[A simulation study on brain hematoma real time monitoring by use of electrical impedance tomography].

It is significant to develop a new set of medical equipment in which electrical impedance tomography is applied to brain hematoma real time monitor. This paper describes some progress in reconstruction algorithm of EIT. The methods make it possible to improve the precision as well as computation speed dramatically. In addition, intensive simulation describes all the process for real time monitoring of brain hematoma by exploiting EIT.

[Extractible properties of PDLLA under different extraction conditions].

This assay was made to investigate the extractible properties of PDLLA under different extraction conditions. The ratio of test sample to extraction medium was 0.2 g to 1 ml. The distilled water, artificial saliva, Eagle's MEM and hexane were selected respectively as the extraction medium. The samples were extracted under 37 degrees C 24 h, 72 h, 1 w, 2 w and 3 w; 50 degrees C 72 h; 70 degrees C 24 h and 72 h; 121 degrees C 1 h. By use of gas chromatography, the ethyl alcohol, xylene and ethyl acetate content were measured. The results showed the ethyl alcohol content < 1.998 ug/ml, xylene contents was < 53.39 ug/ml and ethyl acetate content < 3.647 ug/m of PDLLA in distilled water, artificial saliva and Eagle's MEM under the condition of 37 degrees C from 24 h to 3 w. The ethyl alcohol content and xylene content in hexane were higher than those in the other three aqueous solutions. When the extracted temperature was increased, the contents of above three components were kept at the original level. There was almost no difference in the extractible properties of PDLLA among distilled water, artificial saliva and Eagle's MEM. The results did not change even if the extraction time and temperature were increased. It is a new concept to evaluate the safety of biomaterials by combining chemical and biological extraction tests, which will be significance in narrowing the gap between physical-chemical tests and biological tests for medical devices.

[Model of B immunoblastic lymphomas in the Hu-PBL-SCID mice].

OBJECTIVE: To constitute a model of B immunoblastic lymphomas in the Hu-PBL-SCID mice. METHODS: The SCID mice were reconstituted by intraperitoneal injection (i.p.) of 5 x 10(7) human lymphocytes from Epstein-Barr virus (EBV) seronegative individuals. After one week, the SCID mice were inoculated with EBV by i.p. injection, and subjected to the investigation of whether there was any tumor in the abdomen of such SCID mice four weeks later. The characteristics of the found tumor was observed by the methods of Hematoxylin-eosin (HE) stain, immunohistochemical staining and polymerase chain reaction (PCR). RESULTS: Compared with the control groups, all the EBV-infected Hu-PBL-SCID mice had abdominal solid tumors [(32 +/- 12.5) mm3] developed, often located in the liver. HE staining and immunohistochemical staining showed the tumors were human B cell lymphomas. EBV DNA could be detected in the tumors by the PCR. CONCLUSIONS: The model of B immunoblastic lymphomas in the Hu-PBL-SCID mice is successfully constituted, and may well be useful to the human tumor immunological study.

[Phosphorus transfer between mixed poplar and black locust seedlings].

