction induced by KCl and CaCl2. Effects on endothelial cell denuded aortic strips were similar to those in the normal aortic strips. CONCLUSION: Copper aspirinate possessed different effects from aspirin and CuSO4 on vascular smooth muscles. It inhibited contraction induced by NE with an activity stronger than CuSO4 at the same Cu2+ concentration, this action might be due to blockade of the receptor-operated Ca2+ channels, and it might not be linked to the endothelium.

Field amplified stacking injection--capillary electrophoresis for quantitative analysis of released methacrylic acid from dental composites.

Field-amplified stacking injection-capillary electrophoresis (FASI-CE) applied for the first time to the study of dental materials is demonstrated in this report. CE was used to separate methacrylic acid from the other degraded components. Due to the background electrolyte being weakly basic and methacrylic acid being negatively charged, in-capillary enrichment of the analyte was achieved by FASI. A detection limit of less than 50 ppb was obtained with good reproducibility (relative standard deviation < 3%). The method was applied to the analysis of four commercially available dental composites. Results showed that FASI-CE was effective in monitoring the fabrication process and product quality of restorative materials fabricated from methacrylate-based composites by analyzing the amount of methacrylic acid released from the residual methacrylate monomers.

[The role of NO/ET and the effect of electroacupuncture on injuried gastric mucosa in rats].

The aim of the present work is to observe changes in endothelium-derived factors--nitric oxide (NO) and endothelin (ET), with special reference to the regulative role of electroacupuncture (EA) in injured gastric mucosa of rat by ethanol. It was found that gastric mucosal blood flow (GMBF), transmucosal potential difference (PD) and the content of NO in the serum all decreased (P < 0.01), while the content of ET in the plasma and the lesion index (LI) of gastric mucosa increased (P < 0.01). After administration of L-Arg and SNP, the content of NO and GMBF increased significantly (P < 0.01), whereas the content of ET and LI decreased (P < 0.01). These effects were enhanced by EA at ZuSanLi (ST-36) points, which, however, were inhibited by NO biosynthetic inhibitor--L-NNA. The latter effect could be reversed by concurrent administration of L-Arg. The above results suggest that NO plays a critical part in protecting gastric mucosa against injury, which is also responsible for the protective effect of EA on injury of gastric mucosa.

Effects of copper-aspirin complex on platelet aggregation and thrombosis in rabbits and mice.

The effects of intragastric and intraduodenal copper-aspirin complex on rabbit platelet aggregation were observed by Born's method. Myers's method was used to evaluate the antithrombotic effect of copper-aspirin complex in mice. In-vitro copper-aspirin complex selectively inhibited arachidonic acid-induced platelet aggregation with an IC50 value (concentration resulting in 50% inhibition) of 13.2 microM (95% confidence limits 9.1-16.8 microM). Copper-aspirin complex (10 mg kg(-1) given intragastrically or intraduodenally) was more potent than aspirin in inhibiting arachidonic acid-induced platelet aggregation. Copper-aspirin complex (10 mg kg(-1)) had a stronger inhibitory effect and a longer duration of action when given intragastrically than when given intraduodenally. It was shown by radioimmunoassay that copper-aspirin complex significantly reduced the level of thromboxane B2 in plasma while markedly increasing that of 6-ketoprostaglandin F1alpha (6keto-PGF1alpha). Copper-aspirin complex (10 mg kg(-1) given intragastrically for 7 days) significantly reduced mouse mortality caused by intravenous injection of arachidonic acid. The results suggest that both in-vitro and in-vivo copper-aspirin complex is more potent in selectively inhibiting arachidonic acid-induced platelet aggregation than aspirin. When given intragastrically the complex has a more potent antiplatelet effect and a longer duration of action than when given intraduodenally. The antithrombotic effect of the complex was more potent than that of aspirin.

Inhibitory effects of copper-aspirin complex on platelet aggregation.

AIM: To study the inhibitory effects of copper-aspirin complex (CuAsp) on platelet aggregation. METHODS: With adenosine diphosphate the effects of CuAsp on platelet aggregation in vitro or in vivo were investigated. Radioimmunoassay and fluorophotometry were used to measure thromboxane B2 (TXB2) generation from platelets, the levels of TXB2 and of 6-keto-PGF1 alpha in plasma and the platelet serotonin release reaction. RESULTS: In vitro, CuAsp inhibited arachidonic acid (AA)-induced aggregation (IC50 = 17 mumol.L-1, 95% confidence limits: 9-33 mumol.L-1), the release of 5-HT (IC50 = 19 mumol.L-1, 95% confidence limits: 10-30 mumol.L-1), and TXB2 generation from platelets (P < 0.05). CuAsp 10 mg.kg-1 i.g. selectively inhibited AA-induced aggregation, and increased the 6-keto-PGF1 alpha concentration in plasma while decreased that of TXB2. CONCLUSION: CuAsp, in vitro or in vivo, shows more potent inhibitory effects on AA-induced aggregation than aspirin (Asp), related to the inhibition of platelet cyclooxygenase and the release of active substances from platelets.

