Changes in neocortical and hippocampal GABAA receptor subunit distribution during brain maturation and aging.

gamma-Aminobutyric acid type A (GABA(A)) receptors are the most important inhibitory receptors in the central nervous system, playing a pivotal role in the regulation of brain excitability. The pentameric receptor is commonly composed of different alpha, beta, and gamma subunits which mediate the function and pharmacology of the receptor and show regional- and temporal-specific expression patterns. Under varying physiological and pathophysiological conditions, this diversity allows a multitude of adaptive changes in subunit composition leading to distinct biological and pharmacological properties of the receptor. Here, we investigated the expression of five major GABA(A) receptors subunits (alpha1, alpha2, alpha3, alpha5, gamma2) in early postnatal, adult, and aged rat brains. Immunohistochemistry was performed at postnatal day 10, 30, 60, 90, 180, 360, and 540. Morphological and semi-quantitative evaluations of regional optical densities revealed specific regional and temporal expression patterns for all subunits. The study clearly demonstrated that changes in GABA(A) receptor distribution not only occur in the early postnatal cortex and hippocampal formation but also during later periods in the adolescent and aging brain. These findings contribute to a better understanding of age-related changes in brain excitability and further elucidate the distinct pharmacological effects of different GABAergic drugs in young and elderly patients.

Coherence current, coherence vortex, and the conservation law of coherence.

Introducing scalar and vector densities for a mutual coherence function, we present a new conservation law for optical coherence of scalar wave fields in the form of a continuity equation. This coherence conservation law provides new insights into topological phenomena for the complex coherence function. Some properties related to the newly introduced coherence vector density, such as a circulating coherence current associated with a coherence vortex, are investigated both theoretically and experimentally for the first time.

Plasmid pWW115, a cloning vector for use with Moraxella catarrhalis.

The plasmid shuttle vector pWW102B is able to replicate in only a modest number of Moraxella catarrhalis strains. Plasmid pWW115, a spontaneous deletion mutant of pWW102B, was shown to lack both the pACYC184-derived origin of replication and the associated chloramphenicol-resistance gene but was able to replicate in every M. catarrhalis strain tested in this study, including one strain that had been previously refractory to all types of genetic manipulations. To test the utility of this plasmid, a M. catarrhalis gene encoding the UspA2 serum-resistance factor was cloned into pWW115 and the resultant recombinant plasmid was shown to confer serum-resistance on a serum-sensitive M. catarrhalis uspA2 mutant.

[In vitro gene transfection into rabbit articular chondrocytes mediated by recombinant adeno-associated virus vector].

OBJECTIVE: To investigate the in vitro efficiency of recombinant adeno-associated virus (rAAV2) vector-mediated gene transfection into the rabbit articular chondrocytes. METHODS: Articular chondrocytes were isolated from New Zealand rabbits, cultured, and transfected with rAAV2 containing enhanced green fluorescent protein (eGFP) gene at different values multiplicity of infection (MOI). Successful transfection of chondrocytes was confirmed by detection of recombinant enhanced green fluorescent protein using inverted fluorescence microscopy. The transfection efficiency was determined using the fluorescence-activated cell sorter. A maximum level of 4.76% was set as the background autofluorescence in live, uninfected chondrocytes. RESULTS: The percentage of eGFP-positive chondrocytes increased in a vector dose-dependent manner. When the MOI value was <1 x 10(5) v.g./cell the transfection rate was positively linearly correlated with the dose. However, the MOI > 10(5) v.g./cell did not markedly increase the transfection rate. The greatest population of eGFP-expressing cells was 97.7% and the highest mean fluorescence intensity was 17.4 7 days after transfection. EGFP gene expression remained high until 56 days after transfection. CONCLUSION: The in vitro efficiency of rAAV2 vector-mediated eGFP gene transfection into rabbit articular chondrocytes was high. The eGFP expression can be maintained for a rather long time. EGFP gene is an ideal tracer for chondrocytes transduction.

Effect of cocaine on germ cell apoptosis in rats at different ages.