In this paper, the 32P radio-tracer technique was applied to study the ways of phosphorus transfer between poplar (Populus euramericana cv. 'I-214') and black locust (Robinia pseudoacacia). A five compartment root box (18 cm x 18 cm x 26 cm) was used for testing the existence of the hyphal links between the roots of two tree species when inoculated with vesicular-arbuscular (VA) mycorrhizal fungus (Glomus mosseae). Populus I-214 (donor) and Robinia pseudoacacia (receiver) were grown in two terminal compartments, separated by a 2 cm root-free soil layer. The root compartments were lined with bags of nylon mesh (38 microns) that allowed the passage of hyphae but not roots. The top soil of a mixed stand of poplar and black locust, autoclaved at 121 degrees C for one hour, was used for growing seedlings for testing. In 5 compartment root box, mycorrhizal root colonization of poplar was 34%, in which VA mycorrhizal fungus was inoculated, whereas 26% mycorrhizal root colonization was observed in black locust, the other terminal compartment, 20 weeks after planting. No root colonization was observed in non-inoculated plant pairs. This indicated that the mycorrhizal root colonization of black locust was caused by hyphal spreading from the poplar. Test of tracer isotope of 32P showed that the radioactivity of the treatment significantly higher than that of the control (P < 0.05), 14 days from the tracer applied, to 27 days after, when VA mycorrhizal fungus was inoculated in poplar root. Furthermore, mycorrhizal interconnections between the roots of poplar and black locust seedlings was observed in situ by binocular in root box. All these experiments showed that the hyphal links was formed between the roots of two species of trees inoculated by VA mycorrhizal fungus. Four treatments were designed according to if there were two nets (mesh 38 microns), 2 cm apart, between the poplar and black locust, and if the soil in root box was pasteurized. Most significant differences of radioactivity among four treatments appeared 44 days after feeding 32P, the radioactivity of the day was applied to estimating the contribution of the various possible transfer ways to the total amount of nutrient transfer. Level of 32P radioactivity was found to be significantly (P < 0.05) higher in leaves of the treatment of "no separated and soil non-pasteurized" (17.1 pulse.g-1.s-1) than in leaves of "net separated and soil non-pasteurized" (5.3 pulse.g-1.s-1), and also significantly higher in leaves of "no net separated and soil pasteurized" (11.5 pulse.g.s-1) than in leaves of "net separated and soil pasteurized" (2.3 pulse.g-1.s-1), and very significantly (P < 0.01) higher in leaves of "no net separated and soil non-pasteurized" than in leaves of "net separated and soil pasteurized", whereas the levels of 32P radioactivity were not significantly different between the other treatments. The results showed that root contact and root exudations were the main ways of phosphorus transfer between the two species and the amount of phosphorus transfer through these two ways accounted for 62% of the total. The activity of the microorganisms including VA mycorrhizal fungi and the interaction between the microorganisms and root contact and root exudations made up 38% of total amount of phosphorus. The effect of mycorrhizal hyphal links in the direct nutrient transfer between poplar and black locust through separate mesh (38 microns) was little.

Hybrid methods for automated diagnosis of breast tumors.

OBJECTIVE: To design and analyze a new family of hybrid methods for the diagnosis of breast tumors using fine needle aspirates. STUDY DESIGN: We present a radically new approach to the design of diagnosis systems. In the new approach, a nonlinear classifier with high sensitivity but low specificity is hybridized with a linear classifier having low sensitivity but high specificity. Data from the Wisconsin Breast Cancer Database are used to evaluate, computationally, the performance of the hybrid classifiers. RESULTS: The diagnosis scheme obtained by hybridizing the nonlinear classifier ellipsoidal multisurface method (EMSM) with the linear classifier proximal support vector machine (PSVM) was found to have a mean sensitivity of 97.36% and a mean specificity of 95.14% and was found to yield a 2.44% improvement in the reliability of positive diagnosis over that of EMSM at the expense of 0.4% degradation in the reliability of negative diagnosis, again compared to EMSM. At the 95% confidence level we can trust the hybrid method to be 96.19-98.53% correct in its malignant diagnosis of new tumors and 93.57-96.71% correct in its benign diagnosis. CONCLUSION: Hybrid diagnosis schemes represent a significant paradigm shift and provide a promising new technique to improve the specificity of nonlinear classifiers without seriously affecting the high sensitivity of nonlinear classifiers.

[Study on cyto-compatibility of PGLA film for periodontal guided tissue regeneration].

This study was aimed to evaluate the cyto-compatibility of PGLA film for periodontal guided tissue regeneration (GTR) and their degradable products. Different extraction temperature, time and ratio were used to assess the cell relative growth rate (RGR) for PGLA. The degradable solution were brought into contact with cultured cells in vitro to observe the effects of cytotoxicity at 2, 4, 6, 8 and 10 weeks. The results showed when the extraction ratio was 0.1 g/ml, the extraction time has no effects on cytotoxicity at 37 degrees C. When the ratio was 0.1 cm2/ml, slight cytotoxic reaction appeared with the increase of extraction temperature (50 degrees C or 70 degrees C). When the extraction ratio was 0.5 cm2/ml in contract with the degradable solution at 37 degrees C for 72 h, the cell growth rate decreased. When the ratio was 6 cm2/ml, the cytotoxicity existed in some degree even if the temperature was at 37 degrees C for 24 h. After 2-4 w, the degradable solution had no obvious toxic effects on cells. However, the RGR gradually decreased as the degradation period increased. In conclusion, the effects of the extraction temperature, time, ratio and the degradation products accumulating in solution on RGR may exist. PGLA film has a good cytocompatibility.

Effects of scutellarin on liver function after brain ischemia/reperfusion in rats.

