Nontoxic lipopolysaccharide from Rhodopseudomonas sphaeroides ATCC 17023.

Chemical analysis of the lipopolysaccharide from Rhodopseudomonas sphaeroides ATCC 17023, isolated by the phenol-chloroform-petroleum ether method, revealed the presence of glucuronic acid, 2-keto-3-deoxyoctonate, threonine, and phosphorus in the polysaccharide moiety. The lipid A component contained glucosamine, glucosamine phosphate, amide-bound 3-oxotetradecanoic acid and 3-hydroxytetradecanoic acid, and ester-bound 3-hydroxydecanoic acid and 7-tetradecenoic acid. Structural similarity of the lipid A from R. sphaeroides ATCC 17023 to enterobacterial lipid A is indicated by the existence of a serological cross-reaction occurring between the lipid A from R. sphaeroides ATCC 17023 and that from Salmonella minnesota R595. The lipopolysaccharide and lipid A of R. sphaeroides, however, were found to be neither toxic in mice nor pyrogenic in rabbits.

Cdc2 phosphorylation of nucleolin demarcates mitotic stages and Alzheimer's disease pathology.

Nucleolin is a major multifunctional nuclear phosphoprotein that is phosphorylated by Cdc2 kinase in mitosis and that participates in a number of cellular processes. The monoclonal antibody TG-3 generated against neurofibrillary tangles (NFT) found in Alzheimer's disease (AD) is highly specific for mitotic cells in culture. We here demonstrate that phosphorylation of nucleolin by Cdc2 kinase generates the TG-3 epitope. The unique pool of TG-3 immunoreactive nucleolin appears abruptly during the prophase. It is associated with chromosomes through the metaphase and it gradually disappears during separation of chromosomes and exit from mitosis. In the brain, nucleolin was localized not only to nuclei but also to neuronal cytoplasm, and it is a marker for early NFT. In patients with AD, Cdc2 phosphorylated nucleolin was present in NFT. These findings suggest that phosphorylation of nucleolin by Cdc2 kinase is a critical event and the point of convergence of two distinct pathways, mitosis and neurodegeneration.

Immunogenic properties of lipid A.

Lipid A, the endotoxic principle of gram-negative bacterial lipopolysaccharides, when suitably exposed on the surface of bacterial cells, becomes immunogenic, eliciting the formation of antibodies specific to lipid A. Antibodies to lipid A occur naturally in the serum of normal humans and many animal species; the two exceptions found so far are mice (all strains tested) and guinea pigs. Antibodies to lipid A occur more frequently in patients with infections caused by gram-negative bacteria, such as urinary tract infections. Antibodies to lipid A interacted with free lipid A but not with lipid A as present in the intact lipopolysaccharide molecule. Antibodies to lipid A were induced in rabbits, but not in mice, following a single injection of immunogenic lipid A. Because of similarities between the lipid A of many gram-negative bacteria, antibodies to lipid A showed wide cross-reactions. With regard to biologic activity, antibodies to lipid A have been variously shown to be protective, damaging, or without effect.