AIM: To investigate the effect of cocaine on apoptosis and caspase-3 activity in germ cells in male rats at different ages. METHODS: Cocaine hydrochloride was given (15 mg/kg body weight s.c.) to male Sprague-Dawley rats of 3 weeks (n = 8), 6 weeks (n = 8) and 12 weeks (n = 8) of age, daily for 28 Days. The serum levels of follicle stimulating hormone (FSH), luteinizing hormone (LH), prolactin (PRL), testosterone (T) and estrogen (E2) were assayed, and the DNA fragmentation of germ cells was determined by gel eletronphoresis. The cell cycle, apoptosis and caspase-3 activity of germ cells were tested by flow cytometry. RESULTS: After the 28-day cocaine treatment, testes weight of the 3-week-old rats, the testes and body weights of the 6-week-old rats were decreased significantly compared to those of their corresponding controls (P < 0.05). The serum level of T was decreased significantly in the 3-week-old and 6-week-old rats, and the serum level of PRL was also decreased significantly in 12-week-old rats compared to the controls (P < 0.05). In all the three cocaine-treated groups, the isolated DNA displayed a clear ladder pattern, especially in the 6-week old rats. The number of apoptosic germ cells increased significantly in 3- and 6-week-old rats treated with cocaine (P < 0.05). The caspase-3 activity in all three groups increased significantly compared to the controls (P < 0.05), especially in the 6-week-old rats. CONCLUSION: Cocaine exposure for 28 Days leads to significant damage to male gonad and apoptosis elevation in testes of rats of different ages, especially in those of 6 weeks of age. The increase in caspase-3 activity might be a key pathway related to the early stage of apoptosis as the mechanism of cocaine-induced germ cell loss.

Electron transport dynamics in a room-temperature Au nanoparticle molten salt.

A room-temperature Au38 nanoparticle polyether melt has been prepared by exchanging poly(ethylene glycol) (PEG) thiolate ligands, HS-C6-PEG163, into the organic protecting monolayer of Au38(PhC2)24 nanoparticles. Spectral and electrochemical properties verify that the Au38 core size is preserved during the exchange. Adding LiClO4 electrolyte, free PEG plasticizer, and/or partitioned CO2 leads to an ionically conductive nanoparticle melt, on which voltammetric, chronoamperometric, and impedance measurements have been made, respectively, of the rates of electron and ion transport in the melt. Electron transport occurs by electron self-exchange reactions, or electron hopping, between diffusively relatively immobile Au38(0) and Au38(1+) nanoparticles. The rates of physical diffusion of electrolyte ions (diffusion coefficients DCION) are obtained from ionic conductivities. The measured rates of electron and of electrolyte ion transport are very similar, as are their thermal activation energy barriers, observations that are consistent with a recently introduced ion atmosphere relaxation model describing control of electron transfer in semisolid ion and electron-conductive media. The model has been previously demonstrated using a variety of metal complex polyether melts; the present results extend it to electron transfers between Au nanoparticles. In ion atmosphere relaxation control, measured rates and energy barriers for electron transfer are not intrinsic values but are instead characteristic of competition between back-electron transfer caused by a Coulombic disequilibrium resulting from an electron transfer and relaxation of counterions around donor-acceptor reaction partners so as to reachieve local electroneutrality.

[Association of baseline cholesterol level and its 10-year change with carotid atherosclerosis in the population of Peking University community].

OBJECTIVE: To explore the association of baseline cholesterol level in 1992 and its 10-year change from 1992 to 2002 with the prevalence of carotid atherosclerosis. METHODS: In 1992, 1985 subjects of the community of Peking University, aged 35 - 64, underwent physical examination and blood-lipid test to study the cardio-vascular disease (CVD) risk factors. In 2002, the second survey on CVD risk factors was conducted among the existing 1932 subjects, now aged 45 - 74. In addition, B-mode ultrasonography of the carotid artery was performed too. The complete data of 1331 subjects were analyzed. RESULTS: (1) In 2002, the prevalence rates of intimal medial thickening (IMT) and carotid plaque in the study population were 47.8% and 29.9% respectively. (2) The post-10-year prevalence rate of carotid plaque was 19.7% in the subjects with the baseline cholesterol < 3.64 mmol/L, and was 46.8% in those with the baseline cholesterol 6.24 mmol/L, with a thickening rate 3.1 times that of the subjects with the baseline cholesterol < 3.64 mmol/L. The higher the baseline TV level, the higher the prevalence rates of carotid plaque and IMT. (3) The prevalence rate of carotid plaque was 32.0% in the subjects with normal TC in both 1992 and 2002, and 50.0% in those with hypercholesterolemia in both 1992 and 2002. CONCLUSION: Carotid atherosclerosis is a prevalent disorder in the people aged 45 - 74 in Beijing. The prevalence rates of carotid plaque and IMT increase with the elevation of baseline cholesterol level.

