Spatial scaling of metagenomic diversity reveals ecological disruption in the gut microbiome of gout patients.

Gout, a painful inflammatory arthritis, is characterized by hyperuricemia and monosodium urate crystal deposition, with growing evidence linking its pathogenesis to gut microbiome dysbiosis. However, traditional diversity metrics fail to capture the complex spatial organization of microbial communities. This study addresses this gap by applying the novel metagenomic Diversity-Area Relationship (m-DAR) model to investigate scaling laws in the gout microbiome-quantifying how metagenomic diversity changes with the number of individuals sampled. Our analysis of gut microbiomes from gout patients and healthy controls revealed fundamental ecological disruptions. We found that gout microbiomes exhibited significantly altered scaling patterns: they showed greater inter-individual dissimilarity (higher z-values) at the level of rare genes (q = 0), but weaker scaling of dominant genes (q = 1-3) compared to healthy controls. Crucially, the maximal accrual diversity (MAD) was substantially lower in gout patients, indicating a severely constrained potential for total microbial gene diversity. Furthermore, profiling of metagenomic functional gene clusters (MFGCs) uncovered widespread functional perturbations, including increased diversity scaling for carbohydrate-active enzymes (CAZy) but decreased scaling in essential metabolic pathways (KEGG, KO). These results demonstrate that the gout gut microbiome is defined by a loss of ecological structure, featuring reduced homogeneity in dominant taxa, expanded rare biosphere variation, and an overall collapsed diversity capacity. This work introduces an ecological framework for characterizing dysbiosis in gout that complements traditional diversity metrics and may inform the development of microbiome-based therapeutic strategies. Further research is needed to translate these ecological patterns into clinical applications.

Adsorption of hydrogen isotopes on micro- and mesoporous adsorbents with orderly structure.

The equilibrium and dynamic adsorption data of H(2) and D(2) on different micro- and mesoporous adsorbents with orderly structure including 3A, 4A, 5A, Y, and 10X zeolites; carbon CMK-3; silica SBA-15; and so forth were collected. Critical effect of the nanodimension of adsorbents on the adsorption behavior of hydrogen and its isotopes is shown. The highest adsorption capacity was observed at pore size 0.7 nm, but equal or even larger isotope difference in the equilibrium adsorption was observed at larger pore sizes, whereas the largest isotope difference in the dynamic adsorption was observed at 0.5 nm. The adsorption rate of D(2) is larger than that of H(2) in microporous adsorbents, but the sequence could be switched over in mesoporous materials. Linear relationship was observed between the adsorption capacity for hydrogen and the specific surface area of adsorbents although the adsorbents are made of different material, which provides a convincing proof of the monolayer mechanism of hydrogen adsorption. The linear plot for microporous adsorbents has a larger slope than that for mesoporous adsorbents, which is attributed to the stronger adsorption potential in micropores.

Identification and characterization of two rice autophagy associated genes, OsAtg8 and OsAtg4.

Autophagy is an intracellular process for vacuolar degradation of cytoplasmic components. The molecular machinery responsible for yeast and mammalian autophagy has begun to be elucidated at the cellular level. A genome-wide search revealed significant conservation among autophagy genes in yeast and Arabidopsis. Up till now, however, there is no report about rice autophagy associated genes. Here we cloned OsAtg8 and OsAtg4 from Oryza sativa and detected their expression patterns in various tissues. Immunoblotting analysis showed that carboxyl terminus of OsAtg8 can be cleaved in yeast cell. Mutation analysis revealed that the conserved Gly117 residue of OsAtg8 was essential for its characteristic C-terminal cleavage as similar to that found in mammalian and yeast Atg8. We further proved that OsAtg8 interacted with OsAtg4, and this interaction was not affected by the conserved Gly117 mutation. Our results demonstrate that Atg8 conjugation pathway is conserved in rice and may play important roles in rice autophagy.

Pose and position tracking with super image vector inner products.

We introduce a new and efficient distortion-invariant super image tracker and pose estimator based on a linear phase coefficient composite filter. The super image consists of a weighted sum of training images chosen to span the distortion range under analysis. Unlike correlation-based composite filter design, the super image is implemented by means of a complex vector inner product operation. A super image vector inner product is implemented by elementwise multiplication of a super image template by a window of interest in the input scene and summation of the elementwise operations. The resulting amplitude indicates target detection, and the resulting phase indicates the value of scale, orientation, or movement of the target object. The mathematical characteristics of the super image vector inner product are presented, and its application is demonstrated.

Molecular cloning and expression of a cDNA encoding Lon protease from rice (Oryza sativa).