AIM: To investigate the effects of scutellarin (Scu) on liver function after brain ischemia/reperfusion in Wistar rats. METHODS: Rats were pretreated with Scu for 7 d and then subjected to a brain ischemia/reperfusion injury induced by a middle cerebral artery occlusion. The levels of nitric oxide (NO), xanthine oxidase (XOD), alanine transaminase (ALT), and aspartate aminotransferase (AST) in serum or liver tissues and the activities of antioxidant enzymes and cytochrome P450-dependent monooxygenases (CYPs) in liver tissues after brain ischemia/reperfusion were determined. RESULTS: In vehicle-treated rats, XOD, ALT, and AST activities (P<0.01) in serum and the MDA level (P<0.05) in liver tissues were all elevated but were significantly reduced (P<0.05) by Scu pretreatment. The NO levels in serum and liver tissues were decreased (P<0.01) dramatically in vehicle-treated rats and returned to the levels in the sham-operated animals when pretreated with Scu. SOD (P<0.05) and GSH-PX (P<0.01) activities in cytosol fraction were increased significantly by Scu pretreatment. Furthermore, a loss of CYP3A activity (P<0.01), but no changes of CYP1A1, CYP1A2, and CYP2E1 activities in liver were observed after brain ischemia/reperfusion in rats. Scu had no effect on them. CONCLUSION: These results demonstrated that pretreatment with Scu could attenuate hepatocellular damage elicited by brain ischemia/reperfusion in rats and this protection is in major part by its antioxidant activity.

[Reliability of detecting SARS-CoV antibody for diagnosis of SARS].

OBJECTIVE: To discuss the reliability of SARS-CoV antibody detection for SARS diagnosis. METHODS: Using SARS-CoV ELISA kit to detect relevant antibody in fresh serum of healthy, fever, probable, and suspect cases. RESULTS: The positive rate is 0%, 40%, and 95% respectively in healthy, probable, and suspect cases. CONCLUSIONS: It is reliable to detect SARS-CoV antibody in late suspect patients, but there will be high false-positive result in ordinary fever cases.

[Effects of one Chinese herbs on improving cognitive function and memory of Alzheimer's disease mouse models].

OBJECTIVE: To investigate Chinese herbs with the effect on significantly improving the cognitive function and retention of patients with Alzheimer's disease. METHOD: AD mouse models were established by injecting poly-beta-AP25-35 into the cerebral ventricle of mice. The curative effects of traditional Chinese herbs Dangguishaoyaosan, Chaihujialonggumulitang and two herbs (CHP I and CHP II) developed by the authors on improving the memory and cognitive function of AD mouse models were studied by the detection of behavioral and histochemical tests, with piracetam serving as control. RESULT: CHP II has profound curative effects on improving the memory and cognitive function of AD-like animal model. CONCLUSION: The present study indicates that it has a satisfactory prospect to seek new drugs from Chinese herbs to treat AD.

[The relationship between abnormal apoptosis and Fas expression of thymocytes from patients with myasthenia gravis].

AIM: To explore the roles of abnormalities of thymocytic abnormal apoptosis and its Fas expression in patients with myasthenia gravis (MG) in generation of autoimmune response. METHODS: The thymocytes were isolated and thymus extractive was prepared from fresh thymic tissue excised from MG patients. The proliferation of thymocyes was evaluated by MTT colorimetry. The apoptosis of thymocytes were examined by DNA electrophoresis. Expression of Fas was analyzed by a FACScan flow cytometry. Transcription and mutation of Fas gene were detected by RT-PCR and single-stranded conformation polymorphism ( SSCP) analysis. RESULTS: Thymus extractive could inhibit proliferation of normal thymocytes, while had no influence on the thymocytic proliferation of MG patients. The proliferation of thymocytes of both normal persons and MG patients could be inhibited by thymus extractive in the presence of dexamethasone. The inhibition rates were 58. 33% and 54. 26% respectively. The cocultured thymocytes with dexamethasone showed characteristic apoptosis pattern and the DNA electrophoresis exhibited 'ladder' appearance. The abnormal bands in Fas mRNA of partial MG patients' thymocytes were found by RT-PCR-SSCP analysis. CONCLUSION: Abnormalities of apoptosis and Fas gene expression of MG patients' thymocytes, and Fas gene mutation may be related to the pathogenesis and progression of MG.

Anti-nociceptive effects of diazepam binding inhibitor in the central nervous system of rats.