Synthesis and infrared and fluorescence spectra of rare earth complexes with a novel amide-based ligand.

A novel amide-based open-chain crown ether, N,N'-1,3-propanediyl-bis[2-(benzyl -carbamoyl-methoxy)-benzamide] (L) and its solid complexes with rare earth nitrates and picrates have been prepared. The complexes were characterized by elemental analyses, molar conductivity and IR spectra. The fluorescence properties of the Eu(III) and Tb(III) complexes in solid and in organic solvents were studied. Under the excitation of ultraviolet light, these complexes exhibit characteristic emission of europium and terbium ions. The results show that the ligand favor energy transfers to the emitting energy level of Tb(III). Some factors that influence the fluorescent intensity were also discussed.

Enantio- and diastereoselective Michael addition reactions of unmodified aldehydes and ketones with nitroolefins catalyzed by a pyrrolidine sulfonamide.

Chiral (S)-pyrrolidine trifluoromethanesulfonamide has been shown to serve as an effective catalyst for direct Michael addition reactions of aldehydes and ketones with nitroolefins. A wide range of aldehydes and ketones as Michael donors and nitroolefins as acceptors participate in the process, which proceeds with high levels of enantioselectivity (up to 99 % ee) and diastereoselectivity (up to 50:1 d.r.). The methodology has been employed successfully in an efficient synthesis of the potent H(3) agonist Sch 50917. In addition, a practical three-step procedure for the preparation of (S)-pyrrolidine trifluoromethanesulfonamide has been developed. The high levels of stereochemical control attending Michael addition reactions catalyzed by this pyrrolidine sulfonamide, have been investigated by using ab initio and density functional methods. Transition state structures for the rate-limiting C--C bond-forming step, corresponding to re- and si-face addition to the reactive conformation of the key enamine intermediates have been calculated. Analysis of these structures indicates that hydrogen bonding plays an important role in catalysis and that the energy barrier for si-face attack in reactions of aldehydes to form 2R,3S products is lower than that for the re-face attack leading to 2S,3R products. In contrast, the energy barrier for re-face addition is lower than that for si-face addition in reactions of ketones. The computational results, which are in good agreement with the experimental observations, are discussed in the context of the stereochemical course of these Michael addition reactions.

Exteroceptive suppression of temporalis muscle activity in subjects with high and low aggression traits.

STUDY AIM: There is evidence that the second exteroceptive suppression period of temporalis muscle activity (ES2) is modulated by the 5-HT neuronal activity in the brainstem, and the aggression trait is also connected with the cerebral 5-HT neuronal innervation. We therefore studied the temporalis ES2 in subjects with high and low aggression traits. METHODS: Sixty-five subjects with either low or high aggression trends, judged by clinical interview, answered the Zuckerman-Kuhlman Personality Questionnaire (ZKPQ), the Zuckerman Sensation Seeking Scales (SSS) and the Plutchik-van Praag Depression Inventory (PVP). These also underwent a temporalis ES2 test procedure. Twenty-two subjects with a high ZKPQ aggression trait score and 27 with a low score were selected for data analysis. RESULTS: On average, the high aggression group displayed significantly reduced temporalis ES2 duration, elevated ZKPQ Impulsive Sensation Seeking, Neuroticism-Anxiety and Aggression-Hostility, and PVP scores. The personality traits were not related with either latency or duration of temporalis ES2 in any group. The PVP score, however, was negatively correlated with ES2 duration in all 49 subjects. CONCLUSIONS: This study demonstrates neurophysiologic signs of brainstem dysfunction in subjects with high aggression traits.

Polymorphisms of the low-density lipoprotein receptor-related protein 5 (LRP5) gene are associated with obesity phenotypes in a large family-based association study.