The ATP-dependent Lon protease is a highly conserved enzyme that is present in archeae, eubacteria, and eukaryotes, and plays an important role in intracellular protein degradation. We have isolated a Lon protease gene, OsLon1, from Oryza sativa. The cDNA contained a 2,655 bp ORF. Comparative analysis showed that OsLon1 shared significant similarity with the previously reported Lon proteases from maize, Arabidopsis, human, and bacteria. Tissue expression pattern analysis revealed that OsLon1 was highly expressed in young leaves, mature leaves, and leaf sheaths but only weakly in young roots, mature roots, and young panicles. The OsLon1 gene was successfully expressed in E. coli and the detected protein size, about 120 kDa, matched the expected molecular mass of the His-tagged OsLon1 protein.

A series of heteroleptic bis(phthalocyaninato) europium(III) complexes, namely, Eu(Pc)[Pc(15C5)] (2), Eu(Pc)[Pc(opp-15C5)2] (3), Eu(Pc)[Pc(adj-15C5)2] (4), Eu(Pc)[Pc(15C5)3] (5), and Eu(Pc)[Pc(15C5)4] (6) [Pc = unsubstituted phthalocyaninate; Pc(15C5) = 2,3-(15-crown-5)phthalocyaninate; Pc(opp-15C5)2 = 2,3,16,17-bis(15-crown-5)phthalocyaninate; Pc(adj-15C5)2 = 2,3,9,10-bis(15-crown-5)phthalocyaninate; Pc(15C5)3 = 2,3,9,10,16,17-tris(15-crown-5)phthalocyaninate, Pc(15C5)4 = 2,3,9,10,16,17,24,25-tetrakis(15-crown-5)phthalocyaninate], with one, two, three, and four 15-crown-5 voids attached at different positions of one of the two phthalocyaninato ligands in the double-decker molecules, have been devised and prepared by Eu(Pc)(acac)-induced (Hacac = acetylacetone) mixed cyclization of the two corresponding phthalonitriles in the presence of organic base 1,8-diazabicyclo[5.4.0]undec-7-ene (DBU) in n-pentanol. For the purpose of comparative studies, homoleptic counterparts Eu(Pc)2 (1) and Eu[Pc(15C5)4]2 (7) have also been prepared. These sandwich double-decker complexes have been characterized by a wide range of spectroscopic methods in addition to elemental analysis. Their electrochemistry has also been studied by cyclic voltammetry (CV) and differential pulse voltammetry (DPV). The molecular structure of Eu(Pc)[Pc(15C5)4] (6) has been determined by X-ray diffraction analysis. Their supramolecular structure-formation properties, in particular for compounds 5 and 6 in the presence of potassium ions, have also been comparatively studied for the purpose of future functional investigation.

Haplotype variation in structure and expression of a gene cluster associated with a quantitative trait locus for improved yield in rice.

By constructing nearly isogenic lines (NILs) that differ only at a single quantitative trait locus (QTL), we fine-mapped the yield-improving QTL qGY2-1 to a 102.9-kb region on rice chromosome 2. Comparison analysis of the genomic sequences in the mapped QTL region between the donor (Dongxiang wild rice, Oryza rufipogon Griff.) and recurrent (Guichao2, Oryza sativa ssp. indica) parents used for the development of NILs identified the haplotypes of a leucine-rich repeat receptor kinase gene cluster, which showed extensive allelic variation. The sequences between genes in the cluster had a very high rate of divergence. More importantly, the genes themselves also differed between two haplotypes: Only 92% identity was observed for one allele, and another allele was found to have completely lost its allelic counterpart in Guichao2. The other six shared genes all showed >98% identity, and four of these exhibited obvious regulatory variation. The same haplotype segments also differed in length (43.9-kb in Guichao2 vs. 52.6-kb in Dongxiang wild rice). Such extensive sequence variation was also observed between orthologous regions of indica (cv. 93-11) and japonica (cv. Nipponbare) subspecies of Oryza sativa. Different rates of sequence divergence within the cluster have resulted in haplotype variability in 13 rice accessions. We also detected allelic expression variation in this gene cluster, in which some genes gave unequal expression of alleles in hybrids. These allelic variations in structure and expression suggest that the leucine-rich repeat receptor kinase gene cluster identified in our study should be a particularly good candidate for the source of the yield QTL.

Rectification of excitation with bathochromic shift induced by intense absorption of organic ligands during emission measurement of Eu(III) complex.