The present study investigated the effect of diazepam binding inhibitor (DBI) on nociception in the central nervous system of rats. There were dose-dependent increases in hindpaw withdrawal latency (HWL) to noxious thermal and mechanical stimulation after intrathecal injection of 1, 5 or 10 microg of DBI in rats, indicating a DBI-induced anti-nociceptive effect at the spinal levels of rats. Furthermore, it was found that there were no significant influences of intrathecal co-administration of gamma-aminobutyric acid (GABA) on the intrathecal DBI-induced increases in HWLs of rats. Intracerebroventricular administration of 1, 10 or 20 microg of DBI also induced dose-dependent increases in HWL to thermal and mechanical stimulation in rats, suggesting an anti-nociceptive effect of DBI in the brain. Moreover, there were no significant influences of intracerebroventricular co-administration of 2 microg of GABA on the intracerebroventricular DBI-induced increases in HWL of rats. The results of the present study demonstrated that DBI played anti-nociceptive effects in the central nervous system of rats.

Effects of pregnancy cocaine exposure on the mother and fetus: a murine model.

The aim of the experiments was to develop and characterize a murine model for investigating the effects of prenatal cocaine exposure on the mother and fetus. Pregnant mice were separated into three groups: group 1 was treated with cocaine HCl at 10 mg/kg twice daily (COC); group 2 was treated with saline at 10 ml/kg twice daily (SAL); and group 3 was pair-fed with the COC dams and was injected with saline following the same schedule (SPF) from embryonic day (E) 8 to 17. We utilized high-pressure liquid chromatography (HPLC) with UV detector and electrochemical detector to test the concentrations of cocaine, dopamine and serotonin, as well as HE staining to observe morphological alterations of liver and placenta. Though less food intake and lower weight gain were observed in COC and SPF groups but not in SAL dams, lower fetal body weight and brain weight were only seen in COC offspring. Pharmacological analysis revealed that cocaine was found in fetal plasma at 15 min following intraperitoneal administration on E17, accompanied with elevated concentrations of dopamine (DA) and serotonin (5-HT) in fetal brain. We also observed morphological changes in liver and placenta of cocaine-exposed fetuses. The present study indicates that pregnancy cocaine exposure can lead to maternal undernutrition and developmental abnormality of the fetal brain, liver and placenta. It is suggested that the developmental abnormality of the fetuses induced by cocaine is due to the toxicological effect of cocaine but not to maternal undernutrition.

Development of a rapid, simple dipstick dye immunoassay for schistosomiasis diagnosis.

In order to develop a rapid, simple immunodiagnostic assay for schistosomiasis, soluble egg antigen (SEA) of Schistosoma japonicum was conjugated with a blue colloidal dye (D-1) produced in China and used to detect antibodies in the sera of schistosomiasis patients. The antigen-antibody complex was captured by anti-human IgG absorbed onto a nitrocellulose membrane dipstick by means of immunochromatography. The results showed that the sensitivity of the dipstick dye immunoassay (DDIA) was 96.7% in 30 cases of acute schistosomiasis (29/30) and 94.1% (79/84) in 84 cases of chronic schistosomiasis. The specificity of the assay was 96.7% in 60 healthy subjects. Cross-reactions were observed in 10.0% of 20 cases of clonorchiasis and in 70.0% of 20 cases of paragonimiasis. The results were similar to those detected by routine ELISA. In a field evaluation of the DDIA kit, the positive rate of the DDIA was 96.7% in 121 cases of schistosomiasis, compared with 90.1% with the circumoval precipitin test (COPT). The antigen conjugated with dye was stable at room temperature for at least 6 months. The results indicated that the dipstick dye immunoassay provided high sensitivity and good specificity for the detection of schistosomiasis and the assay was rapid, simple and cheap, and did not need any equipment. It was more useful for screening target populations for selective chemotherapy than other immunoassays.

Overexpression of Smad7 results in severe pathological alterations in multiple epithelial tissues.