BACKGROUND: The low-density lipoprotein receptor-related protein 5 (LRP5) gene, essential for glucose and cholesterol metabolism, may have a role in the aetiology of obesity, an important risk factor for diabetes. PARTICIPANTS AND METHODS: To investigate the association between LRP5 polymorphisms and obesity, 27 single-nucleotide polymorphisms (SNPs), spacing about 5 kb apart on average and covering the full transcript length of the LRP5 gene, were genotyped in 1873 Caucasian people from 405 nuclear families. Obesity (defined as body mass index (BMI) >30 kg/m(2)) and three obesity-related phenotypes (BMI, fat mass and percentage of fat mass (PFM)) were investigated. RESULTS: Single markers (12 tagging SNPs and 4 untaggable SNPs) and haplotypes (5 blocks) were tested for associations, using family-based designs. SNP4 (rs4988300) and SNP6 (rs634008) located in block 2 (intron 1) showed significant associations with obesity and BMI after Bonferroni correction (SNP4: p<0.001 and p = 0.001, respectively; SNP6: p = 0.002 and 0.003, respectively). The common allele A for SNP4 and minor allele G for SNP6 were associated with an increased risk of obesity. Significant associations were also observed between common haplotype A-G-G-G of block 2 with obesity, BMI, fat mass and PFM with global empirical values p<0.001, p<0.001, p = 0.003 and p = 0.074, respectively. Subsequent sex-stratified analyses showed that the association in the total sample between block 2 and obesity may be mainly driven by female subjects. CONCLUSION: Intronic variants of the LRP5 gene are markedly associated with obesity. We hypothesise that such an association may be due to the role of LRP5 in the WNT signalling pathway or lipid metabolism. Further functional studies are needed to elucidate the exact molecular mechanism underlying our finding.

Notable norbornene (NBE) incorporation in ethylene-NBE copolymerization catalysed by nonbridged half-titanocenes: better correlation between NBE incorporation and coordination energy.

CpTiCl2(N=CtBu2) exhibits both remarkable catalytic activity and efficient norbornene (NBE) incorporation for ethylene-NBE copolymerization: the NBE incorporation by Cp'TiCl2(X) (X = N=CtBu2, O-2,6-iPr2C6H3; Cp' = Cp, C5Me5, indenyl) was related to the calculated coordination energy after ethylene insertion.

Preparation of spherical iron nanoclusters in ethanol-water solution for nitrate removal.

In this study, a higher surface area spherical nanoscale zero valent iron (HNZVI) cluster (80 nm, 54.25 m(2)g(-1)) was synthesized in ethanol-water mixed solvent in the presence of dispersion agent of polyglycol (PEG). At the same time, a lower surface area nanoscale zero valent iron (LNZVI) particle (80 nm, 8.08 m(2)g(-1)) was also prepared with only de-ioned water as reaction media. Their structures, compositions and physical properties were characterized by transmission electron microscope (TEM), X-ray diffractometer (XRD), inductively coupled plasma atomic emission spectrophotometer (ICP-AES), and Brunauer-Emmett-Teller (BET) surface area analyzer and the results obtained for these two kinds of nanoscale iron were compared with each other and also with those reported in the literatures. The HNZVI clusters seemed to be accumulated by smaller iron particles (<10 nm). At the same time, whiskers were formed in the final produce. Reactivity of the HNZVI was affirmed via denitrification of nitrate. The factors controlling the reduction of nitrate, such as pH, dissolved oxygen (DO), iron content as well as the initial nitrate concentration were also discussed. Finally, kinetic analysis revealed that chemical reduction of nitrate by HNZVI could not be described by the first- or pseudo-first-order kinetic model.

Proteomic characterization of transient expression and secretion of a stress-related metalloprotease in high cell density culture of Bacillus megaterium.

Intracellular and extracellular proteome analysis was carried out by combined two-dimensional gel electrophoresis and mass spectrometric analysis (2DE/MS) for high cell density fed-batch culture of recombinant Bacillus megaterium strains. In the early feeding phase with a constant growth rate of 0.12h(-1) under glucose limitation, high expression and secretion of a metalloprotease (referred as Bmg1465) was detected. The transient appearance of this metalloprotease was found both as cell-associated and as secreted into the culture medium. Searching homologous proteins for functional assignment led to an unambiguous identification of Bmg1465 as a zinc-binding metalloprotease of the type immune inhibitor A (InhA). Metalloproteases of this type are currently considered as typical virulence factors associated with pathogenic Bacillus species. The result raises questions concerning the intrinsic function(s) of Bmg1465 in B. megaterium, which has the GRAS status, with respect to its stress response in high cell density culture.

