Inhibition of human tumor xenograft growth in nude mice by a conjugate of monoclonal antibody LA22 to epidermal growth factor receptor with anti-tumor antibiotics mitomycin C.

Anti-EGFR monoclonal antibodies LA22 and Erbitux bind to different epitopes of EGFR. The chemimmunoconjugates of MMC with LA22 or Erbitux were prepared, and in vitro cytotoxicity assays with A549 cells showed that LA22-MMC was much more potent than Erbitux or Erbitux-MMC. Viabilities of A549 cells treated with LA22-MMC, Erbitux or Erbitux-MMC were 35%, 94%, and 81%, respectively. Immunoscintigraphy of xenografts of human A431 and A549 cells in nude mice both showed that (125)I-labeled-LA22-MMC enriched in tumor sites prominently. Most importantly, in vivo assays showed LA22-MMC was significantly more effective than free drug MMC in the treatment of subcutaneous xenografts of human A431 cells in nude mice (83% inhibition for LA22-MMC and 30% for MMC). We concluded that LA22-MMC could be a very potent drug for treatment of solid tumors.

Analysis of the sound field in finite length infinite baffled cylindrical ducts with vibrating walls of finite impedance.

This paper describes an analytical model of finite cylindrical ducts with infinite flanges. This model is used to investigate the sound radiation characteristics of the gradient coil system of a magnetic resonance imaging (MRI) scanner. The sound field in the duct satisfies both the boundary conditions at the wall and at the open ends. The vibrating cylindrical wall of the duct is assumed to be the only sound source. Different acoustic conditions for the wall (rigid and absorptive) are used in the simulations. The wave reflection phenomenon at the open ends of the finite duct is described by general radiation impedance. The analytical model is validated by the comparison with its counterpart in a commercial code based on the boundary element method (BEM). The analytical model shows significant advantages over the BEM model with better numerical efficiency and a direct relation between the design parameters and the sound field inside the duct.

[Pharmacokinetic studies of quercetin-hydroxypropyl-beta-cyclodextrin inclusion compound in rats].

To estabilish a HPLC method for determination of quercetin-hydroxypropyl-beta-cyclodextrin in rats plasma and investigate the pharmacokinetics characteristics of it in rats. The plasma was extracted with methanol-aceitc acid (9:1). The mobile phase was acetonitrilewater. Absorbance of the effluence was monitored at 360 nm. The linear limit of quercetin was within the range of 1 to 150 microg/ml. The lowest limit of quercetin was 0.5 microg/ml. The concentration-time curve of quercetin in rats was considered of a two-compartment open model, and the t1/2 (beta) of the inclusion compound when ig is 2.220 h, the t1/2 (beta) of the inclusion compound when iv is 90.871 min.

[Study on the inclusion compound of qucertin with hydroxypropyl-beta-cyclodextrin].

The inclusion compound of qucertin-HP-beta-CD was manufactured by aqueous solution-stirring method. The inclusion compound of qucertin-HP-beta-CD was confirmed by IR spectroscopy and differential theramal analysis (DTA). The results indicated that the inclusion compound of qucertion-HP-beta-CD was formed. The content analysis of the inclusion compound showed that the molecular ratio of qucertin to HP-beta-CD was 1:1. The solubility of qucertin increased when the drug was included by HP-beta-CD, which in water has added from 0.0392 mg/ml to 34.227 mg/ml.

[Determination of silicon, manganese and magnesium in cast iron by atomic emission spectrometry].

A spectrograph with high voltage spark source, once pre-burned and continuous photography was used for the quantitative determination of Silicon, Manganese and Magnesium in cast iron. The experimental conditions were optimized including the control of pre-burning time, plate moving rate, slit height of the blackness measuring, the selection of electrode material, the distance between the electrodes, shape of the electrode and the diminution of interferences. The determination range of contents (w/w) are 0.010%-0.090% for Mg, 1.50%-3.50% for Si and 0.40%-1.10% for Mn. The relative standard deviations are 10.4%, 1.9% and 3.0% for Mg, Si, and Mn, respectively.

Parallel profiling of active transcription factors using an oligonucleotide array-based transcription factor assay (OATFA).

Cell/tissue-specific gene expression are tightly regulated by various combinations of multiple transcription factors (TFs). Here, we present an oligonucleotide array-based transcription factor assay (OATFA), which allows the simultaneous assay of multiple active TFs. In this proof-of-principle work, both purified TFs and cell extracts were analyzed using OATFA and further antibody-based validation confirmed the chip data. This method could simplify the assay of multiple TFs and may facilitate high-throughput profiling of large numbers of TFs.
