Nucleotide sequence and transcriptional mapping of the major capsid protein gene of pseudorabies virus.

The gene encoding the 142-kDa major capsid protein (MCP142) of pseudorabies virus (PrV) was isolated and sequenced. Nucleotide sequence analysis revealed that the MCP142 gene has a single open reading frame of 3993 nucleotides (nt) encoding 1330 amino acids. The 4400-nt major RNA from the MCP142 gene was detected in PrV-infected cells. The 5' end of the transcript was located 60 nt upstream of the initiation codon. The 3' end of the transcript was located 18 nt downstream of a putative poly(A) signal sequence TATAAA and 133 nt downstream of the termination codon. In comparing amino acid sequence homology between MCP142 of PrV and other available herpesviruses MCP was shown to have 58% homology with herpes simplex virus type 1 and varicella-zoster virus, 27% with Epstein-Barr virus, and 24% with human herpesvirus 6 and human cytomegalovirus. It has greater homology with those of the alpha-herpesviruses than with those of the beta-herpesviruses and the gamma-herpesviruses.

[Study on identification of a calcium pyrophosphate dihydrate crystal using electron diffraction technique].

Electron diffraction technique was used to identify a single crystal in synovial fluid and tissue obtained from patients with pseudogout. One hundred and ten kinds of reciprocal lattice planes of the triclinic calcium pyrophosphate dihydrate (CPPD) crystal were made from its crystallographical data. Electron diffraction patterns obtained from synthesized crystal closely fitted to the reciprocal lattice planes. Synovial fluid of ten patients who had chondrocalcinosis in the knee joint were examined. Diffraction patterns of a single rhomboid-shaped crystal fitted to the reciprocal lattice planes and these kinds of crystals were identified as triclinic CPPD crystals. A single triclinic crystal in the calcified meniscus prepared for transmission electron microscopy was identified in the same way as above. On the other hand, there weren't enough crystallographical data on the monoclinic CPPD crystal to construct the reciprocal lattice planes. Instead of comparing diffraction patterns with the reciprocal lattice planes, diffraction patterns of a single rod-like crystal were compared with those of a synthesized monoclinic CPPD crystal. This comparison combined with energy dispersive elemental analysis made it possible to identify a single rod-like crystal as a monoclinic CPPD crystal.

Identification and sequence analysis of Escherichia coli purE and purK genes encoding 5'-phosphoribosyl-5-amino-4-imidazole carboxylase for de novo purine biosynthesis.

It has been shown that the Escherichia coli purE locus specifying 5'-phosphoribosyl-5-amino-4-imidazole carboxylase in de novo purine nucleotide synthesis is divided into two cistrons. We cloned and determined a 2,449-nucleotide sequence including the purE locus. This sequence contains two overlapped open reading frames, ORF-18 and ORF-39, encoding proteins with molecular weights of 18,000 and 39,000, respectively. The purE mutations of CSH57A and DCSP22 were complemented by plasmids carrying ORF-18, while that of NK6051 was complemented by plasmids carrying ORF-39. Thus, the purE locus consists of two distinct genes, designated purE and purK for ORF-18 and ORF-39, respectively. These genes constitute a single operon. A highly conserved 16-nucleotide sequence, termed the PUR box, was found in the upstream region of purE by comparing the sequences of the purF and purMN operons. We also found three entire and one partial repetitive extragenic palindromic (REP) sequences in the downstream region of purK. Roles of the PUR box and REP sequences are discussed in relation to the genesis of the purEK operon.

An outbreak of hepatitis A caused by consumption of raw oysters.

An outbreak of hepatitis A occurred among the members of the Japan Overseas Corporation Volunteers (JOCV) stationed in the Philippines. Seven out of 10 youths who ate, but none of five youths who did not eat raw oysters developed hepatitis. All these seven cases were positive for hepatitis A antibody (anti-HAV) and six of them were also positive for anti-HAV IgM (immunoglobulin M) 111 days after the infection.

[Histological study on radiosensitivity of cervical carcinoma with special reference to 3 subtypes of squamous cell carcinoma].

Morphological investigation were made on the radiosensitivity, according to the histological types; keratinizing(K), large cell nonkeratinizing(LNK), and small cell nonkeratinizing(S) type. The results obtained are as follows. Histological and cytological estimation of irradiated effects were examined at the 7th day after 10 Gray test-irradiation: (i) Histological studies were done with the biopsy specimens (76 cases): There were significant differences among 3 histological types (LNK greater than K greater than S, P less than 0.05). (ii) Cytological studies were done as well as histological ones (48 cases): There was a significance between two groups (LNK greater than S, K greater than S, P less than 0.05). (iii) There was a correlation between the histological and cytological irradiation effects. DNA histogram patterns were also examined spectrophotometrically. It seemed that the patterns after test-irradiation was broad and these peaks were situated rather to high ploidy in histologically and cytologically highly effective cases. Early ultrastructural findings after test-irradiation were as follows: (i) appearance of intra-nuclear "pseudoinclusion", (ii) swelling of mitochondria, endoplasmic reticulum, and so on. Although these characteristics presented the same sequence of events without regard to their histological types, they were not synchronized. It seemed that they appeared earlier after test-irradiation in histologically high-sensitivity cases than in low-sensitivity cases.