Detection of alkaline phosphatase using surface-enhanced Raman spectroscopy.

A new approach was developed to detect the activity of alkaline phosphatase (ALP) enzyme at ultralow concentrations using a surface-enhanced Raman scattering (SERS) technique. The approach is based on the use of gold nanoparticles as a SERS material whereas 5-bromo-4-chloro-3-indolyl phosphate (BCIP) is used as a substrate of ALP. The enzymatic hydrolysis of BCIP led to the formation of indigo dye derivatives, which were found to be highly SERS active. For the first time, we were able to detect ALP at a concentration of approximately 4 x 10(-15) M or at single-molecule levels when ALP was incubated with BCIP for 1 h in the Tris-HCl buffer. The same technique also was successfully employed to detect surface-immobilized avidin, and a detection limit of 10 ng/mL was achieved. This new technique allows the detection of both free and labeled ALP as a Raman probe in enzyme immunoassays, immunoblotting, and DNA hybridization assays at ultralow concentrations.

Synthesis and structure-activity relationships of 3,4-diaminocyclobut-3-ene-1,2-dione CXCR2 antagonists.

A novel series of 3,4-diaminocyclobut-3-ene-1,2-diones was prepared and found to show potent inhibitory activity of CXCR2 binding and IL-8-mediated chemotaxis of a CXCR2-expressing cell line. Microsome stability and Caco2 studies were subsequently used to show that compounds of this chemotype are predicted to have good oral bioavailability and are thus suitable for pharmaceutical development.

A primitive fish provides key characters bearing on deep osteichthyan phylogeny.

Osteichthyans, or bony vertebrates, include actinopterygians (teleosts and their relatives) and sarcopterygians (coelacanths, lungfishes and tetrapods). Despite features found in basal actinopterygians (for example, Dialipina and Ligulalepis) and basal sarcopterygians (for example, Psarolepis and Achoania), the morphological gap between the two lineages remains wide and how sarcopterygians developed a dermal surface covering known as cosmine (composed of a pore-canal network and a single layer of odontodes and enamel) is still poorly known. Here we describe a primitive fossil fish, Meemannia eos gen. et sp. nov., that possesses an actinopterygian-like skull roof and a cosmine-like dermal surface combining a pore-canal network (found in various fossil sarcopterygians) with superimposed layers of odontodes and enamel (previously known in actinopterygians and some acanthodians). This 405-million-year-old fish from the Lower Devonian of Yunnan (China) demonstrates that cosmine in many fossil sarcopterygians arose step by step through the acquisition of a pore-canal network followed by the subsequently developed ability to resorb previous generations of odontodes and enamel. Meemannia provides key characters for studying deep osteichthyan phylogeny and indicates a possible morphotype for the common ancestor of actinopterygians and sarcopterygians.

Distribution of G-actin is related to root hair growth of wheat.

BACKGROUND AND AIMS: Actin distribution in root hair tips is a controversial topic. Although the relationship between Ca2+ gradient and actin dynamics in plant tip-growth has been a focus of study, there is still little direct evidence on the exact relationship in root hair tip-growth. METHODS: G-actin was labelled by fluorescein isothiocyanate-DNase I. F-actin was labelled by tetramethylrhodamine isothiocyanate-phalloidin. Actin in root hairs of Triticum aestivum (wheat) was investigated using confocal laser-scanning microscopy. KEY RESULTS: Thick F-actin bundles did not extend into a region of approx. 5-10 microm from the tip of the growing root hairs, although they gave off branches of fine actin filaments in the hair tips. A tip-focused G-actin gradient was shown at the extreme apex of growing root hairs. In full-grown wheat root hairs, the tip-focused G-actin gradient disappeared while the thick F-actin bundles extended into the tips. BAPTA-AM, a Ca2+ disruption agent, also caused the tip-focused G-actin gradient to disappear and the diffuse F-actin bundles to appear in the tips of wheat root hairs. CONCLUSIONS: These results suggest that the tip-focused gradient of intracellular G-actin concentration at the extreme apex may be essential for root hair growth, and that preserving the tip-focused gradient needs a high Ca2+ concentration in the root hair tips.