Biochemical studies have shown that Smad7 blocks signal transduction of transforming growth factor beta (TGFbeta); however, its in vivo functions are largely unknown. To determine the functions of Smad7, we have expressed Smad7 in transgenic mice, utilizing a keratin K5 promoter (K5.Smad7). K5.Smad7 mice exhibited pathological changes in multiple tissues and died within 10 days after birth. These mice were born with open eyelids and corneal defects, significantly delayed and aberrant hair follicle morphogenesis, and hyperproliferation in the epidermis and other stratified epithelia. Furthermore, K5.Smad7 mice developed severe thymic atrophy and massive thymocyte death, suggesting that Smad signaling in thymic epithelia is essential for thymocyte survival. Interestingly, in addition to a reduction in Smad phosphorylation, the protein levels of the receptors for TGFbeta, activin and bone morphogenetic protein were significantly decreased in the affected tissues of K5.Smad7 mice. Our study provides evidence that Smad7 is a potent in vivo inhibitor for signal transduction of the TGFbeta superfamily during development and maintenance of homeostasis of multiple epithelial tissues.

Dual functions of [alpha]4[beta]1 integrin in epicardial development: initial migration and long-term attachment.

The epicardium of the mammalian heart arises from progenitor cells outside the developing heart. The epicardial progenitor (EPP) cells migrate onto the heart through a cyst-mediated mechanism in which the progenitors are released from the tissue of origin as cysts; the cysts float in the fluid of the pericardial cavity and attach to the naked myocardial surface of the heart, and cells in the cysts then migrate out to form an epithelial sheet. In this paper, we show that the gene encoding the alpha4 subunit of alpha4beta1 integrin (alpha4beta1) is essential for this migratory process. We have generated a knockin mutation in mice replacing the alpha4 integrin gene with the lacZ reporter gene, placing lacZ under the control of the alpha4 integrin promoter. We show that in homozygous mutant embryos, the migration of EPP progenitor cells is impaired due to inefficient budding of the cysts and a failure of the cells in the cysts to migrate on the heart. This study provides direct genetic evidence for essential roles for alpha4beta1 integrin-mediated cell adhesion in the migration of progenitor cells to form the epicardium, in addition to a previous finding that alpha4beta1 is essential for maintaining the epicardium (Yang, J.T., H. Rayburn, and R.O. Hynes. 1995. Development. 121:549-560).

[Immunocontraceptive effect of a synthetic chimeric peptide of sperm].

OBJECTIVE: To investigate the immunocontraceptive ability of a new synthetic chimeric peptide with 35 amino acid residues. METHODS: A chimeric peptide with 35 amino acid residues was synthesized based on the amino acid sequence of SP17, a protein specific to murine testis/sperm, and cyritestin. The purified product, with keyhole limpet hemocyanin (KLH), was injected subcutaneously into the pad of female and male Balh/C mice. Blood was extracted from the ophthalmic arteries of the mice to identify the corresponding proteins from organs of mice and/or rat and human being by Western blotting. The chimeric peptide was injected into other female mice that were then raised together with male mice in the same cages to observe their status of pregnancy. Freund' adjuvant or normal saline were injected into the controls. RESULTS: The purity of the new chemiric peptide was 95%. The highest specific IgG titre in mouse serum was 1:6000 and the highest titre of specific IgA in the washing of vaginal mucous membrane of female mice was 1:300. The specific antiserum from the mice immunized by the chimeric peptide and KLH could identify two proteins with the molecular weights of 22 000 and 55 000 from testes of mice, rats, and human being. Western blotting showed no band for the extracts of mouse liver, spleen, and kidney. The pregnancy rate of female immunized mice was 61.11%, significantly lower that those in the controls (P < 0.05). The average number of cubs in a litter was less in the experimental group than in the controls (P < 0.05). CONCLUSION: The new chimeric peptide with 35 amino acid residues stimulates powerfully humoral and mucous immunity in mice and obviously decreases the pregnancy rate and the number of cubs in each litter in mice.

Mild benzene-thermal route to GaP nanorods and nanospheres.

GaP nanorods and nanospheres were synthesized from a mild benzene-thermal route at 240 and 300 degrees C, respectively, using Na, P, and GaCl(3) as the starting materials. The structure of the products was identified as zinc blende phase by X-ray powder diffraction (XRD). Transmission electron microscopy (TEM) images showed that, when the reaction temperature was 240 degrees C, the products were nanorods with widths of 20-40 nm and lengths of 200-500 nm and nanospheres with diameters of 20-40 nm. However, when the reaction temperature was increased to 300 degrees C, the products were only nanospheres, and the diameters increased to 40-60 nm. The reaction proceeded through a metallic gallium intermediate, and a solution-liquid-solid (SLS) mechanism was proposed for the one-dimensional growth. The products were also investigated by UV-vis absorption and X-ray photoelectron spectroscopy.