Bathochromic shift in excitation spectrum was observed during emission measurement of Eu(DBM)(3)Phen containing dilute solution in methyl methacrylate (MMA). Detailed analysis shows that the reason of bathochromic shift is not the formation of molecule aggregation. It is caused by the intense absorption of ligands in the complex. Based on this model, a new method has been established to rectify excitation spectra before emission measurement of systems with different concentration. There exists a critical value of the absorption strength, which is 0.87 from calculation. Higher absorption than this value will cause the bathochromic shift of excitation peak. The wavelength whose absorbance is 0.87 will be the position of the strongest excitation peak. With 200 ppm and 500 ppm Eu(DBM)(3)Phen as the standard sample, relations between relative concentration and wavelength of excitation peak in Eu(DBM)(3)Phen system were deduced and plotted. Theoretical curves are in good agreement with experiment data except extra-dilute concentration, for partial decomplexation of the beta-diketonate and phenanthroline ligands.

Differential expression of the intermediate filament protein nestin during renal development and its localization in adult podocytes.

Nestin, an intermediate filament protein, is widely used as stem cell marker. Nestin has been shown to interact with other cytoskeleton proteins, suggesting a role in regulating cellular cytoskeletal structure. These studies examined renal nestin localization and developmental expression in mice. In developing kidney, anti-nestin antibody revealed strong immunoreactivity in vascular cleft of the S-shaped body and vascular tuft of capillary loop-stage glomerulus. The nestin-positive structures also were labeled by endothelial cell markers FLK1 and CD31 in immature glomeruli. Nestin was not detected in epithelial cells of immature glomeruli. In contrast, in mature glomerular, nestin immunoreactivity was observed only outside laminin-positive glomerular basement membrane, and co-localized with nephrin, consistent with podocyte nestin expression. In adult kidney, podocytes were the only cells that exhibited persistent nestin expression. Nestin was not detected in ureteric bud and its derivatives throughout renal development. Cell lineage studies, using a nestin promoter-driven Cre mouse and a ROSA26 reporter mouse, showed a strong beta-galactosidase activity in intermediate mesoderm in an embryonic day 10 embryo and all of the structures except those that were derived from ureteric bud in embryonic kidney through adult kidney. These studies show that nestin is expressed in progenitors of glomerular endothelial cells and renal progenitors that are derived from metanephric mesenchyme. In the adult kidney, nestin expression is restricted to differentiated podocytes, suggesting that nestin could play an important role in maintaining the structural integrity of the podocytes.

High luminescence quantum yields and long luminescence lifetime from Eu(III) complex containing two crystal water based on a new beta-diketonate ligand.

New members of family of Eu(III) complex based on the thenoylacetophenone have been synthesized and characterized. The compounds were found for high metal luminescence quantum yields and long luminescence lifetime, especially for compound with two crystal water, corresponding with other compounds containing two crystal water. The result is attributed to high molar absorption coefficients of the Eu(III) complex according to UV-vis and emission spectra. The high molar absorption coefficients balance quenching effect from OH oscillators of water contained in compound.

Cloning, characterization and tissue-specific expression of a cDNA encoding a novel EMBRYONIC FLOWER 2 gene (OsEMF2) in Oryza sativa.

EMBRYONIC FLOWER 2 (EMF2) gene plays a major role in maintain vegetative development and repress flower development. Here, we present the cloning, characterization and tissue-specific expression of a putative EMF2 (OsEMF2) gene in Oryza sativa. The full-length cDNA of OsEMF2 was 1899 bp and contained an 1872 bp open reading frame (ORF) encoding a 624 amino acid protein. Homologous analysis showed that OsEMF2 contain a single conserved C2H2-type zinc finger motif. Sequence alignment shows that there is a homology between the deduced amino acid sequence of OsEMF2 and EMF2 in Zea mays (55%). Moreover, pI of OsEMF2 are predicted. The tissue-specific expression pattern of OsEMF2 reveals that it is abundant in shoot apical meristem and inflorescence meristem, while its expression level is much lower in leaf, root, immature seed and callus.

Photodermatitis with minimal inflammatory infiltrate: clinical inflammatory conditions with discordant histologic findings.

