[Experimental study of controlled release microencapsulated Salmonella typhi capsular polysaccharide vaccines immunized mice].

Salmonella Typhi capsular polysaccharide vaccines were encapsulated in the Micro-particles made from polyethylene glycol-poly-DL-lactide (PELA). BALB/c mouse were divided into three groups with 20 mice in each. Mouse were immunized respectively with controlled release microencapsulated Salmonella Typhi capsular polysaccharide vaccines and Salmonella Typhi capsular polysaccharide vaccines by oral and subcutaneous administration. The mice blood and salvia were collected at the 2nd, 4th and 8th weeks respectively for the titrating of IgG and sIgA antibodies by RIA. At the 8th week, live typhoid bacteria were injected into the immunized mice for the calculation of the rate of immunization protection. The IgG titers of the controlled release microencapsulated Salmonella Typhi capsular polysaccharide vaccines group were higher than those of the other groups(P < 0.05). The IgA titers of the low groups of controlled release microencapsulated Salmonella Typhi capsular polysaccharide vaccines (oral and subcutaneous) were higher than those of the group of Salmonella Typhi capsular polysaccharide vaccines (P < 0.05). The immunization protection rates of the three groups were 40%, 100% and 60% respectively. The controlled release microencapsulated Salmonella Typhi capsular polysaccharide vaccines possess the advantages of releasing slowly in vivo and persisting long time immunogenicity.

A rapid and sensitive liquid chromatography/negative ion tandem mass spectrometry method for the determination of an indolocarbazole in human plasma using 96-well diatomaceous earth plates for solid-liquid extraction [correction of using internal standard (IS) 96-well diatomaceous earth plates for solid-liquid extraction].

A sensitive liquid chromatography/tandem mass spectrometry (LC/MS/MS) method for the quantitation of a novel topoisomerase I inhibitor (indolocarbazole derivative I) in human plasma was developed to support clinical studies. Drug and internal standard were isolated from plasma by solid-liquid extraction using 96-well diatomaceous earth plates. Various extraction solvents were evaluated for extraction of I and 9% isopropyl alcohol (IPA) in methyl-tert-butyl ether (MtBE) was chosen as the optimal extraction solvent. The sensitivity of this LC/MS/MS method is 10x higher in negative ion mode using alkaline conditions than in positive ion mode using a wide range of pH's. A mobile phase with 2 mM ammonium hydroxide enhanced the sensitivity in negative ion mode over other volatile bases. The calibration curve for compound I is linear over the range 0.05-200 ng/mL in plasma and the lower limit of quantification (LLOQ) of the assay is 0.05 ng/mL, when 0.25 mL of plasma is processed. The method was fully validated and successfully applied to plasma samples from clinical studies. Performing chromatography at high pH, for enhanced negative ion sensitivity, eliminates the need for post-column addition of base. Furthermore, the 96-well diatomaceous earth plate extraction offers the following advantages over liquid-liquid extraction (LLE) or solid-phase extraction (SPE): clean sample extracts with reduced sample preparation time; increased sample throughput; no conditioning or washing steps; and a neutral eluate applicable to acid/base labile compounds.

Chromosome loss from par mutants of Pseudomonas putida depends on growth medium and phase of growth.

The proteins encoded by chromosomal homologues of the parA and parB genes of many bacterial plasmids have been implicated in chromosome partitioning. Unlike their plasmid counterparts, mutant phenotypes produced by deleting these genes have so far been elusive or weakly expressed, except during sporulation. Here the properties of Pseudomonas putida strains with mutations in parA and parB are described. These mutants do not give rise to elevated levels of anucleate bacteria when grown in rich medium under standard conditions. However, in M9-minimal medium different parA and parB mutations gave between 5 and 10% anucleate cells during the transition from exponential phase to stationary phase. Comparison of the DNA content of bacteria at different stages of the growth curve, in batch culture in L-broth and in M9-minimal medium, suggests that the par genes are particularly important for chromosome partitioning when cell division reduces the chromosome copy number per cell from two to one. This transition occurs in P. putida during the entry into stationary phase in M9-minimal medium, but not in L-broth. It is proposed that the partition apparatus is important to ensure proper chromosome segregation primarily when the bacteria are undergoing cell division in the absence of ongoing DNA replication.

[Using a two-stage SBR process for removal of organics and nitrification-denitrification via nitrite].

High efficiency removal of organics and nitrogen by using a two-stage SBR process was introduced in this paper. Most of organics was removed in the first stage SBR reactor(SBR1) under the aerobic condition. Subsequently the second stage SBR reactor(SBR2) firstly operated under the aerabic condition for simultaneous nitrification and removal of a small amount of residual organics. Nitrification was controlled to the nitrite-type nitrification. Then denitrification happened in SBR2 under the anoxic condition. The petrochemical industry wastewater was used as external carbon sources in the denitrification. The experimental results indicated that in a two-stage SBR system, two kinds of biomass with the different function existed in the different reactors, which was beneficial to improve the treatment efficiency. The effluent COD reduced again because SBR2 removed COD which was left in SBR1 effluent. It resisted the disturbance of the high organic loading to nitrification. Consequently, as compared to a single SBR process, a two-stage SBR not only improved the treatment efficiency, but also saved the energy cost.