Nasal T/NK cell lymphoma: a clinico pathologic study of 30 west Chinese patients with special reference to proliferation and apoptosis.

Midfacial T-cell lymphomas are more prevalent in Asia than in Europe or North America. Clinically, these lymphomas are noted as one major differential diagnosis in the malignant midline granuloma syndrome. During the past years, the group of nasal T/NK cell lymphomas has been recognized that is frequently associated with EBV-infection. The aim of the current publication was to describe the clinical presentation and course of 30 patients attending the West China University of Medical Sciences, Chengdu, P.R. China, between 1991 and 1994. Clinical records were assessed and the patients were followed for 6 to 29 (mean 12.4) months. Several microscopic features thought to be associated with this entity were carefully evaluated together with immunohistochemical data. The proliferation of the tumour cells was assessed by determining the mitotic index and the ratio of MIB-1 labelled cells. In addition, the incidence of apoptotic cells was investigated by means of the in-situ end labelling (ISEL) technique. Our data confirm the expression of T-cell markers by T/NK cell lymphomas as determined by the immunohistochemistry. The apoptotic index was found to correlate with the ratio of MIB-1 labelled cells. Expression of the bcl-2 oncoprotein was not associated with increased or diminished proliferation or cell death, respectively. Eight of the thirty patients succumbed to their disease during the follow-up period. Kaplan-Meier cumulative survivals and log-rank tests revealed a significant impact of MIB-1 labelling on mean survival times.

[The photolytical products of aqueous carboplatin solution].

Carboplatin, an abbreviation for 1, 1-cyclobutanedicarboxylatodiamine platinum (II), is the second generation platinum anticancer drug. The stability of its aqueous solution is of great importance to clinical effects. It has been known that the solution is relatively stable with t1/2 being about three months when kept in dark place, and unstable to light as shown by the rapid change in the UV spectrum. But, the photolytic products remain unidentified. As a knowledge of the products plays an important role in understanding the influence of photolysis of carboplatin on the clinical effects, we have recently studied the photolytical products of aqueous carboplatin solution upon 313 and 254 nm irradiation, and now report our results here.

[Epstein-Barr virus infection in midline malignant reticulosis].

An oligonucleotide probe directed against small RNAs encoded by EBV gene was used to detect EBV genome by in situ hybridization combined with immunophenotype analysis. The results showed that (1) in 18 of 19 cases (94.7%) atypical lymphoid cells (ALC) expressed T-cell differentiation antigen, 15 cases were CD3 positive, 9 cases were UCHL1 positive, 6 cases were both CD3 and UCHL1 positive, no ALC gave positive reaction to B-cell and histiocyte markers. (2) 15 of 19 cases gave positive reaction to EBER (small RNAs encoded by EBV gene) by in situ hybridization analysis and ALC were reactive to T-cell markers in all 15 cases but one. The results of this study suggest that EBV infection is also present in midline malignant reticulosis, which is characterized by active T-cell proliferation. The role and effect of EBV infection in pathogenesis of lymphoproliferative disorders was discussed.

Determination of diclofop-methyl and diclofop residues in soil and crops by gas chromatography.

A method is described for the determination of residues of the herbicide diclofop-methyl and its metabolite, diclofop, in soil and crops. The residues were extracted with acetone-light petroleum and extracts were concentrated (diclofop was derivatized to its pentafluorobenzyl derivative), and then the products were purified on a chromatographic column containing alumina, silver-alumina and Florisil. Finally, they were determined by gas chromatography using an electron-capture detector. The detection limits of diclofop-methyl and diclofop were between 0.01 and 0.05 mg/kg. The average recoveries were 76.4-97.2% and 72.8-105.2%, respectively, making the method suitable for statutory residue testing purposes.

[Pathomorphologic and immunohistochemical study of midline malignant reticulosis].

The histopathology, immunophenotype and clinical presentation in 35 cases of midline malignant reticulosis (MMR) were studied with 8 cases of midfacial non-specific inflammation as the control during immunohistochemical staining. The result showed that atypical lymphoid cells (ALC) of MMR could be divided into small, intermediate and large ALC subtypes basing on their sizes. Twenty-three of the 35 cases (65.7%) showed dominant small and medium-sized ALC proliferation, meanwhile, ALC infiltrating into the mucosa and vascular walls were also small and medium-sized cell predominant. Immunohistochemical study showed that the UCHL1 positive small lymphocytes proliferated predominantly in MMR; in contrast, many Ki-B5 positive lymphocytes appeared in midfacial inflammation. ALC of MMR in 27 cases (71%) also showed UCHL1 positive reaction but negative reaction to Ki-B5 and lysozyme. The result indicates that ALC of MMR belongs to T-lymphocyte origin and MMR may be a kind of peripheral T-cell lymphoma originated from mucosa-associated lymphoid tissue (MALT). The relations between MMR, malignant lymphoma and other related disorders of MALT are discussed.