Dermatoses associated with cutaneous photosensitivity are a group of photodistributed skin eruptions caused or exacerbated by light. Multiple clinical variants of photosensitive dermatoses have been characterized including polymorphous light eruption, chronic actinic dermatitis, solar urticaria, phototoxic and photoallergic dermatitis, reticular erythematous mucinosis, acute cutaneous lupus erythematosus, and dermatomyositis. As there may be significant overlap among the clinical presentation of these conditions, the specific diagnosis of individual photodermatosis relies heavily on characteristic histopathologic features. We present here 5 cases of photodistributed eruptions with virtual absence of histologic epidermal changes and dermal inflammation, yet all were described clinically as being "inflammatory" and erythematous. All cases of this "pauci-inflammatory photodermatitis" presented with photodistributed bright red macular erythema or slightly indurated plaques that developed over a period of weeks to months and clinically resembled photoallergic or phototoxic drug reactions or polymorphous light eruption. Microscopically, however, only very sparse dermal lymphocytic infiltrate was noted with no or minimal epidermal changes. To our knowledge, the observation of clinically evident photodistributed dermatoses that demonstrate such minimal histopathologic findings has not been reported. Clinicians and histologists should be aware of the disparity that may be encountered in this setting, as the clinical features are usually far more impressive than those seen histologically.

Euthyroid sick syndrome in trauma patients with severe inflammatory response syndrome.

OBJECTIVE: To investigate the alternations of thyroid hormone in traumatic patients with severe inflammatory response syndrome (SIRS). METHODS: Fifty traumatic patients with severe SIRS were enrolled and divided into two groups according to whether they presented multiorgan dysfunction syndrome (MODS). Thyroid hormone measurements were taken, including total triiodothyronine (TT3), total thyroxine (TT4), free triiodothyronine (FT3), free thyroxine (FT4) and thyroid stimulating hormone (TSH). The acute physiology and chronic health evaluation II (APACHE II) score was calculated according to clinical data. The outcomes of recovery or deterioration were recorded, as well as the length of time from the onset of SIRS to the time thyroid hormones were measured. RESULTS: Euthyroid sick syndrome (ESS) was presented in 45 cases. TT3 level was negatively correlated with APACHE II score (r = -0.330, P<0.05), and TT3/TT4 value was negatively correlated with the duration of SIRS( r = -0.316, P<0.05). TT3, TT4 and FT3 levels in MODS patients were significantly lower than those without MODS (P<0.05). MODS patients got low TT4 or FT4 level more frequently than those without MODS (P<0.05). Compared with the patients in normal TSH group, the patients with decreased TSH had lower T3, T4, recovery rate and higher APACHE II scores, MODS incidence, but there was no difference between two groups (P>0.05). CONCLUSIONS: Trauma patients with severe SIRS have high possibility to get ESS, which occurs more frequently and severely in MODS patients. It shows the influences of SIRS on the thyroid axes. With the persistence and aggravation of SIRS, there is a progressive reduction of thyroid hormone.

Changes of dipalmitoyl phosphatidyl choline after mechanical ventilation in patients with acute cerebral injury.

OBJECTIVE: To detect the levels of dipalmitoyl phosphatidyl choline (DPPC) in the sputum of the patients with acute cerebral injury without primary pulmonary injury after mechanical ventilation treatment. METHODS: DPPC levels in sputum of 35 patients with acute cerebral injury but without pulmonary injury were detected with high performance liquid chromatography at the beginning of ventilation and 16-20 days, 21-40 days, and 41-60 days after ventilation, respectively. RESULTS: There was no significant difference of the DPPC levels between 16-20 days after ventilation (3.36+/-0.49) and at the beginning of ventilation (3.37+/-0.58) (P>0.05). The mean levels of DPPC decreased significantly at 21-40 days (2.87 mg/ml+/-0.26 mg/ml, P<0.05) and 41-60 days (1.93 mg/ml+/-0.21 mg/ml, P<0.01) after ventilation compared with that at the beginning of ventilation. At the same period, the peak inspiratory pressure and the mean pressure of airway increased significantly, whereas the static compliance and the partial pressure of oxygen in artery decreased significantly. Among the 25 patients who received ventilation for more than 20 days, 8 (32%) had slightly-decreased partial pressure of oxygen in artery compared with that at the beginning of ventilation. CONCLUSIONS: Mechanical ventilation can decrease the DPPC levels, decrease the lung compliance and increase the airway pressure, even impair the oxygenation function in patients with acute cerebral injury. Abnormal DPPC is one of the major causes of ventilator-associated lung injury.

High performance organic field-effect transistors based on amphiphilic tris(phthalocyaninato) rare earth triple-decker complexes.

LB films of three amphiphilic tris(phthalocyaninato) rare earth triple-decker complexes with crown-ethers as hydrophilic heads and long alkyl chains as hydrophobic tails have been prepared and found to display very well ordered layer structures, as proved by pi-A isotherms, UV-vis and polarized absorption spectra, X-ray diffraction experiments, and microscopic morphology characterization. These LB films have been fabricated into field-effect transistor (FET) devices, which show carrier mobilities as high as 0.24-0.60 cm2 V-1 s-1, among the highest mobilities achieved thus far for all LB film-based OFETs.