Composite ganglioneuroblastoma--a case report and short term culture of tumor cells.

A 1-year-old male infant showed cerebellar signs and symptoms, and excess urinary excretion of vanillylmandelic acid (VMA). Each of them disappeared spontaneously but a large abdominal tumor occurred at 3 years of age. Surgical specimen of the tumor was diagnosed as neuroblastoma and the patient died at 4 years of age. Autopsy revealed the huge tumor originated from the right adrenal, and occupying the retroperitoneum, pelvic cavity and posterior mediastinum. Histology of the primary lesion was chiefly that of ganglioneuroma, while all of the metastatic ones neuroblastoma. Transitional histology from neuroblastoma to ganglioneuroma was also observed in the primary lesion. The diagnosis was designated as composite ganglioneuroblastoma. The surgical material of the metastatic neuroblastoma was cultured for 2 months in vitro and the tumor cell clumps extended spontaneously abundant long neurites. This phenomenon suggests the high maturation ability of the neuroblastoma cells.

[Anaphylactoid purpura and the complement system: particularly in terms of C3a and IgA (author's transl)].

We measured the immunoglobulin, beta1 C globulin, beta1E globulin, C3a, and CH50 in 20 patients with anaphylactoid purpura (Schönlein-Henoch purpura). IgA values proved to be elevated in 5 out of 11 cases. beta1 C globulin values were lower in 2 out of 8 cases and fell within the range of normalcy in the remaining 6 cases. beta1 E globulin values were normal in all cases measured (5 patients). A 130% increase in C3a was found in 7 out of 9 cases; patients with higher C3a values suffered from lasting petechial hemorrhages. It can, therefore, be concluded that the activation of the "alternative pathway" induced by IgA led to the release of C3a.

Mitogenic activities of heteroglycan and heteroglycan-protein fractions from culture medium of Lentinus edodes mycelia.

The water soluble material (LEM) was prepared from the solid culture medium in which Lentinus edodes mycelia were growing actively. An alcohol insoluble material was prepared from LEM and subjected to Sepharose 6B gel filtration. The void fraction (LAP1) was composed mainly of xylose-rich heteroglycan and protein. From LAP1, a heteroglycan fraction (LAF1) was prepared by DEAE-Sepharose CL-6B column chromatography. LAP1 and LAF1 enhanced the incorporation of [3H]thymidine into mouse splenic cells (SPs). At each of the optimum doses, the rate of the incorporation was about 10 times as high with LAF1 as with LAP1. Such mitogenic responses were not induced in nylon-column effluent SPs and thymocytes. sIg-expressed cells were responsive to LAF1, but not to LAP1. Moreover, with each fraction, the incorporation was enhanced more in plastic adherent splenic cells (ADs) than in SPs. The flow cytometric assay revealed that the number of Mac-1+ cells is about 13 times as many in ADs as in SPs and that the number of Ly-5+ or Thy-1.2+ cells is considerably reduced in ADs compared with that in SPs. Thus, the present studies suggest that LAP1 and LAF1 act as mitogens predominantly for mouse splenic macrophages and/or monocytes.

Comparisons between invasive and noninvasive determinations of embryonic heart rate in chickens.

The embryonic heart rate (HR) of chicken eggs was determined by ballistocardiography, electrocardiography, impedance-cardiography and acoustocardiography. The effects of these different methods on the HR were examined for young (12-day-old) and late (16- and 18-day-old) embryos. Ballistocardiography is a non-invasive method. Nevertheless, the mean HRs determined using this method at 5-hr intervals were significantly different in many embryos. This difference was caused by the natural variability of embryonic HR. The statistical analyses indicated that any of these four methods did not influence the HR measurement and can be used as a simple means for determination of the embryonic HR.

Range of motion after Bankart repair. Vertical compared with horizontal capsulotomy.

We studied the range of shoulder motion of patients who underwent vertical as compared with horizontal capsulotomies during open Bankart repair for recurrent anterior dislocations of the shoulder. A vertical capsulotomy was used in 10 shoulders and a horizontal capsulotomy was used in 14 shoulders. Except for the method of capsulotomy, the surgical procedure and postoperative rehabilitation were the same. The range of motion was measured at 1.5, 2, 3, 4, 5, 6, 9, and 12 months after the surgery, and at the final follow-up (average, 49 months for the vertical and 26 months for the horizontal group). No dislocations recurred, and the anterior apprehension test was negative in all of the patients in both groups. External rotation in abduction was greater in the horizontal group than in the vertical group; the differences were significantly greater at 9 months and 12 months after surgery and at the final follow-up. External rotation in adduction, flexion, and internal rotation were not significantly different between the groups. We conclude that Bankart repair through a horizontal capsulotomy preserves a better range of external rotation in abduction than does a vertical approach.
